陸地棉核心種質(zhì)表型性狀遺傳多樣性分析及綜合評(píng)價(jià)

【目的】分析陸地棉核心種質(zhì)的遺傳多樣性和表型性狀遺傳變異規(guī)律,并探討核心種質(zhì)的綜合評(píng)價(jià)方法?!痉椒ā坷?7個(gè)表型性狀數(shù)據(jù)分析419份陸地棉核心種質(zhì)的遺傳多樣性。用Shannon-weaver信息多樣性指數(shù)計(jì)算表型性狀的遺傳多樣性,用Nei’s 1973法計(jì)算表型性狀遺傳距離,并使用NTSYS-pc 2.20q軟件對(duì)核心種質(zhì)進(jìn)行聚類(lèi)分析;用SAS9.2對(duì)表型性狀數(shù)據(jù)進(jìn)行最佳線性無(wú)偏估計(jì)(BLUE),然后根據(jù)最佳線性無(wú)偏估計(jì)值計(jì)算出表型性狀的最佳值。同時(shí),結(jié)合主成分、回歸和相關(guān)分析,研究核心種質(zhì)的綜合評(píng)價(jià)指標(biāo)和方法?!窘Y(jié)果】核心種質(zhì)表型性狀分析發(fā)現(xiàn),單株鈴數(shù)、單鈴重、衣分、子指等性狀的變異系數(shù)均較大,變異系數(shù)超過(guò)10%。而斷裂比強(qiáng)度、馬克隆值以及上半部平均長(zhǎng)度的變異程度較小,變異系數(shù)均在10%以下。方差分析發(fā)現(xiàn),各表型性狀地點(diǎn)間、年份間、地點(diǎn)和年份間、品種間均有極顯著差異;不同地理來(lái)源的種質(zhì)表型性狀差異較大,長(zhǎng)江流域地理來(lái)源的種質(zhì)生育期、伸長(zhǎng)率、上半部平均長(zhǎng)度、衣分等性狀均高于其他的地理來(lái)源,西北內(nèi)陸地理來(lái)源的種質(zhì)纖維強(qiáng)度,單鈴重、整齊度指數(shù)、株高、紡紗均勻性指數(shù)等綜合性狀最好,美國(guó)種質(zhì)的產(chǎn)量和纖維品質(zhì)的性狀優(yōu)于其他國(guó)家的總和。表型性狀的遺傳多樣性指數(shù)范圍為0.351—3.796,平均為1.715。分析不同地理來(lái)源種質(zhì)的遺傳多樣性,發(fā)現(xiàn)黃河流域的遺傳多樣性和遺傳豐富度最高,中國(guó)南部區(qū)域最低。類(lèi)群聚類(lèi)結(jié)果發(fā)現(xiàn)陸地棉整體分散,沒(méi)有比較明顯的類(lèi)群關(guān)系,部分具有相似特點(diǎn)的種質(zhì)聚類(lèi)13個(gè)組群。核心種質(zhì)綜合評(píng)價(jià)表明在累計(jì)貢獻(xiàn)百分比高于85%時(shí),共發(fā)現(xiàn)7個(gè)主成分,陸地棉核心種質(zhì)的表型性狀綜合值(值)平均為1.740,來(lái)自澳大利亞的N74-250值最高(2.302),遼陽(yáng)綠絨棉的值最低(0.624)。對(duì)17個(gè)表型性狀與值的相關(guān)分析,發(fā)現(xiàn)除馬克隆值、子指和黃度外,單鈴重、衣分、斷裂比強(qiáng)度、上半部纖維長(zhǎng)度等14個(gè)表型性狀與值間的相關(guān)性具有極顯著差異,最后構(gòu)建了以吐絮期、單鈴重、伸長(zhǎng)率、花期、馬克隆值、株高、果枝數(shù)、紡紗均勻性指數(shù)8個(gè)表型性狀為自變量的回歸方程,綜合評(píng)價(jià)核心種質(zhì)資源?!窘Y(jié)論】中國(guó)保存的陸地棉核心種質(zhì)具有較為豐富的遺傳多樣性,不同地理來(lái)源遺傳變異有較大的差異,不同生態(tài)區(qū)的核心種質(zhì)具有獨(dú)特的性狀特性。

【研究意義】棉花種質(zhì)資源是棉花遺傳改良的基礎(chǔ)[1-3],其數(shù)量在一定程度上制約著品種改良的廣度和深度。中國(guó)共收集和保存8 828份棉花種質(zhì)資源,其中,陸地棉7 362份、陸地棉野生種系350份、海島棉633份、亞洲棉433份、草棉18份、野生種32份,種質(zhì)資源的保存數(shù)量居世界第四位[4-5]。隨著棉花種質(zhì)資源收集和保存數(shù)量的日益增加,高效利用這些種質(zhì)資源顯得十分困難,評(píng)價(jià)這些種質(zhì)顯得越來(lái)越重要?!厩叭搜芯窟M(jìn)展】Frankel等[6]和Brown等[7]在此基礎(chǔ)上提出了核心種質(zhì)(core germplam)的概念,意圖以最少數(shù)量的遺傳資源最大限度地保存整個(gè)資源群體的遺傳多樣性和整個(gè)群體的地理分布,以此來(lái)提高種質(zhì)資源的利用和基因挖掘的效率。目前,小麥[8]、水稻[9]、玉米[10]、大豆[11]等作物核心種質(zhì)已構(gòu)建完成,大量作物的核心種質(zhì)正在構(gòu)建中。中國(guó)農(nóng)業(yè)科學(xué)院棉花研究所種質(zhì)資源課題組依托國(guó)家棉花種質(zhì)資源中期庫(kù)保存的陸地棉種質(zhì)資源優(yōu)勢(shì),已經(jīng)初步完成了陸地棉核心種質(zhì)的構(gòu)建,將進(jìn)一步進(jìn)行遺傳多樣性分析。遺傳多樣性分析方法有很多,主要從形態(tài)學(xué)數(shù)據(jù)(主要是表型數(shù)據(jù))[12-14]、細(xì)胞水平[15]、生物化學(xué)記[16]和DNA分子標(biāo)記[17-32]4個(gè)方面進(jìn)行分析。