小鼠肉瘤細胞分泌的生長因子對裸鼠坐骨神經(jīng)損傷修復的探討

目的 探討小鼠肉瘤細胞S180分泌的神經(jīng)生長因子(Nerve Growth Factor,NGF)與外源性神經(jīng)生長因子對損傷坐骨神經(jīng)的修復效果。方法 將裸鼠分為8組,實驗操作后觀察其肢體運動恢復情況,采用改良Tarlov評分,取血液標本檢測NGF濃度;取脊髓腰膨大處做切片染色,觀察受體Trka和p75。結果 小鼠肉瘤細胞S180分泌NGF,且接種量不同,NGF產生量也不同,接種1×104為0.390±0.018,接種1×105為0.445±0.012,接種2×105為0.469±0.017,接種1×106為0.520±0.049,接種2×106為0.571±0.042;接種組神經(jīng)損傷恢復效果明顯優(yōu)于未接種組;改良評分接種組接種1×104為2.00±0.00,接種1×105為2.25±0.50,接種2×105為2.75±0.50,接種1×106為2.75±0.50,接種2×106為2.50±0.57,均優(yōu)于其他組;切片顯示接種組Trka和P75均表達增高。結論 接種肉瘤細胞S180可產生內源性NGF,內源性NGF對周圍神經(jīng)損傷修復效果優(yōu)于外源性NGF。

使用神經(jīng)生長因子(NGF)治療周圍神經(jīng)損傷一直是研究的熱點,NGF能夠促進神經(jīng)的修復[1-2]。動物實驗中有采用外源性NGF及通過構建模型分泌內源性NGF來研究NGF對坐骨神經(jīng)損傷后的修復效果[3-6],但實驗缺乏內源性NGF與外源性NGF治療效果的對照研究,給藥途徑和內源性NGF持續(xù)釋放也理想,本實驗對此進行研究,報告如下。
1.1 實驗動物及分組:SPF級BALB/c-nu小鼠32只,雄性,鼠齡35 d,購于北京華阜康生物科技股份有限公司,由中國醫(yī)學科學院醫(yī)學動物研究所提供,合格證號:SCXK(京)2009-0004。將小鼠分為正常組(4只)、吻合組(4只)、注射NGF組(4只)和接種肉瘤細胞組(分為5個肉瘤細胞數(shù)量組,每組4只)。
1.2 藥品及試劑:S180小鼠肛門肉瘤細胞(購自中科院),Trka(E492)一抗為兔抗人多克隆抗體,p75為單克隆抗體,以上試劑由美國GBI公司提供,NGF由寧夏泰康元生物科技有限公司提供。
1.3 實驗方法
1.3.1 動物模型建立:接種肉瘤細胞組接種S180肉瘤細胞,接種于小鼠背部,細胞數(shù)量分別為1×104、1×105、2×105、1×106、2×106/mL,接種后切斷坐骨神經(jīng),隨后顯微鏡下吻合;注射NGF組切斷坐骨神經(jīng)后吻合,每只注射NGF 30 μg,使用注射用水溶解于2 mL中;吻合組切斷坐骨神經(jīng),顯微鏡下吻合,不注射NGF;正常組不采取任何操作。
1.3.2 NGF濃度測定和觀察NGF受體:模型建立后5周取小鼠尾靜脈血2 μl,按試劑說明行ELISA法測定NGF濃度;其后腹腔麻醉后以甲醛溶液灌注,取脊髓腰膨大處,病變組織包埋在石蠟塊里,切片,染色,顯微鏡下觀察NGF受體,胞漿有棕褐色或黃色顆?;虬咂邽镹GF受體陽性細胞[7-8]。
1.4 改良Tarlov評分標準:改良Tarlov評分[9]分為0~5分:0分,肢體無力;1分,肢體可見活動,不能負重;2分,肢體活動頻繁,不能負重;3分,可以站立但無法行走;4分,肢體可以行走但有障礙;5分,行走正常。
2.1 改良Tarlov評分和NGF濃度檢測:接種肉瘤細胞各組Tarlov評分和NGF濃度均高于吻合組和注射NGF組(P<0.05),接種肉瘤細胞各組NGF濃度均高于正常組(P<0.05),見表1。接種肉瘤細胞組神經(jīng)生長因子濃度曲線。

2.2 觀察Trka和p75數(shù)量:脊髓腰膨大處接種肉瘤細胞組Trka受體和p75受體染色均明顯高于正常組,說明受體都有增加。
1951年Levi-Montalcini[10-11]發(fā)現(xiàn)小鼠肉瘤細胞S180能夠刺激雞胚交感神經(jīng)節(jié)的生長,其后她們將分離純化得到的物質命名為NGF,通過外源性給予NGF或細胞移植技術內源性給予NGF均能促進神經(jīng)損傷的恢復,但它們之間的恢復效果沒有比較,另外采用的內源性分泌NGF不能逃避宿主的免疫反應,不能準確反映NGF的效果。本實驗裸鼠無免疫系統(tǒng),同時S180可以持續(xù)分泌NGF,解決了NGF濃度問題,改良評分結果優(yōu)于其他方式,說明內源性NGF更能促進神經(jīng)損傷的修復。
不同濃度NGF對周圍神經(jīng)損傷修復效果有差異[12],本研究表明,實驗接種組間NGF濃度是有差異的,且都高于其他實驗組,但接種組之間修復無差異性,說明當NGF濃度達到某一程度后對神經(jīng)修復的效果就不存在劑量依賴性,高于一定濃度時可以明顯促進神經(jīng)損傷的修復。
接種組較其他組NGF受體Trka和p75均出現(xiàn)上調,Trka為高親和力受體,p75為低親和力受體,NGF對神經(jīng)修復的作用通過對受體的調節(jié)來實現(xiàn)。Trka和p75介導了神經(jīng)細胞的分化增殖凋亡,通過調控神經(jīng)細胞的生長周期來影響修復,不同受體在NGF濃度下對神經(jīng)細胞電位的影響不同[18],實驗表明NGF不是通過單一提高某一受體而促進神經(jīng)修復的。

The effects of nerve growth factor S180 on repairing sciatic nerve in mice

Objective To study the repair function of nerve growth factor (NGF) which secrete by sarcomas 180 and extrinsic nerve growth factor on sciatic nerve.Methods Rats were randomly divided into eight groups,included five groups,NGF group,normal group and four rats in each group,except normal group.Others were cut sciatic nerve and anastomosed,and observed the hindlimb function,NGF concentration,Trka and p75.Results Sarcomas 180 secreted nerve growth factor,also the concentration of nerve growth factor were different,the rats inoculated group were better recovery than those in other groups.The Trka and p75 which rats inoculated had elevation than those in other groups.Neurotrophin receptors Trka and p75.Conclusion Rats inoculated sarcomas can secrete endogenous nerve growth factor,it has a good effects than extrinsic nerve growth factor.
