摘要:本年度預(yù)期研究目標(biāo)包括構(gòu)建siRNA,培養(yǎng)人臍靜脈內(nèi)皮細(xì)胞,完成各分組,給予不同的剪切力觀察不同的剪切力觀察信號(hào)轉(zhuǎn)導(dǎo)通路膜整合素→MAPKs→核轉(zhuǎn)錄因子→MMP和P4Hα1及底物的關(guān)系。同時(shí)觀察炎性因子的改變;建立動(dòng)脈粥樣硬化小型豬模型,獲取斑塊的OCT或光聲圖像,進(jìn)行圖像和回波波譜分析,對(duì)比組織學(xué)實(shí)驗(yàn)結(jié)果,建立彈性數(shù)據(jù)和斑塊組織成分間的映射關(guān)系; 結(jié)合真實(shí)血管三維重建構(gòu)型,利用Comsol Multiphysics 有限元軟件建立仿真模擬系統(tǒng)。建立兔動(dòng)脈粥樣硬化動(dòng)物模型,定期檢測(cè)獲取不同階段的相關(guān)數(shù)據(jù)。主要研究發(fā)現(xiàn)包括(1)證實(shí)斑塊部位不同的機(jī)械應(yīng)力特征與不同的病理學(xué)變化具有顯著相關(guān)性;(2)首次提出了細(xì)胞內(nèi)NADPH 氧化酶介導(dǎo)的氧化還原信號(hào)傳導(dǎo)機(jī)制可能是細(xì)胞應(yīng)激反應(yīng)體系中一個(gè)固有的組成部分;(3)嘗試?yán)蔑@微超聲粒子圖像測(cè)速技術(shù)(Micro Ultrasonic Particle Image Velocimetry)獲得血管內(nèi)血流場(chǎng)速度矢量分布.

Abstract:This year we aims to construct siRNA, culture HUVEC cells and test the effects of shear stress on signaling pathways including integrin-->MAPKs-->transcription factors-->MMP and P4Hα1 expression, and observe the changes in inflammatory responses; Obtain OCT or optical-sonic images and perform image or echo spectrum analysis, and compare the results with real histology images; clarify the relationships between plaque elasticity data and the biological components in the plaques; Using Comsol Multiphysics software to create hemodynamic simulation platform, and test the correlations in rabbit models of atherosclerosis. Our major findings include: (1) we have shown that mechanic properties of atherosclerotic plaque are correlated with histology characteristics; (2) we propose NADPH oxidase-dependent redox signaling may be a critical component in cellular stress response; (3) we attempted to use Micro Ultrasonic Particle Image Velocimetry technique to demonstrate the distribution of blood flow hemodynamic parameters.
