摘 要:探討8周有氧運(yùn)動(dòng)預(yù)干預(yù)對(duì)72h睡眠剝奪大鼠血清氧化應(yīng)激水平的影響。8周齡SD大鼠分為安靜對(duì)照組(CC)、運(yùn)動(dòng)組(EX)、睡眠剝奪組(SD)、運(yùn)動(dòng)預(yù)干預(yù)睡眠剝奪組(ES)。結(jié)果說(shuō)明,8周有氧運(yùn)動(dòng)預(yù)干預(yù)可降低血清中因72h睡眠剝奪引起氧化應(yīng)激升高的水平,提示有氧運(yùn)動(dòng)預(yù)干預(yù)可減輕因睡眠剝奪引起的血清中氧化應(yīng)激的損傷,減少自由基對(duì)血管系統(tǒng)的損害,對(duì)血管系統(tǒng)具有一定的保護(hù)意義。

Abstract: To investigate the effect of 8-week aerobic exercise pre-intervention on serum oxidative stress level in 72-hour sleep deprived rats. 8-week-old SD rats were divided into four groups: control group (CC), exercise group (EX), sleep deprivation group (SD), and exercise preintervention sleep deprivation group (ES). The results showed that 8-week aerobic exercise pre-intervention could decrease the level of oxidative stress induced by 72-hour sleep deprivation in serum, suggesting that aerobic exercise pre-intervention could alleviate the oxidative stress injury in serum caused by sleep deprivation. Reducing the damage of free radicals to the vascular system, the vascular system has a certain degree of protection.

在工作壓力不斷增多的當(dāng)今社會(huì),睡眠缺失已成為一種普遍的現(xiàn)象;睡眠剝奪的發(fā)生率明顯升高,廣泛影響著機(jī)體的健康,睡眠缺失已成為導(dǎo)致亞健康的因素之一[1,2]。本實(shí)驗(yàn)從自由基角度,觀察8周的有氧運(yùn)動(dòng)預(yù)干預(yù)對(duì)睡眠剝奪大鼠血清、心肌組織氧化應(yīng)激水平的影響。
1 材料與方法
1.1 實(shí)驗(yàn)動(dòng)物
健康雄性SD大鼠,8周齡,體重180~220g。
1.2 實(shí)驗(yàn)分組
大鼠進(jìn)行適應(yīng)性跑臺(tái)訓(xùn)練,剔除不適應(yīng)跑臺(tái)訓(xùn)練者,大鼠32只進(jìn)行綜合分組,隨機(jī)分為4組,安靜對(duì)照組(CC,N=8),運(yùn)動(dòng)組(EX,N=8),睡眠剝奪組(SD,N=8),運(yùn)動(dòng)預(yù)干預(yù)睡眠剝奪組(ES,N=8)。
1.3 運(yùn)動(dòng)方案
按Bedford[3]方法改進(jìn)運(yùn)動(dòng)方案,前兩周30min/d,跑臺(tái)速度10m/min,坡度0°,分兩小節(jié)進(jìn)行;兩周45min/d,跑臺(tái)速度15m/min,坡度0°,分三小節(jié)進(jìn)行;后四周60min/d,跑臺(tái)速度15m/min,坡度0°,分四小節(jié)進(jìn)行;每小節(jié)15min,兩小節(jié)的運(yùn)動(dòng)間隔時(shí)間3~5min,每周運(yùn)動(dòng)6天,休息1d。
1.4 統(tǒng)計(jì)學(xué)處理
實(shí)驗(yàn)數(shù)據(jù)均以均值±標(biāo)準(zhǔn)差()表示,采用SPSS 19.0軟件進(jìn)行t檢驗(yàn),差異顯著性意義為P<0.05。
2 結(jié)果及分析
2.1 各組大鼠血清SOD活性的變化
各組大鼠血清SOD活性的變化如表1所示。

3 討論
本實(shí)驗(yàn)結(jié)果顯示,睡眠剝奪組(SD)大鼠血清MDA含量顯著高于對(duì)照組,SOD活性與對(duì)照組(CC)相比有降低趨勢(shì)。GSH-px活性與對(duì)照組(CC)相比顯著升高。雖然與上述前人研究的睡眠剝奪的時(shí)間不同,但本研究表明了72小時(shí)的睡眠剝奪也可使血清中和心肌組織脂質(zhì)過(guò)氧化程度加劇,抗氧化酶活性降低,過(guò)量的自由基可損害血管系統(tǒng),提示72小時(shí)的睡眠剝奪可損害血管系統(tǒng)和心肌組織。本文以有氧運(yùn)動(dòng)預(yù)干預(yù)作為干預(yù)因子,研究其對(duì)睡眠剝奪大鼠氧化應(yīng)激水平的影響。實(shí)驗(yàn)結(jié)果顯示有氧運(yùn)動(dòng)預(yù)干預(yù)睡眠剝奪組(ES)大鼠血清MDA含量顯著低于睡眠剝奪組(SD);運(yùn)動(dòng)預(yù)干預(yù)睡眠剝奪組(ES)血清中GSH-px/MDA比值顯著高于睡眠剝奪組(SD)。表明運(yùn)動(dòng)預(yù)干預(yù)睡眠剝奪組(ES)血清脂質(zhì)過(guò)氧化程度低于睡眠剝奪組(SD),運(yùn)動(dòng)預(yù)干預(yù)組睡眠剝奪組(ES)血清抗脂質(zhì)過(guò)氧化的能力明顯高于睡眠剝奪組(SD)。提示8周有氧運(yùn)動(dòng)可減輕血清中因睡眠剝奪引起的脂質(zhì)過(guò)氧化的程度。
8周有氧運(yùn)動(dòng)預(yù)干預(yù)可減輕血清中因72小時(shí)睡眠剝奪引起的氧化應(yīng)激的損傷。
