傳統(tǒng)中藥華中五味子(Schisandra sphenantheraRehd.et Wils.)來源于五味子屬植物華中五味子的干燥成熟果實(shí),又稱為“南五味子”,是中醫(yī)常用的滋補(bǔ)強(qiáng)壯藥。近年來,通過大量臨床實(shí)驗(yàn),發(fā)現(xiàn)其在諸多方面都有較好的療效,臨床應(yīng)用有更加廣泛的前景。
質(zhì)譜(EI-MS)條件為安捷倫-5973型,直接進(jìn)樣,在70eV下測定,檢索用NIST數(shù)據(jù)庫;核磁共振譜(1H-NMR,13C-NMR 和DEPT)均在INOVA-400核磁共振儀上測定(TMS為內(nèi)標(biāo))。柱層析用硅膠(200~300目,40~80目,硅膠H),薄層層析用硅膠(GF254)及GF254薄層層析板均為青島海洋化工廠產(chǎn)品。反相材料RP-18(40~63μm) 及RP-18薄層板為默克公司產(chǎn)品(Merck,Darmstadt,Germany);Sephadex LH-20(25~100 μm)為Amersham Biosciences,Sweden公司產(chǎn)品。薄層層析板采用UV-254和365 nm、碘蒸氣、5% 硫酸-乙醇溶液、顯色觀察其斑點(diǎn)。所有化學(xué)溶劑均為重蒸的工業(yè)級試劑,氘代試劑購于武漢中興科技。實(shí)驗(yàn)用植物樣品(約15kg)于2010年8月購自貴州省三橋藥材市場,產(chǎn)自四川,經(jīng)貴陽中醫(yī)學(xué)院陳德媛教授鑒定為五味子屬植物華中五味子(Schisandra sphenantheraRehd.et Wils.)的干燥成熟果實(shí)。
華中五味子成熟干燥果實(shí)(15.0 kg)粉碎后用95%乙醇回流提取3次(時間依次為5h、4h、4h),提取液合并,并減壓濃縮至浸膏。用水制成混懸液后,依次用石油醚、乙酸乙酯、正丁醇萃取(每種溶劑至少萃取3次)?;厥杖軇┖蠓謩e得到石油醚萃取物部位、乙酸乙酯萃取物部位、正丁醇萃取物部位及最后的水部分,經(jīng)薄層層析(TLC)分析檢測后,發(fā)現(xiàn)石油醚萃取物部位和乙酸乙酯萃取物部位基本相同,故合并一起,濃縮成浸膏約1731g。浸膏分別用硅膠柱和凝膠柱反復(fù)層析的方法,石油醚∶丙酮(1∶0~0∶1)梯度洗脫分離得到化合物五味子酚111.08g(schisanhenol,1),五味子酯乙523mg(schisantherin B,2),chicanine 28mg(3),pregomisin 6mg(4)、甘五酸123mg(schizandronic acid,5)、去氧五味子素 18mg(6)、前戈米辛 9mg(7)、內(nèi)消旋二氫愈創(chuàng)木脂素 22mg(8),五味子酯甲 13mg(9),戈米辛P順芷酸酯 15mg(10)。其中前戈米辛(7)首次從該植物中分離得到。
