        熱應(yīng)激對(duì)兔下丘腦-垂體-睪丸軸NMB及其受體基因表達(dá)和睪酮分泌的影響

        摘要:研究熱應(yīng)激對(duì)公兔下丘腦-垂體-睪丸(HPT)軸神經(jīng)介素B(NMB)及其受體(NMBR)基因表達(dá)水平及血清皮質(zhì)醇和睪酮分泌的影響,探討熱應(yīng)激對(duì)神經(jīng)肽和相關(guān)激素分泌的影響。建立熱應(yīng)激模型,采用熒光定量PCR(RT-PCR)檢測(cè)兔NMB和NMBR mRNA在HPT軸的表達(dá)情況,采用放射免疫分析法(RIA)檢測(cè)兔血清中皮質(zhì)醇和睪酮濃度的變化規(guī)律。結(jié)果表明,兔NMB和NMBR基因在HPT軸中表達(dá);NMB和NMBR mRNA在HPT軸的表達(dá)模式相似,且熱應(yīng)激后在一定程度上可以促進(jìn)NMB和NMBR基因的表達(dá),并按時(shí)序在下丘腦(熱應(yīng)激0.5 h)、垂體(熱應(yīng)激1.0 h)和睪丸(熱應(yīng)激2.0 h)促進(jìn)作用最為明顯;熱應(yīng)激可以促進(jìn)皮質(zhì)醇的分泌,并抑制睪酮的分泌,但這種抑制作用隨著熱應(yīng)激時(shí)間的增加(熱應(yīng)激2.0 h)逐漸恢復(fù)正常。表明熱應(yīng)激可能通過NMB/NMBR系統(tǒng)調(diào)節(jié)HPT軸的功能,進(jìn)而影響動(dòng)物繁殖。

        Abstract: The aim of this study was to detect the effect of heat stress on the expression levels of Neuromedin B (NMB) and its receptor (NMBR) genes in the hypothalamus-pituitary-testis(HPT) axis of male rabbit, and the secretion of serum cortisol (Cor.) and testosterone (T). Establishment The heat stress model was established, the expression levels of NMB and NMBR genes in rabbit HPT axis were detected by real-time PCR(RT-PCR), and the concentration of Cor. and T in rabbit serum were tested using radioimmunoassay (RIA). Results showed that NMB and NMBR genes were expressed in rabbit HPT axis. The expression pattern of NMB and NMBR mRNA in HPT axis were similar, and heat stress could promote the NMB and NMBR gene expression, which the role was mostly obvious according to the time sequence in the hypothalamus (heat stress 0.5 h), pituitary (heat stress 1.0 h) and testis (heat stress 2.0 h); RIA results indicated that heat stress could improve the concentration of Cor. and inhibit T secretion which the inhibition was gradually weakened over time of heat stress. The results indicated that heat stress may regulate the HPT axis through the NMB/NMBR system to affect reproductive function, and then affected the reproduction of animals.
