[摘 要] 目的 了解浙江省部分漢族人群乙醛脫氫酶2(ALDH2)基因
多態(tài)性分布頻率?
方法 在浙江省地區(qū)收集1370例漢族人血液標(biāo)本,對(duì)其ALDH2基因12號(hào)外顯
子的特定片段進(jìn)行特異性擴(kuò)增,應(yīng)用基因芯片法判定基因型?
結(jié)果 ALDH2變異型等位基因在浙江省部分漢族人群中的分布頻率為44.5%
;此人群的ALDH2基因型頻率分布與上海人群之間的差異不存在統(tǒng)計(jì)學(xué)意義,而與洛陽人群
之間存在統(tǒng)計(jì)學(xué)意義?
結(jié)論 浙江省部分漢族人群ALDH2基因變異型有將近一半,表明此人群中AL
DH2酶活性缺乏者占近半數(shù);中國(guó)漢族人群的ALDH2基因多態(tài)性似存在南北地區(qū)差異?

[Abstract] Objective To investigate the ALDH2 gene polymorphis
m in part of Han population in Zhejiang province.
Methods With 1370 blood samples of Han population
in Zhejiang province, the specific fragment of ALDH2 gene exon 12 were amplifie
d using gene chip technique to judge the genotypes.
Results The frequency distribution of ALDH2 mutant alleles was 4
4.5% in Han population of Zhejiang province.
There was no statistical differences between the distributions of ALDH2 genotyp
e
frequencies of the population in Zhejiang province compared with that in Shangh
ai district; while there was a difference when compared to the population of Luo
yang population.
Conclusions The percentage of ALDH2 gene variant in Han populat
ion of Zhejiang province was nearly half, which showed that the percentage of pe
ople lacking ALDH2 enzyme activity was nearly half; and there was a regional dif
ference of ALDH2 gene polymorphism between north and south in Chinese Han popula
tion.

乙醛脫氫酶( aldehyde dehydrogenases, ALDH )是一組NAD(P)+依賴性酶,廣泛參與由體內(nèi)
乙醇?氨基酸?生物胺?維生素?類固醇及脂質(zhì)的代謝過程中產(chǎn)生的醛類物質(zhì)的氧化反應(yīng),
成為相應(yīng)的羧酸,對(duì)減輕醛類物質(zhì)對(duì)機(jī)體的毒性作用具有重要意義[1]?ALDH有19
種同工酶,其中以位于線粒體內(nèi)的ALDH2活性最強(qiáng),
在清除體內(nèi)醛類物質(zhì)中起主要作用[2]?ALDH2的活性變化與ALDH2基因多態(tài)性有關(guān)
,突變的
等位基因形成人群中三種基因型:具有正常催化活性的ALDH2野生純合子型(ALDH2*1/*1);
活性下降的ALDH2突變雜合子型(ALDH2*1/*2);活性喪失的ALDH2突變純合子型(ALDH2*2/*2
)?國(guó)內(nèi)外研究發(fā)現(xiàn)ALDH2基因多態(tài)性在不同人種和地區(qū)有不同的頻率分布,其與心血管疾病
?癌癥和藥物代謝等各方面均存在很大的關(guān)聯(lián)性,因此對(duì)我國(guó)漢族人群ALDH2基因多態(tài)性的
研究具有一定的重要性?
1 資料和方法
1.1 一般資料:所有標(biāo)本來自本院體檢和住院病例,且均為漢族人?其中男
974例,年齡19~79(49.30±9.86)歲;女396例,年齡15~79(49.81±10.64)歲,
人群之間無血緣關(guān)系?每人抽取2ml外周靜脈血,EDTA_NA2抗凝,儲(chǔ)存于4℃冰箱中?
1.2 實(shí)驗(yàn)方法及儀器:試劑采用上海百傲生物科技有限公司生產(chǎn)的試劑盒?儀器包括:Ba
iO BR_526_24全自動(dòng)雜交儀?BaiO生物芯片識(shí)讀儀?BioRadC1000 Thermal Cycler PCR儀?
Ep
pendorf Centrifuge 5418和Eppendorf Centrifuge 5415R高速離心機(jī)?按血液基因組DNA提
取試劑盒使用說明書來提取和純化DNA;將提取的1μl DNA加入擴(kuò)增液后放入PCR儀中,
按下列條件擴(kuò)
增:50℃5min,94℃ 5min,94℃25s,60℃25s,72℃30s,35個(gè)循環(huán);將PCR產(chǎn)物放入BaiO
雜交顯色試劑盒,放入雜交儀中,運(yùn)行雜交程序,反應(yīng)結(jié)束后,取出芯片;將芯片放入生
物芯片識(shí)讀儀中,用BaiO基因芯片圖像分析軟件V2.0進(jìn)行圖像掃描與數(shù)據(jù)分析,輸出檢測(cè)

