填飼應(yīng)激對(duì)北京鴨生產(chǎn)性能、腿部肌肉品質(zhì)、血清激素和血?dú)庵笜?biāo)的影響

摘要:為了探究填飼應(yīng)激對(duì)北京鴨生產(chǎn)性能、腿部肌肉品質(zhì)、血清激素和血?dú)庵笜?biāo)的影響,本試驗(yàn)選擇160只39日齡、平均體重為(2.573±0.096)kg的北京鴨,分為2組,每組80只,分別為基礎(chǔ)日糧填飼組和基礎(chǔ)日糧對(duì)照組。試驗(yàn)6 d后,各處理組隨機(jī)選取8只肉鴨分別稱取體重,計(jì)算平均日增重,并進(jìn)行腿部肌肉品質(zhì)(pH、滴水損失率)、血清激素(ACTH、CORT、T3、T4)和血?dú)鈾z測(cè)。結(jié)果表明,填飼組肉鴨填飼后體重和平均日增重均顯著高于對(duì)照組(P<0.05);血清CORT、AnGap、Hct和Hb含量極顯著高于對(duì)照組(P<0.01);而填飼組45 min和24 h時(shí)的脛骨前肌pH極顯著低于對(duì)照組(P<0.01)。結(jié)果顯示,填飼能顯著提高肉鴨的增重效率;但同時(shí)會(huì)顯著降低肉鴨腿部肌肉的肉品質(zhì);填飼會(huì)使肉鴨血清中CORT的含量顯著升高,使其出現(xiàn)應(yīng)激征象;填飼還會(huì)在一定程度上影響肉鴨的血液循環(huán)系統(tǒng)。

中國(guó)是世界第一的養(yǎng)鴨大國(guó),養(yǎng)鴨業(yè)的發(fā)展在養(yǎng)殖業(yè)中占有比較重要的位置。生產(chǎn)實(shí)踐中,填飼的目的主要是為“北京烤鴨”提供原材料,其機(jī)理是在短期內(nèi)、超量飼喂以碳水化合物為主的能量物質(zhì)時(shí),碳水化合物會(huì)通過(guò)磷酸戊糖和糖酵解等途徑轉(zhuǎn)化為甘油三酯,最終動(dòng)物會(huì)將多余的甘油三酯以脂肪的形式貯存起來(lái)[1-2],這樣的方式會(huì)使肉鴨快速沉積皮下脂肪,從而保證烤制后的風(fēng)味。
但是當(dāng)進(jìn)行人工填鴨時(shí),驅(qū)趕、抓鴨、插管等一系列操作會(huì)給肉鴨造成較大程度的應(yīng)激[3],多項(xiàng)關(guān)于家禽應(yīng)激的研究都發(fā)現(xiàn),應(yīng)激會(huì)導(dǎo)致家禽抵抗力下降、易遭受感染,繼而出現(xiàn)腹瀉、生長(zhǎng)不良、死亡等癥狀[4-5],這將給企業(yè)帶來(lái)較大經(jīng)濟(jì)損失。本試驗(yàn)通過(guò)對(duì)填飼與不填飼肉鴨的生產(chǎn)性能、腿部肌肉品質(zhì)、血清激素和血?dú)獾纫幌盗兄笜?biāo)的對(duì)比,來(lái)進(jìn)一步探索填飼應(yīng)激對(duì)肉鴨機(jī)體產(chǎn)生的影響。
1材料與方法
1.1試驗(yàn)設(shè)計(jì)與飼養(yǎng)管理
試驗(yàn)選擇健康的、體重相近的(2.573 ± 0.096) kg 39日齡北京鴨,共160只(公母各半),隨機(jī)分成兩個(gè)處理組:第一組為填飼組,每天填飼時(shí)間分別為03:00、09:00、15:00和21:00,每天每只肉鴨平均填飼276.37 g基礎(chǔ)日糧;第二組為對(duì)照組,肉鴨自由采食基礎(chǔ)日糧。填飼時(shí)舍內(nèi)平均溫度為28~33 ℃,相對(duì)濕度為58%~65%。
1.2試驗(yàn)基礎(chǔ)日糧
配方及營(yíng)養(yǎng)成分見表1。
1.3樣品采集與指標(biāo)測(cè)定
1.3.1北京鴨生產(chǎn)性能肉鴨填飼6 d后,每個(gè)處理組隨機(jī)選取8只肉鴨并分別稱取體重,計(jì)算平均末重和平均日增重(Average daily gain,ADG)。
1.3.2北京鴨腿部肌肉品質(zhì)將肉鴨右腿腓腸肌剪下約10 g肉塊懸吊于紙杯中,肉塊不能接觸任何物體,紙杯外再套一層自封袋,密封后置于4 ℃冰箱中等待檢測(cè)。最后分別于屠宰后45 min和24 h用吸水紙吸去肉塊表面水分,并用電子稱稱取肉塊的準(zhǔn)確重量,分別記為W1(g)和W2(g),則滴水損失率= (W1-W2)/ (W1)×100%。

