摘要:目的? 觀察電針聯(lián)合兩色金雞菊提取物對(duì)糖尿病大鼠腎臟波形蛋白(Vimentin)、α-平滑肌肌動(dòng)蛋白(α-SMA)、轉(zhuǎn)化生長因子-β1(TGF-β1)及p-smad2表達(dá)的影響,探討其作用機(jī)制。方法? 將36只SD大鼠隨機(jī)分為正常組、模型組、電針組、兩色金雞菊組、電針+兩色金雞菊組和二甲雙胍組,每組6只。采用高糖高脂喂養(yǎng)聯(lián)合腹腔注射鏈脲佐菌素建立2型糖尿病模型。正常組、模型組不進(jìn)行干預(yù);電針組針刺大鼠雙側(cè)“腎俞”“脾俞”,連接電針,留針30 min;兩色金雞菊組予兩色金雞菊乙酸乙酯提取物(300 mg/kg)灌胃;電針+兩色金雞菊組予電針聯(lián)合兩色金雞菊乙酸乙酯提取物灌胃;二甲雙胍組予二甲雙胍(200 mg/kg)藥液灌胃。每日1次,治療4周。HE染色觀察腎組織形態(tài)學(xué)及纖維化程度,Western blot檢測腎組織Vimentin、α-SMA、TGF-β1及p-smad2蛋白表達(dá)。結(jié)果? HE染色結(jié)果顯示,電針組、兩色金雞菊組、電針+兩色金雞菊組和二甲雙胍組均可緩解模型大鼠腎小管擴(kuò)張,減輕腎小球皺縮現(xiàn)象,改善炎性細(xì)胞浸潤,減少基底膜增生和融合,且以電針+兩色金雞菊組改善更明顯;Western blot檢測結(jié)果顯示,電針和兩色金雞菊提取物均可降低模型大鼠腎組織Vimentin、α-SMA、TGF-β1及p-smad2蛋白表達(dá),且以兩者聯(lián)合效果最佳。結(jié)論? 電針聯(lián)合兩色金雞菊提取物可明顯改善糖尿病大鼠腎臟纖維化,其作用機(jī)制可能與抑制TGF-β1/Smad2信號(hào)通路有關(guān)。

Abstract: Objective To observe the effects of electroacupuncture combined with coreopsis tinctoria extract for expressions of Vimentin, α-SMA, TGF-β1 and p-smad2 in renal tissue of diabetic rats; To discuss its mechanism of action. Methods Totally thirty-six SD rats were randomly divided into normal group, model group, electroacupuncture group, coreopsis tinctoria group, electroacupuncture combined with coreopsis tinctoria group and metformin group, with 6 rats in each group. Type 2 DM model was established by high-fat and high-glucose diet and STZ injection intraperitoneally. Normal group and model group were given no treatment; Electroacupuncture was at the acupoints of “Pishu” and “Shenshu” once daily in electroacupuncture group for 30 min; coreopsis tinctoria group was given intragastric administration of 300 mg/kg coreopsis tinctoria extract for gavage; electroacupuncture combined with coreopsis tinctoria group was given electroacupuncture and intragastric administration of coreopsis tinctoria extract for gavage; metformin group was given intragastric administration of 200 mg/kg metformin for gavage. Histological changes and fibrosis degree of kidneys were observed by HE staining and expressions of Vimentin, α-SMA, TGF-β1 and p-smad2 were detected by Western blot. 糖尿病腎?。╠iabetic nephropathy,DN)已成為終末期腎病的第2位致病因素[1]。終末期腎病一旦形成,治療極為棘手。因此,及時(shí)治療DN,防止其向終末期腎病轉(zhuǎn)化意義重大。腎臟纖維化是DN主要的病理特征之一,前期研究發(fā)現(xiàn),兩色金雞菊提取物對(duì)高糖高脂誘導(dǎo)的大鼠腎小球系膜纖維化有明顯抑制作用[2-3],而電針也可改善DN腎功能和超微結(jié)構(gòu)[4]。本實(shí)驗(yàn)擬將電針聯(lián)合兩色金雞菊提取物干預(yù)DN大鼠,觀察其對(duì)DN大鼠腎臟波形蛋白(Vimentin)、α-平滑肌肌動(dòng)蛋白(α-SMA)、轉(zhuǎn)化生長因子-β1(TGF-β1)及p-smad2表達(dá)的影響,明確兩者是否存在協(xié)同作用,為臨床治療DN提供依據(jù)。