陳光等[12]對(duì)43份陸地棉基礎(chǔ)種質(zhì)按照不同時(shí)期、來(lái)源、生態(tài)區(qū)分別進(jìn)行了表型性狀的遺傳多樣性研究,發(fā)現(xiàn)基礎(chǔ)種質(zhì)間在農(nóng)藝性狀、品質(zhì)、產(chǎn)量等表型性狀上差異顯著或極顯著;衛(wèi)澤等[13]研究了國(guó)內(nèi)外57份棉花種質(zhì)的遺傳多樣性,基于表型性狀聚類(lèi)結(jié)果和分子標(biāo)記聚類(lèi)的結(jié)果大體一致,但存在差異;周忠麗等[14]通過(guò)對(duì)200份不同地理來(lái)源的亞洲棉代表性樣本進(jìn)行了表型遺傳多樣性分析,表明亞洲棉表型多樣性豐富,種質(zhì)間表型性狀差異顯著或極顯著。但是利用少量的棉花種質(zhì)表型數(shù)據(jù)來(lái)了解種群的遺傳變異狀況是不夠的,必須擴(kuò)大群體的數(shù)目以及多種特色的種質(zhì)材料(核心種質(zhì))來(lái)分析比較可靠。關(guān)于細(xì)胞標(biāo)記和生物化學(xué)標(biāo)記研究較少且結(jié)果復(fù)雜,限制其廣泛應(yīng)用。目前研究最廣最常用的方法是相繼發(fā)展的DNA分子標(biāo)記技術(shù),如RFLP[17]、RAPD[18-20,22]、ISSR[19-21]、AFLP[22-24]、SSR[25-30]、SCAR[31]、SNP[32]等,這些分子標(biāo)記技術(shù)已經(jīng)被廣泛地應(yīng)用到棉花種質(zhì)資源遺傳多樣性研究中?!颈狙芯壳腥朦c(diǎn)】陸地棉是中國(guó)栽培面積最大、產(chǎn)量最高的栽培種,因其龐大的種質(zhì)資源數(shù)目給研究帶來(lái)不便,其多達(dá)幾十種的表型性狀給田間調(diào)查和研究均帶來(lái)較大困難,國(guó)內(nèi)對(duì)于陸地棉核心種質(zhì)多樣性的研究和綜合評(píng)價(jià)尚未見(jiàn)報(bào)道?!緮M解決的關(guān)鍵問(wèn)題】本研究利用表型性狀對(duì)現(xiàn)有陸地棉核心種質(zhì)進(jìn)行遺傳多樣性分析,以揭示陸地棉核心種質(zhì)的表型遺傳規(guī)律及篩選出相對(duì)合理的表型評(píng)價(jià)指標(biāo),為陸地棉育種和基礎(chǔ)的研究提供一定的參考和依據(jù)。
1.1 試驗(yàn)材料
按照品種特性、農(nóng)藝性狀以及廣泛的地理來(lái)源差異,從中國(guó)農(nóng)業(yè)科學(xué)院棉花研究所中期庫(kù)中保存的7 362份陸地棉種質(zhì)中根據(jù)品種主要突變性狀和品種類(lèi)型分為11組群,在分組的基礎(chǔ)上利用21個(gè)表型性狀,用非加權(quán)類(lèi)平均聚類(lèi)分析法,構(gòu)建了281份陸地棉核心種質(zhì),占全部種質(zhì)資源總量的4.71%。利用不同性狀的均值測(cè)驗(yàn)、方差測(cè)驗(yàn)、變異系數(shù)、多樣性指數(shù)檢驗(yàn)、均值、極差、表型方差、變異系數(shù)、均值差異百分率、方差差異百分率、極差符合率、變異系數(shù)變化率、表型變異百分率及主成分分析等參數(shù)進(jìn)行核心種質(zhì)代表性檢驗(yàn)和評(píng)價(jià),所構(gòu)建的陸地棉核心種質(zhì)的表型變異解釋率達(dá)到80.67%,利用相似的原理,構(gòu)建了419份陸地棉核心種質(zhì)[4](植物資源遺傳學(xué)報(bào)已接受)。這些種質(zhì)來(lái)自中國(guó)、美國(guó)等17個(gè)國(guó)家(電子附表1),其中,國(guó)內(nèi)材料分別來(lái)自23個(gè)?。ㄊ校?。為了便于分析,根據(jù)材料地理來(lái)源將材料劃分為7個(gè)生態(tài)區(qū):長(zhǎng)江流域棉區(qū)(100份)、黃河流域棉區(qū)(164份)、西北內(nèi)陸棉區(qū)(28份)、北部早熟棉區(qū)(19份)、中國(guó)南部棉區(qū)(9份)、美國(guó)(55份)以及其他國(guó)家(44份)。
1.2 試驗(yàn)設(shè)計(jì)
田間試驗(yàn)于2014年和2015年連續(xù)兩年分別在河南安陽(yáng)中國(guó)農(nóng)業(yè)科學(xué)院棉花研究所實(shí)驗(yàn)農(nóng)場(chǎng)、湖北荊州長(zhǎng)江大學(xué)實(shí)驗(yàn)基地和甘肅敦煌甘肅省農(nóng)業(yè)科學(xué)院經(jīng)濟(jì)作物研究所實(shí)驗(yàn)基地進(jìn)行。小區(qū)面積6 m2,每小區(qū)1行,行距0.8 m、株距0.3 m,觀測(cè)道0.8 m,隔離道1.2 m,每重復(fù)20株。試驗(yàn)采取完全隨機(jī)區(qū)組設(shè)計(jì),3次重復(fù),正常的大田栽培管理。
1.3 數(shù)據(jù)采集
根據(jù)陸地棉材料的特點(diǎn),由于試驗(yàn)材料多,小區(qū)多,為了控制田間試驗(yàn)系統(tǒng)誤差一方面選擇肥力相差不大的試驗(yàn)田,另一方面種植方式和調(diào)查方法均需要統(tǒng)一規(guī)范,最后要增加調(diào)查株數(shù),選取每小區(qū)10株(不包括兩端植株)作調(diào)查,生育期按小區(qū)調(diào)查,其他性狀調(diào)查在取樣行中調(diào)查。測(cè)定性狀包括:株高(plant height,PH,cm)、果枝數(shù)(sympodial brand number,SBNU)、果枝節(jié)位(sympodial brand type,SBT)、單株鈴數(shù)(bolls per plant,BPP)、開(kāi)花期(flowering date,F(xiàn)D)、吐絮期(boll open date,BOD)。