1.3 統(tǒng)計(jì)學(xué)處理:采用SPSS 19.0軟件進(jìn)行統(tǒng)計(jì)學(xué)分析?計(jì)數(shù)資料用百分比表示,率的比

用χ2檢驗(yàn),P<0.05為差異有統(tǒng)計(jì)學(xué)意義?基因型分布采用Hardy_Weinberg平衡定律

2 結(jié)果
2.1 ALDH2基因型種類:檢測(cè)出ALDH2基因型有3種:ALDH2*1/*1型?ALDH2*1/*2型和ALDH2
*2/*2型?
2.2 ALDH2基因型在年齡和性別上的分布:浙江省漢族人群ALDH2基因野生型分布頻率為55
.5%,變異型分布頻率為44.5%?ALDH2基因型在性別和年齡分布上均無統(tǒng)計(jì)學(xué)意義(P>0.05),見表1?
2.4 Hardy_Weinberg平衡定律:ALDH2基因第12號(hào)外顯子504位點(diǎn)基因型分布符合Hardy_W
einberg平衡定律,χ2=1.92,P>0.05?
3 討論
近年來,對(duì)ALDH2的研究越來越廣泛,國(guó)內(nèi)外的研究表明ALDH2的基因多態(tài)性有民族?種族性
差異,東方人中ALDH2變異者較為普遍,而在白人和黑人中很少見[3]?曹西蓉等
[4]在
中國(guó)五個(gè)民族的研究結(jié)果中發(fā)現(xiàn)中國(guó)漢族和其他少數(shù)民族間存在ALDH2基因型分布的顯著差
異性?在中國(guó)上海和洛陽漢族人群的研究中也發(fā)現(xiàn)不同地區(qū)的漢族人群ALDH2存
在一定的差異性[5,6]?本文的研究結(jié)果發(fā)現(xiàn)浙江省部分漢族人群的ALDH2基因型頻
率分布與上海漢族人群之間
的差異無統(tǒng)計(jì)學(xué)意義,而與洛陽漢族人群之間存在統(tǒng)計(jì)學(xué)差異?可見中國(guó)南北地區(qū)的漢族人
群其ALDH2基因型頻率分布存在地域性的差異?
最新的單因素和多因素的薈萃回歸分析結(jié)果表明ALDH2基因多態(tài)性與亞洲人群的冠心病和心
肌梗死的風(fēng)險(xiǎn)增加密切相關(guān)[7]?Xu F等[8]的研究發(fā)現(xiàn):ALDH2突變型有
更高的心肌梗死率
和糖尿病的發(fā)病率;ALDH2突變型可能是中國(guó)漢族人群心肌梗死的獨(dú)立危險(xiǎn)因素;并表明ALD
H2基因多態(tài)性與心肌梗死的發(fā)病相關(guān),但與冠狀動(dòng)脈病變程度并無明顯的關(guān)系?ALDH2突變

群心肌梗死的發(fā)病率更是明顯高于正常人群[9]?動(dòng)物實(shí)驗(yàn)[10]也證明ALD
H2體內(nèi)的高表達(dá)能
減輕心力衰竭小鼠心功能的惡化(顯著增加EF和FS值)?抑制心梗受損區(qū)域心肌細(xì)胞的凋亡?
同時(shí)抑制心肌細(xì)胞內(nèi)p53蛋白的表達(dá)?王洪巨等[11]的研究證實(shí),乙醇激動(dòng)ALDH2表
達(dá)增高后
可發(fā)揮抗心肌細(xì)胞凋亡作用?Budas等[12]也證實(shí)外源性ALDH2激動(dòng)劑ALDA_1可產(chǎn)生
心肌保護(hù)
作用,抑制心肌細(xì)胞的凋亡?Li Y等[13]發(fā)現(xiàn)舌下含服硝酸甘油對(duì)部分心絞痛無效
的患者AL
DH2基因突變是主因?段富交等[14]發(fā)現(xiàn)ALDH2的變異基因型能夠增加中國(guó)人群食管
癌患病的
風(fēng)險(xiǎn)?郭李柯等[15]認(rèn)為ALDH2是口腔鱗狀細(xì)胞癌的易患因素?ALDH2基因型與心血
管疾病?
癌癥和藥物代謝等各方面的研究均存在很大的關(guān)聯(lián)性,而我國(guó)漢族人群ALDH2的基因型存在
普遍的變異性?本研究結(jié)果顯示浙江省漢族人群ALDH2基因變異型分布頻率為44.5%,表明

人群中ALDH2酶活性缺乏者占近半數(shù),因此在今后對(duì)該類人群無論從某些藥物的應(yīng)用上,還
是在心血管病和某些癌癥等疾病的研究中都有重要的啟示性?