1).預(yù)混料為每千克飼糧提供:Cu(CuSO4·5H2O)10 mg,F(xiàn)e(FeSO4·7H2O)50 mg,Zn(ZnO)60 mg,Mn(MnSO4·H2O)80 mg,Se(NaSeO3)0.25 g,I(KI)0.2 mg,Cr(Cr2O3)0.15 mg,氯化膽堿1 000 mg,VA 1 000 IU,VD33 000 IU,VE 20 IU,VK32 mg,硫胺素2 mg,核黃素8 mg,VB64 mg,VB120.06 mg,煙酸50 mg,葉酸1 mg,生物素0.2 mg,植酸酶0.50 mg,5%甘露醇60 mg,D-泛酸50 mg。2).營(yíng)養(yǎng)水平為計(jì)算值。3).根據(jù)肉鴨飼料原料表觀代謝能進(jìn)行計(jì)算

將右腿脛骨前肌完整剪下,于自封袋中保存,密封后放置于4 ℃冰箱中等待檢測(cè)。最后分別于屠宰后45 min和24 h測(cè)定pH,并記錄。
1.3.3北京鴨血清激素指標(biāo)血清樣品送至北京華英生物技術(shù)研究所,檢測(cè)血清中促腎上腺皮質(zhì)激素(ACTH,pg·mL-1)、皮質(zhì)酮(CORT,ng·mL-1)、三碘甲狀腺素(T3,ng·mL-1)和四碘甲狀腺素(T4,ng·mL-1)的含量。
1.4數(shù)據(jù)分析
用格魯布斯檢查法剔除異常值。用SPSS 20.0軟件對(duì)所有數(shù)據(jù)進(jìn)行獨(dú)立樣本t-檢驗(yàn)。統(tǒng)計(jì)結(jié)果用“平均值±標(biāo)準(zhǔn)差”表示。
2結(jié)果
2.1填飼應(yīng)激對(duì)北京鴨生產(chǎn)性能的影響
由表2可知,兩組肉鴨初始體重均為(2.573±0.096),填飼6 d后,填飼組平均體重及平均日增重均顯著高于對(duì)照組(P=0.021;P=0.021),填飼組平均末重比對(duì)照組提高了8.5%。

2.2填飼應(yīng)激對(duì)北京鴨腿部肌肉品質(zhì)的影響
由表3可知,屠宰后45 min和24 h時(shí),填飼組脛骨前肌的pH均極顯著低于對(duì)照組(P=0.001;P=0.001),說(shuō)明填飼會(huì)顯著降低肉鴨腿部肌肉pH,45 min和24 h時(shí)分別降低了10.26%和12.31%;而填飼組腿部肌肉的滴水損失高于對(duì)照組(P=0.082),但并未達(dá)到顯著水平,這只能說(shuō)明填飼會(huì)在一定程度上加快肉鴨腿部肌肉失水速度,使其增加60%左右。