每行收取植株中部完全吐絮鈴50個(gè),室內(nèi)測(cè)定衣分(lint percentage,LP,%)、單鈴重(boll weight,BW,g)、子指(seed index,SI,g)、伸長(zhǎng)率(elongation,EL,%)、斷裂比強(qiáng)度(fiber strength,F(xiàn)S,cN/tex)、整齊度指數(shù)(length uniformity,LU)、馬克隆值(micronaire,MIC)、反射率(reflectance rate,F(xiàn)R,%)、紡紗均勻性指數(shù)(spinning consistent index,SCI)、上半部平均長(zhǎng)度(upper half mean length,UHML,mm)、黃度(yellowness,YD)等數(shù)據(jù)。皮棉樣品送河北農(nóng)業(yè)大學(xué)進(jìn)行纖維品質(zhì)檢測(cè)。數(shù)據(jù)采集時(shí)期和采集方法均參照棉花種質(zhì)資源描述規(guī)范[33]。
1.4 數(shù)據(jù)分析
1.4.1 表型性狀的描述性統(tǒng)計(jì)方法 用SAS9.2對(duì)419份陸地棉種質(zhì)群體17個(gè)表型性狀進(jìn)行描述性統(tǒng)計(jì),SAS規(guī)定當(dāng)樣本含量n≤2000時(shí),結(jié)果以Shapiro-Wilk(W檢驗(yàn))為準(zhǔn),根據(jù)樣本量計(jì)算一個(gè)統(tǒng)計(jì)量,即檢驗(yàn)統(tǒng)計(jì)量W把樣本分布的形狀和正態(tài)分布進(jìn)行比較,比較得出一個(gè)數(shù)值w(0<w<1,及即實(shí)際的顯著水平)來(lái)描述對(duì)這個(gè)想法的可疑程度,即當(dāng)w<W時(shí)數(shù)據(jù)符合正態(tài)分布。
1.4.2 表型性狀的遺傳多樣性指數(shù)及遺傳豐富度計(jì)算 模糊隸屬函數(shù)計(jì)算出各個(gè)表型性狀的隸屬函數(shù)值,及將各個(gè)表型性狀定義到[0,1]閉區(qū)間[34]。

式中,(x為某種質(zhì)第個(gè)表型性狀的隸屬函數(shù)值,x為某種質(zhì)的第個(gè)表型性狀的最大值和最小值,每一級(jí)別性狀的相對(duì)頻率用于計(jì)算遺傳多樣性指數(shù)[23]。表型性狀遺傳多樣性分析用Shannon-weaver信息多樣性指數(shù)(index of genetic diversity,)來(lái)表示,其計(jì)算公式:,表示任一組中某一性狀的多樣性程度。其中P為某性狀第級(jí)別的種質(zhì)份數(shù)占總種質(zhì)材料的百分比,表示自然對(duì)數(shù)。
平均遺傳多樣性指數(shù):
遺傳豐富度(genetic richness index,)是指全部性狀的級(jí)別總數(shù),計(jì)算公式:=∑/∑,其中∑為全部種質(zhì)的某一性狀的級(jí)別總數(shù),∑為某一組該性狀的級(jí)別總數(shù)[35]。
1.4.3 表型性狀的遺傳距離計(jì)算 采用Nei’1973[36]方法計(jì)算Nei式遺傳距離:
式中,X是群體第個(gè)性狀第個(gè)級(jí)別的分布頻率。
聚類(lèi)分析采用高偉[37]、劉方等[38]的方法,使用軟件為NTSYS-pc 2.20q。
1.4.4 核心種質(zhì)資源表型性狀的綜合評(píng)價(jià) 為了得到基因型的最佳表現(xiàn)值,盡可能地減小因不同時(shí)間和不同地點(diǎn)所引起的誤差,用SAS9.2通過(guò)混合線性模型:y=+x+z+v+ε,其中y是田間試驗(yàn)?zāi)橙后w中第個(gè)材料在第個(gè)矩陣模塊中的表型值;ε是整體均值和殘差誤差的綜合值;某種質(zhì)資源群體x田間試驗(yàn)數(shù)據(jù)z與v矩陣作為隨機(jī)效應(yīng)模型;對(duì)于某個(gè)種質(zhì)資源群體中某個(gè)材料的某個(gè)性狀的最佳線性無(wú)偏估計(jì)值(the best linear unbiased estimate,BLUE)為,對(duì)419份陸地棉核心材料的表型性狀數(shù)據(jù)進(jìn)行最佳線性無(wú)偏估計(jì)(BLUE),進(jìn)而計(jì)算出兩年三地17個(gè)表型性狀的最佳線性無(wú)偏估計(jì)值,利用最佳線性無(wú)偏估計(jì)值進(jìn)行描述性統(tǒng)計(jì)、方差分析、相關(guān)分析、聚類(lèi)分析及主成分分析。
核心種質(zhì)的綜合評(píng)價(jià)方法[34]:(1)將標(biāo)準(zhǔn)化的核心種質(zhì)表型數(shù)據(jù)代入每個(gè)主成分中,計(jì)算各主成分的得分,再利用模糊隸屬函數(shù)對(duì)各主成分歸一化處理,計(jì)算出各主成分的權(quán)重系數(shù),最后計(jì)算各個(gè)核心材料的綜合得分(值)。(2)以綜合得分(值)為依變量,以17個(gè)表型性狀作為自變量,并通過(guò)逐步回歸分析構(gòu)建回歸方程,進(jìn)一步篩選核心種質(zhì)資源的綜合評(píng)價(jià)指標(biāo)。
2.1 陸地棉核心種質(zhì)表型性狀的遺傳多樣性分析
