【摘要】 目的 研究雷公藤甲素對(duì)哮喘小鼠哮喘小鼠白三烯B4、C4表達(dá)的影響,探討其治療哮喘的機(jī)制。方法 建立小鼠卵蛋白哮喘模型,隨機(jī)分為7組,分別為對(duì)照組、地塞米松治療組及不同劑量雷公藤甲素治療組,用ELISA測(cè)定支氣管肺泡灌洗液(BALF)及肺組織勻漿中白三烯 B4、C4表達(dá)水平。結(jié)果 哮喘組白三烯 B4、C4表達(dá)明顯高于正常對(duì)照組(P<0.01),雷公藤甲素治療組及地塞米松組明顯低于哮喘組(P<0.01)。結(jié)論 雷公藤甲素抑制哮喘氣道炎癥的機(jī)制可能與其抑制白三烯B4、C4表達(dá)的表達(dá)活性有關(guān)。

Effects of triptolide on the expression of TLB4 and TLC4 in asthmatic mice[HJ1*2][HJ]

【Abstract】 Objective To study the effect s of triptolide on the expression of TLB4 and TLC4 in asthmatic mouse model,and to explore the possible therapeutic mechanism of t riptolide in asthma.
Methods The asthmatic model was established by ovalbumin injection and inhalation. The mice were randomly divided into 7 groups andtreated respectively with normal saline (as control) ,dexamethasone or different dose of triptolide. The expression of TLB4 and TLC4 in ranchoalveolar avage fluid (BAL F) and in lung tissue was were detected by ELISA respectively.Results The expression of TLB4 and TLC4 in airway epithelium of asthmatic group significantly increased compared with those of controlgroup(P<0.01). The expression of TLB4 and TLC4 expression in mice t reated with different dose of t riptolide and dexamethson were significantly lower than those in asthmatic group(P<0.01).Con
clusion The mechanism of t riptolide inhibiting asthmatic airway inflammation may be related to its inhibiting activities of TLB4 and TLC4.

支氣管哮喘是一種常見病、多發(fā)病,由于對(duì)其病因及發(fā)病機(jī)制不甚清楚,雖然采用現(xiàn)有的方法,已能有效地控制大多數(shù)病人的癥狀,但對(duì)部分病人療效欠佳,尋求和開發(fā)有效的治療哮喘的藥物對(duì)哮喘的防治具有十分重要的意義。本實(shí)驗(yàn)研究雷公藤甲素對(duì)哮喘小鼠白三烯B4、C4表達(dá)的影響,為開發(fā)雷公藤甲素治療哮喘在臨床上應(yīng)用提供理論依據(jù)。