2.3填飼應(yīng)激對(duì)北京鴨血清激素的影響
由表4可知,填飼會(huì)顯著影響肉鴨血清CORT值,使填飼組血清CORT值極顯著高于對(duì)照組(P<0.001),但是填飼并不會(huì)顯著影響肉鴨血清ACTH、T3及T4的含量。

3討論
3.1填飼應(yīng)激對(duì)北京鴨生產(chǎn)性能的影響
在肉鴨生產(chǎn)中,填飼的主要目的是快速沉積脂肪以及生產(chǎn)鴨肥肝,關(guān)于填飼對(duì)動(dòng)物生產(chǎn)性能的研究已經(jīng)有許多比較深入的報(bào)道,其他研究表明,對(duì)肉鴨或鵝進(jìn)行填飼后,其生產(chǎn)性能會(huì)有很大提高[2,6-7]。本試驗(yàn)結(jié)果(表2)同樣直觀地證明了填飼能顯著提高肉鴨的增重效率,與前人得出的試驗(yàn)結(jié)論相一致。
3.2填飼應(yīng)激對(duì)北京鴨腿部肌肉品質(zhì)的影響
肉類質(zhì)檢中通常用pH、系水力等指標(biāo)來(lái)評(píng)價(jià)肌肉的品質(zhì)。肌肉pH的變化與肌細(xì)胞的糖酵解反應(yīng)有關(guān),其不僅是肌肉酸度的直觀表現(xiàn),還對(duì)肉的嫩度、滴水損失等有直接影響[8]。當(dāng)組織對(duì)氧氣利用不良,或由于循環(huán)障礙造成組織供氧不足時(shí),會(huì)加強(qiáng)糖酵解反應(yīng),使乳酸產(chǎn)生增多。M.P.Mollica等的研究表明,填飼后肉鴨肝體積將會(huì)增大,當(dāng)從食物中攝入的能量過(guò)多時(shí),肝的耗氧量會(huì)顯著上升達(dá)32%[9]。這會(huì)使腿部肌肉血管內(nèi)氧氣含量相對(duì)下降,供氧量不足,加強(qiáng)了肌肉細(xì)胞糖酵解反應(yīng),產(chǎn)生更多乳酸導(dǎo)致腿部肌肉pH下降,這就是填飼組腿部肌肉pH顯著低于對(duì)照組的原因。
pH是影響肌肉系水力的重要因素,肌肉pH的下降會(huì)導(dǎo)致其系水力降低[8],從而導(dǎo)致填飼組腿肌滴水損失率高于對(duì)照組,但不顯著。
3.3填飼應(yīng)激對(duì)北京鴨血清激素的影響
當(dāng)肉鴨處于應(yīng)激狀態(tài)時(shí),將激活下丘腦—垂體—腎上腺皮質(zhì)系統(tǒng),使ACTH分泌增加,進(jìn)而促進(jìn)CORT的分泌,血清中高含量CORT在應(yīng)激初期中具有重要的作用:升高血糖,促進(jìn)糖異生,并對(duì)兒茶酚胺、胰高血糖素等的脂肪動(dòng)員起容許作用,還具有穩(wěn)定溶酶體膜,減少溶酶體酶對(duì)細(xì)胞損傷的作用。本試驗(yàn)發(fā)現(xiàn),填飼組肉鴨的CORT值顯著高于對(duì)照組,表明填飼可導(dǎo)致肉鴨出現(xiàn)生理應(yīng)激現(xiàn)象。在V.M.Darras等的研究中發(fā)現(xiàn),血液中高CORT含量會(huì)使T4水平顯著下降、T3水平輕微下降[10]。通過(guò)表4可知,短期填飼不僅顯著提高了血清中CORT的含量,還使血清中T3、T4的含量輕微下降,這與V.M.Darras等的研究結(jié)果基本一致。G.A.Cremaschi和A.J.Klecha等的研究表明,正常的甲狀腺激素水平對(duì)于抗體和細(xì)胞介導(dǎo)免疫反應(yīng)的發(fā)育、維持、功能都是必不可少的[4,11],因此本試驗(yàn)表明短期填飼可能會(huì)對(duì)肉鴨的代謝及免疫反應(yīng)產(chǎn)生一定影響。
3.4填飼應(yīng)激對(duì)北京鴨血?dú)夂脱弘x子濃度的影響