2.1.1 陸地棉核心種質(zhì)間表型性狀差異 對(duì)19份陸地棉種質(zhì)群體17個(gè)表型性狀進(jìn)行描述性統(tǒng)計(jì)(表1),發(fā)現(xiàn)全部數(shù)據(jù)均符合正態(tài)分布,單株鈴數(shù)(BPP)變幅10.26—22.60個(gè),均值為17.02個(gè);單鈴重(BW)變幅3.34—7.45 g,均值5.27 g;衣分(LP)變幅14.99%—49.64%,均值40.41%;子指(SI)變幅7.83—17.64 g,均值10.50 g。研究表明,單株鈴數(shù)、單鈴重、衣分、子指等性狀的變異系數(shù)均較大,變異系數(shù)超過(guò)10%,如無(wú)極一枝花的單鈴重7.45 g,南丹巴地大花單鈴重為3.43 g;美國(guó)材料V83-013衣分高達(dá)49.64%,而稀絮H10衣分只有14.99%;無(wú)極一枝花子指17.64 g,中6331子指7.83 g。而斷裂比強(qiáng)度、馬克隆值以及上半部平均長(zhǎng)度的變異程度較小,變異系數(shù)均在10%以下,說(shuō)明供試材料中纖維品質(zhì)性狀的變異程度低于產(chǎn)量性狀的變異。方差分析后發(fā)現(xiàn)(表1),各表型性狀地點(diǎn)間、年份間、地點(diǎn)和年份間、品種間均有極顯著差異,表明供試的種質(zhì)群體間的遺傳變異極其豐富,有利于表型性狀的優(yōu)異基因位點(diǎn)的發(fā)掘,同時(shí)說(shuō)明各表型性狀受環(huán)境影響較大。

2.1.2 不同地理來(lái)源核心種質(zhì)的表型性狀差異 9份陸地棉種質(zhì)中美國(guó)材料占13.13%,其他國(guó)家的材料占10.50%,而中國(guó)自育的材料中長(zhǎng)江流域棉區(qū)材料占23.63%,黃河流域棉區(qū)占39.14%,西北內(nèi)陸地理來(lái)源占6.68%,北部特早熟棉區(qū)占4.53%,南方地理來(lái)源占2.15%。
通過(guò)比較從不同國(guó)家引進(jìn)的材料,發(fā)現(xiàn)不同來(lái)源的種質(zhì)間的表型性狀差異明顯,17個(gè)表型性狀中,除子指和黃度2個(gè)性狀差異不顯著外,其他15個(gè)性狀差異均達(dá)到極顯著或顯著水平(表2),說(shuō)明不同地理來(lái)源間種質(zhì)的表型性狀存在較大的差異。其中,長(zhǎng)江流域地理來(lái)源的生育期、伸長(zhǎng)率、上半部平均長(zhǎng)度、衣分等性狀均高于其他的地理來(lái)源,馬克隆值與西北內(nèi)陸地理來(lái)源一樣均低于其他地理來(lái)源;北部早熟區(qū)的生育期最短,第一果枝節(jié)位最小,說(shuō)明該地理來(lái)源的早熟性好;西北內(nèi)陸地理來(lái)源纖維強(qiáng)度、單鈴重、整齊度指數(shù)、株高、紡紗均勻性指數(shù)等綜合性狀最好;南方地理來(lái)源單株鈴數(shù)、果枝數(shù)最多,馬克隆值最大;長(zhǎng)江流域種質(zhì)的產(chǎn)量和纖維品質(zhì)大部分性狀均優(yōu)于其他國(guó)家。以上結(jié)果表明不同陸地地理來(lái)源的表型性狀差異較大,受地理影響和種質(zhì)適應(yīng)性選擇變異,不同生態(tài)區(qū)種質(zhì)具有獨(dú)特性的形狀特性。

2.1.3 不同地理來(lái)源核心種質(zhì)的多樣性及遺傳豐富度 遺傳多樣性指數(shù)涵蓋了遺傳豐富度,但多樣性更側(cè)重于性狀的等級(jí)分布均勻度,單純地用它去衡量種質(zhì)群體的遺傳多樣性的程度是不全面的,因此,在評(píng)價(jià)種質(zhì)資源群體的遺傳多樣性時(shí),需要對(duì)這兩個(gè)指標(biāo)進(jìn)行權(quán)衡分析[4]。
通過(guò)分析不同地理來(lái)源的遺傳多樣性和遺傳豐富度(表3),發(fā)現(xiàn)平均遺傳多樣性指數(shù)分布范圍為1.251—1.667,平均遺傳豐富度分布在0.310—0.826,而黃河流域的遺傳多樣性和遺傳豐富度最高,中國(guó)南部區(qū)域最低。中國(guó)長(zhǎng)江流域和西北2個(gè)主產(chǎn)棉區(qū),以及來(lái)自美國(guó)的種質(zhì)的多樣性也較好。黃河、長(zhǎng)江流域和西北3個(gè)地理來(lái)源的棉花是中國(guó)陸地棉的主產(chǎn)區(qū),上百年以來(lái)經(jīng)過(guò)多種途徑的品種改良,不但提高了品種的綜合性能,而且豐富了品種的遺傳變異。美國(guó)種質(zhì)是中國(guó)早期引種較多的,中國(guó)大部分自育品種多來(lái)源于此,有較為豐富的遺傳變異,其遺傳變異處在0.335—3.294,最小的遺傳變異是伸長(zhǎng)率,只有0.335,而最大的遺傳變異則是達(dá)到了3.294的紡紗均勻性指數(shù),整個(gè)遺傳變異性較好。北部早熟地理來(lái)源以早熟品種為主,南方地理來(lái)源因環(huán)境因素所育品種相對(duì)較少,美國(guó)以外的其他國(guó)家初期引種材料并沒(méi)有較多的衍生品種,因此,中國(guó)北部特早熟、南部山地棉區(qū)和除美國(guó)以外地理來(lái)源的種質(zhì)遺傳多樣性和遺傳豐富度均不高。

2.1.4 型遺傳距離與聚類(lèi) 表型性狀對(duì)419份陸地棉核心種質(zhì)進(jìn)行聚類(lèi)(圖1),結(jié)果表明,中國(guó)的陸地棉種質(zhì)在表型性狀上的聚類(lèi)整體分散沒(méi)有比較明顯的類(lèi)群關(guān)系,部分具有相似特點(diǎn)的材料聚在一起,說(shuō)明這些種質(zhì)之間可能具有獨(dú)特的遺傳背景,涵蓋比較豐富的遺傳變異。所有種質(zhì)的相似系數(shù)變動(dòng)范圍是0.04—0.90,平均為0.122。在相似系數(shù)0.316處可將419份材料分為13類(lèi),聚類(lèi)的結(jié)果與生態(tài)區(qū)的關(guān)系并不明顯。
第Ⅰ類(lèi)有22份材料;第Ⅱ類(lèi)中有遼陽(yáng)綠絨棉、綠絮1號(hào)和Greenlint 3份種質(zhì),均為綠色棉材料,419份陸地棉材料中全部的綠絮的材料都聚在了一起;大部分材料都聚集在第Ⅲ類(lèi),這組材料沒(méi)有明顯的特點(diǎn),包含大鈴(軍棉1號(hào)、無(wú)極一枝花、蘇遠(yuǎn)04-3等)、高衣分(泗棉3號(hào)、荊55173、蘇棉12等)等豐產(chǎn)性較好的材料,高產(chǎn)、優(yōu)質(zhì)的材料岱紅岱、洞庭1號(hào)等,抗性較強(qiáng)的材料Miscot7803-52、泗陽(yáng)630、蘇抗310等,陸地棉標(biāo)準(zhǔn)系TM-1,纖維高比強(qiáng)材料J02-247;第Ⅳ類(lèi)種質(zhì)均是早熟陸地棉,如酒棉8號(hào)、新陸早3號(hào)等;第Ⅴ類(lèi)只有3份材料,均來(lái)自黃河流域地理來(lái)源;第Ⅵ類(lèi)是棕色棉,如CC28;第Ⅶ類(lèi)是豐產(chǎn)、廣適、推廣面積大的品種;第Ⅷ類(lèi)是15份材料,是一些與抗逆性狀有關(guān)的材料;第Ⅸ類(lèi)中新疆的材料居多且生育期比較短,纖維品質(zhì)也比較好;第Ⅹ類(lèi)是纖維品質(zhì)好的種質(zhì)如蘇優(yōu)系列材料、川206、蔡科510、J02-508等;第Ⅺ類(lèi)中僅有稀絮H10一份材料;第Ⅻ類(lèi)材料的纖維品質(zhì)較差,馬克隆值較大,纖維短粗;紅雞腳葉單獨(dú)分在第ⅩⅢ類(lèi)。
2.2 陸地棉核心種質(zhì)表型性狀綜合評(píng)價(jià)
2.2.1 表型性狀的相關(guān)性及主成分分析 對(duì)17個(gè)表型性狀進(jìn)行相關(guān)性分析(表4),結(jié)果表明,性狀間存在不同程度相關(guān)性且絕大部分為顯著或極顯著相關(guān)。吐絮期與花期、果枝節(jié)位呈顯著正相關(guān);單株鈴數(shù)與果枝數(shù)呈顯著正相關(guān),而與子指呈顯著負(fù)相關(guān);單鈴重與子指、長(zhǎng)度整齊度呈顯著正相關(guān);伸長(zhǎng)率與斷裂比強(qiáng)度、長(zhǎng)度整齊度、紡紗均勻性指數(shù)、上半部分長(zhǎng)度都呈顯著正相關(guān);花期與果枝數(shù)呈顯著正相關(guān);斷裂比強(qiáng)度與長(zhǎng)度整齊度、紡紗均勻性指數(shù)、上半部分長(zhǎng)度顯著正相關(guān);衣分與長(zhǎng)度整齊度、子指均顯著相關(guān);長(zhǎng)度整齊度與紡紗均勻性指數(shù)、上半部分長(zhǎng)度顯著正相關(guān);第一果枝節(jié)位與上半部分長(zhǎng)度顯著正相關(guān);紡紗均勻性指數(shù)與上半部分長(zhǎng)度顯著正相關(guān);以上信息表明這些性狀之間存在或大或小的相關(guān)性,因此如果直接依據(jù)這些信息評(píng)價(jià)這些核心種質(zhì)材料特性會(huì)影響其真實(shí)性。

為了消除這類(lèi)不利因素的影響,用主成分分析對(duì)核心種質(zhì)綜合評(píng)價(jià),在累計(jì)貢獻(xiàn)百分比高于85%時(shí),共發(fā)現(xiàn)7個(gè)主成分(表5)。其主成分的特征值分別為:4.935、2.798、2.526、1.392、1.182、1.00和0.74;主成分貢獻(xiàn)率分別為29.03%、16.46%、14.86%、8.19%、6.95%、5.85%和4.36%。前7個(gè)主成分的累計(jì)貢獻(xiàn)率達(dá)到85.70%,可以解釋不同陸地棉品種的主要表型性狀的大部分信息。第一主成分的特征值及方差貢獻(xiàn)率最大,所包含的表型形狀信息也比較多,但其貢獻(xiàn)率只有29.03%,可能的原因是這17個(gè)表型性狀包括了農(nóng)藝性狀、產(chǎn)量性狀以及纖維品質(zhì)性狀等,這些性狀間的差異較大,線性關(guān)系比較少。
第一主成分的伸長(zhǎng)率、斷裂比強(qiáng)度、整齊度指數(shù)、紡紗均勻性及上半部平均長(zhǎng)度的特征向量值最大,且均為正值,可以理解為第一主成分是跟纖維品質(zhì)相關(guān)的綜合指標(biāo),第一主成分的值越高,供試陸地棉的纖維品質(zhì)就越好。第二主成分的單株鈴數(shù)、花期、衣分、第一果枝節(jié)位等特征向量值較大,其次子指的特征向量為負(fù)值且絕對(duì)值較大,說(shuō)明第二主成分是與產(chǎn)量相關(guān)的綜合指標(biāo)。第三主成分是跟生育期有關(guān)的因子。第四主成分是控制反射率的因子。第五主成分是與單鈴重有關(guān)的因子。第六主成分是控制果枝數(shù)的因子。第七主成分是與馬克隆值有關(guān)的因子,控制棉花纖維的粗細(xì)。以上結(jié)果說(shuō)明中國(guó)陸地棉核心種質(zhì)資源群體纖維品質(zhì)的多樣性較高,其次是產(chǎn)量。

2.2.2 核心種質(zhì)的表型性狀綜合評(píng)價(jià) 將標(biāo)準(zhǔn)化的核心種質(zhì)表型數(shù)據(jù)代入上述7個(gè)主成分中,可得各個(gè)種質(zhì)資源7個(gè)主成分的得分(電子附表2),其中第一主成分的線性方程為:

用模糊隸屬函數(shù)將7個(gè)主成分的得分進(jìn)行歸一化處理后,計(jì)算出7個(gè)主成分的權(quán)重系數(shù)為:0.339、0.192、0.173、0.096、0.081、0.068和0.051,再利用各核心材料的綜合得分(值),對(duì)核心種質(zhì)進(jìn)行綜合評(píng)價(jià)。結(jié)果表明,陸地棉核心種質(zhì)的表型性狀綜合值(值)平均為1.740,來(lái)自澳大利亞的N74-250的值最高(2.302),中國(guó)北部特早熟地理來(lái)源的遼陽(yáng)綠絨棉的值最低(0.624),說(shuō)明N74-250的綜合性能最好,其單鈴重、衣分、上半部平均長(zhǎng)度、斷裂比強(qiáng)度、馬克隆值分別為6.42 g、40.98%、29.28 mm、29.71(cN/tex)和4.32;而遼陽(yáng)綠絨棉的綜合性能最差。對(duì)17個(gè)表型性狀與值的相關(guān)分析(表6),結(jié)果表明,除馬克隆值、子指和黃度外,單鈴重、衣分?jǐn)嗔驯葟?qiáng)度、上半部纖維長(zhǎng)度等14個(gè)表型性狀與值間的相關(guān)性具有極顯著差異,其中值與果枝數(shù)為負(fù)相關(guān),與其余13個(gè)性狀為極顯著正相關(guān)。