造成填飼組Hct和Hb顯著高于對(duì)照組的原因有很多:首先,在本試驗(yàn)中填飼組Hct升高,通常表示原發(fā)或繼發(fā)性紅細(xì)胞增多癥[13]。慢性低氧血癥時(shí),由于血液中氧分壓持續(xù)降低,刺激腎分泌的促紅細(xì)胞生成素(EPO)增多,導(dǎo)致繼發(fā)性紅細(xì)胞增多;其次,紅細(xì)胞生成和紅細(xì)胞數(shù)量還受腎上腺皮質(zhì)、甲狀腺、卵巢、睪丸和垂體前葉所分泌的激素影響[14],本試驗(yàn)填飼組的高CORT數(shù)值也是導(dǎo)致紅細(xì)胞原發(fā)性增多的原因之一;最后,當(dāng)紅細(xì)胞凋亡時(shí),需要由巨噬細(xì)胞吞噬、分解,A.V.S.Gomes等研究表明,應(yīng)激會(huì)顯著降低巨噬細(xì)胞的活性[5],從而降低紅細(xì)胞被破壞的數(shù)量。所以,紅細(xì)胞生成增多而破壞減少導(dǎo)致了填飼組Hct、Hb顯著高于對(duì)照組,血液中紅細(xì)胞增多的直接結(jié)果就是增加了血液粘度與血流阻力,容易導(dǎo)致血栓的形成[12],這會(huì)對(duì)肉鴨血液循環(huán)系統(tǒng)造成潛在影響。
4結(jié)論
本試驗(yàn)發(fā)現(xiàn),填飼能顯著提高肉鴨的增重效率,但同時(shí)會(huì)顯著降低肉鴨腿部肌肉的pH,在一定程度上影響肉品質(zhì);填飼會(huì)使肉鴨血清中CORT的含量顯著升高,使其出現(xiàn)應(yīng)激征象;填飼還會(huì)在一定程度上影響肉鴨的血液循環(huán)系統(tǒng)。因此,未來(lái)的研究將著重探究何種添加劑能在不影響增重效率的條件下提高肉鴨腿部肌肉品質(zhì)并緩解填飼應(yīng)激。

Abstract:The objective of this study was to evaluate effects of force-feeding stress on growth performance,meat characteristics of legs,serum hormone and blood gas parameters of Peking ducks.A total of 160 thirty-nine-day-old Peking ducks with average body weight (2.573±0.096) kg were randomly divided into 2 groups (force-feeding group and control group) with 80 ducks per treatment.After 6 days,8 ducks from each treatment were selected randomly to be weighted for calculating average body weight and average daily gain,and meat characteristics of leg(pH and drip loss rate),serum hormone (ACTH,CORT,T3 and T4) and blood gas parameters were tested.The results showed that the average body weight and average daily gain in force-feeding group was significantly higher than those in control group (P<0.05).And the CORT,AnGap,Hct and Hb in force-feeding group was significantly higher than that in control group (P<0.01).The pH at 45 min and 24 h of tibialis anterior in force-feeding group were significantly lower than those in control group (P<0.01).It was concluded that force-feeding could enhance gaining weight significantly.However,force-feeding could affect the meat characteristics of leg significantly and induce physiological stress of Peking ducks by increasing serum CORT.At last,force-feeding could also influence the blood circulation system of Peking ducks.