同時(shí),用已經(jīng)求出的綜合得分(值)與表型性狀構(gòu)建回歸方程,篩選核心種質(zhì)資源的綜合評(píng)價(jià)指標(biāo)。構(gòu)建回歸方程為:

式中1、3、4、5、9、10、12、14等分別代表吐絮期、單鈴重、伸長(zhǎng)率、花期、馬克隆值、株高、果枝數(shù)、紡紗均勻性指數(shù)8個(gè)表型性狀。綜合得分(值)與表型性狀的回歸分析結(jié)果表明吐絮期、單鈴重、伸長(zhǎng)率、花期、馬克隆值、株高、果枝數(shù)、紡紗均勻性指數(shù)8個(gè)表型性狀回歸方程的通徑系數(shù)分別為0.261、0.498、0.168、0.225、0.124、0.132、0.164和0.112,其決定系數(shù)2為0.991,8個(gè)表型性狀可決定值的總變異為99.1%,值為246.495,方程結(jié)果極顯著,表明以上所述表型性狀可以作為核心種質(zhì)的綜合評(píng)價(jià)指標(biāo)。主成分綜合值與該回歸方程預(yù)測(cè)值的相關(guān)系數(shù)=0.995**,這說(shuō)明該方程可以用來(lái)綜合評(píng)價(jià)陸地棉核心種質(zhì)群體,且可靠性達(dá)到99.1%。
3.1 陸地棉核心種質(zhì)表型性狀遺傳多樣性
對(duì)表型性狀的統(tǒng)計(jì)發(fā)現(xiàn),單株鈴數(shù)、單鈴重、衣分、子指等性狀的變異系數(shù)均大于10%,而斷裂比強(qiáng)度、馬克隆值以及上半部平均長(zhǎng)度的變異系數(shù)在10%以下,變異的程度較小,說(shuō)明供試材料的纖維品質(zhì)性狀有關(guān)的變異程度低于產(chǎn)量性狀的變異,這與錢(qián)能[39]陸地棉遺傳多樣性與育種目標(biāo)性狀基因(QTL)的關(guān)聯(lián)分析的研究結(jié)果是一致的;不同地理來(lái)源的變異系數(shù)存在同樣的規(guī)律;產(chǎn)生這種現(xiàn)象的原因可能是產(chǎn)量性狀的地理和生態(tài)變異引起的遺傳變異較大,而纖維品質(zhì)較小。
419份陸地棉核心種質(zhì)聚類(lèi)結(jié)果表明,雖然大部分種質(zhì)之間沒(méi)有明顯的分類(lèi),但局部具有相似特點(diǎn)的材料依然聚成了一類(lèi),如高產(chǎn)、早熟、棕色棉、綠色棉、優(yōu)異纖維、稀絮等材料,通過(guò)表型性狀的遺傳分析,一方面說(shuō)明陸地棉種質(zhì)的遺傳基礎(chǔ)較為狹窄,中國(guó)初期引進(jìn)的基礎(chǔ)陸地棉栽培種主要從美國(guó)引進(jìn)[25],然而近期的研究表明美國(guó)陸地棉栽培種同樣存在遺傳多樣性變小,遺傳基礎(chǔ)狹窄等問(wèn)題[40]。另一方面,部分材料之間有明晰的類(lèi)群特征表明這些材料間有較大的遺傳變異,在后續(xù)的研究工作中可以通過(guò)分子標(biāo)記分析基因間的遺傳規(guī)律兩者結(jié)合能更加全面地解釋中國(guó)陸地棉種質(zhì)資源的遺傳變異,更好地為現(xiàn)代育種服務(wù)。
遺傳多樣性為種質(zhì)資源優(yōu)異基因資源的挖掘提供了重要信息,表型性狀是育種工作者進(jìn)行棉花育種研究的重要依據(jù)[12-14,41-42]。國(guó)外對(duì)陸地棉種質(zhì)遺傳多樣性研究的報(bào)道很多,F(xiàn)ang等[43]利用193份陸地棉栽培種進(jìn)行表型性狀遺傳多樣性及其關(guān)聯(lián)分析研究;Alves等[44]對(duì)巴西本土陸地棉的遺傳多樣性和種群結(jié)構(gòu)進(jìn)行了分析;Badigannavar等[45]分析了與種子性狀有關(guān)性狀的遺傳多樣性;Tyagi等[40]用SSR對(duì)381份美國(guó)陸地棉的種群結(jié)構(gòu)和遺傳多樣性進(jìn)行了分析。陳光等[25]用43份中國(guó)陸地棉基礎(chǔ)種質(zhì),分析了中國(guó)不同地理來(lái)源及其衍生品種的遺傳多樣性;Zhao等[46]利用分子標(biāo)記技術(shù)對(duì)157份陸地棉材料的種群結(jié)構(gòu)和遺傳多樣性進(jìn)行分析;趙戰(zhàn)勝等[47]分析了中國(guó)北疆早熟陸地棉的遺傳多樣性。本研究對(duì)中國(guó)已構(gòu)建陸地棉核心種質(zhì)的表型性狀進(jìn)行了遺傳多樣性分析,表明中國(guó)收集保存的陸地棉核心種質(zhì)遺傳多樣性較高,在不同地理來(lái)源的種質(zhì)在遺傳多樣性的表現(xiàn)上有差異。所選用的材料為中國(guó)現(xiàn)有陸地棉的核心材料,為后續(xù)的育種工作奠定了堅(jiān)實(shí)的基礎(chǔ)。
3.2 核心種質(zhì)表型性狀的綜合評(píng)價(jià)
表型性狀是結(jié)構(gòu)基因的功能表現(xiàn),是功能基因的產(chǎn)物,是長(zhǎng)期自然和人工選擇的結(jié)果,易受時(shí)間、環(huán)境因素影響,但表型性狀是種質(zhì)資源收集、保存、鑒定和利用的基礎(chǔ),其觀察結(jié)果是直觀的,利用表型性狀進(jìn)行多樣性分析是最直接的。分子標(biāo)記是在基因組水平上對(duì)個(gè)體基因結(jié)構(gòu)差異的部分揭示。
本研究結(jié)合了隸屬函數(shù)法與主成分分析法綜合評(píng)價(jià)了陸地棉核心種質(zhì),這一方法已在植物的遺傳多樣性評(píng)價(jià)中得到應(yīng)用[48],方法比較可靠,不僅可以評(píng)價(jià)各個(gè)品種的綜合表現(xiàn)的高低,同時(shí)能將推斷出評(píng)價(jià)某個(gè)種質(zhì)群體中的關(guān)鍵性狀[34]。結(jié)果表明,來(lái)自澳大利亞的N74-250的值最高,中國(guó)北部特早熟地理來(lái)源的遼陽(yáng)綠絨棉的值最低,表型性狀的綜合值與吐絮期、單鈴重、伸長(zhǎng)率、花期、斷裂比強(qiáng)度、衣分、長(zhǎng)度整齊度、株高、第一果枝節(jié)位、紡紗均勻性指數(shù)及上半部分纖維長(zhǎng)度與值之間極顯著相關(guān),因此綜合值也越高,上述各個(gè)性狀的表型值也較高,這為育種工作中親本的選擇提供了依據(jù)。
由于陸地棉的表型性狀較多,且各性狀間存在不同程度的相關(guān),阻礙了棉花種質(zhì)資源高效的研究與利用,因此,有必要篩選出相對(duì)重要的性狀進(jìn)行重點(diǎn)研究,加速在育種和農(nóng)業(yè)生產(chǎn)上的應(yīng)用。本研究結(jié)合主成分分析和逐步回歸分析,篩選出陸地棉種質(zhì)資源的評(píng)價(jià)方程及綜合指標(biāo)。研究結(jié)果表明吐絮期、單鈴重、伸長(zhǎng)率、花期、馬克隆值、果枝數(shù)、紡紗均勻性指數(shù)8個(gè)表型性狀為陸地棉種質(zhì)資源的重要性狀,可以作為核心種質(zhì)綜合評(píng)價(jià)的指標(biāo),在棉花性狀考察及育種時(shí)間中應(yīng)著重這8個(gè)性狀。生育期,產(chǎn)量性狀,品質(zhì)性狀本研究對(duì)中國(guó)陸地棉核心種質(zhì)表型性狀進(jìn)行遺傳多樣性評(píng)價(jià),為深入解析和闡明中國(guó)陸地棉核心種質(zhì)的重要復(fù)雜性狀的基因遺傳機(jī)理奠定了基礎(chǔ),進(jìn)而促進(jìn)這些種質(zhì)更加高效地為現(xiàn)代育種服務(wù)。
3.3 核心種質(zhì)在育種上的運(yùn)用
隨著時(shí)間的推移,遺傳多樣性不斷地降低。現(xiàn)代品種較之早期和中期品種的遺傳多樣性較低。隨著現(xiàn)代育種商業(yè)化的發(fā)展,在實(shí)際育種中,人們更注重的市場(chǎng)需要的品種和盡可能快的育出商業(yè)化的品種而忽略了品種的遺傳多樣性,這些商業(yè)化品種的遺傳多樣性越來(lái)越低的問(wèn)題越來(lái)越嚴(yán)重,在品種改良過(guò)程中由于現(xiàn)代品種改良注重產(chǎn)量、品質(zhì),目標(biāo)單一,遺傳背景狹窄,從而導(dǎo)致多樣性差,最終導(dǎo)致遺傳多樣性降低。
為解決這一問(wèn)題,從遺傳多樣性好的早期和中期品種中,篩選資源,轉(zhuǎn)育到近現(xiàn)代品種中,如本研究構(gòu)建的核心種質(zhì)群體,不僅代表了80%以上的遺傳變異,且有較多可供選擇的優(yōu)異資源,如較好的伸長(zhǎng)率、馬克隆值、紡紗均勻性指數(shù),較多的果枝數(shù)、較多的棉鈴和較大的單鈴重等,都可以作為改良現(xiàn)有品種,選育新品種的資源。同時(shí)也可以進(jìn)行遠(yuǎn)緣雜交,選育出較好的品種,但遠(yuǎn)緣雜交雖然有利于提高遺傳多樣性,選育出較好的品種,但由于雜交后代的瘋狂分離而使育種周期就更長(zhǎng),成本更高,這與現(xiàn)代育種理念不符,特別是現(xiàn)有的育種,相當(dāng)一部分是由種業(yè)公司完成,不可能允許這樣做。再者,也可以采用MAGIC (multiparent advanced generation inter-cross)群體等方法,MAGIC即多親本高級(jí)世代互交系,是一種比通常用的F2、RIL、CSSLs(2個(gè)親本獲得)等作圖群體更為復(fù)雜的群體,由于MAGIC群體應(yīng)用了多親本互交,其后代不會(huì)出現(xiàn)多態(tài)性下降,后代衰退的現(xiàn)象,該群體可以聚合多樣性的種質(zhì)資源,增加多樣性。但這樣的群體結(jié)構(gòu)比較難于把握,而且獲得這樣一個(gè)作圖群體也需要耗費(fèi)多年的時(shí)間才能完成。以上各種方法各有優(yōu)缺點(diǎn),但核心種質(zhì)庫(kù)的構(gòu)建是非常必要的,不管是改良現(xiàn)有品種,進(jìn)行遠(yuǎn)緣雜交,或是構(gòu)建MAGIC群體,都需要好的種質(zhì)資源做支撐。
中國(guó)陸地棉核心種質(zhì)的遺傳多樣性較高,不同地理來(lái)源遺傳變異有較大的差異,黃河流域棉區(qū)整體遺傳多樣性最好,其次是美國(guó);吐絮期、單鈴重、伸長(zhǎng)率、花期、馬克隆值、株高、果枝數(shù)、紡紗均勻性8個(gè)指標(biāo)可以作為核心種質(zhì)評(píng)價(jià)的綜合指標(biāo)。

Comprehensiveevaluation and genetic diversity analysis of phenotypic traits of core collection in upland cotton

【Objective】The objective of this study is to analyze the genetic diversity and the law of genetic variation of phenotypic traits and explore the comprehensive evaluation techniques for core collection in upland cotton.【Method】The genetic diversity of 419 core collections in upland cotton was analyzed with 17 phenotypic traits. Genetic diversity of phenotypic traits was calculated using Shannon-weaver Information Index. Phenotypic distance was analyzed using Nei’s 1973, and then NTSYS-pc 2.20q software was used for cluster analysis of core collections. With SAS9.2, the best linear unbiased estimate (BLUE) value was gotten from phenotypic trait data, and then the optimum value of phenotypic traits was calculated according to the best linear unbiased estimator. Meanwhile, the principal component, regression and correlation analysis were also used to study the comprehensive evaluation index and method of core collection. 【Result】 Based on the analysis of phenotypic traits of core collection, it was found that the variation of boll number, boll weight, lint percentage, seed index and other traits were greater with coefficient of variation over 10% , while the variation of fiber breaking strength, micronaire level and the upper half of the average length was smaller which their coefficient of variation was below 10%. Variance analysis found that there were significant differences in the phenotypic traits between various locations, between years, and between locations and years. The phenotypic traits of different geographical origin germplasms were quite different. The growing stage elongation, upper half mean length, lint percentage and other traits from the geographical origin of the Yangtze River areas were higher than those of other geographic origins, and the comprehensive properties including the fiber strength, boll weight, uniformity index, plant height, and spinning uniformity index from the Northwest areas were the best. The yield and fiber quality traits from the US germplasms were superior to the sum of the other countries. Genetic diversity indices of phenotypic traits were 0.351-3.796, with an average of 1.715. The result of analysis on the genetic diversity of different geographical origins indicated that the genetic diversity and genetic richness in the Yellow River areas was the highest, but that in south China was the lowest. Cluster analysis showed that the upland cotton lines were overall dispersed with no obvious groups, but the partial accessions with similar characteristics can be clustered into 13 groups. After comprehensive evaluating these core collections, seven principal components were found when the cumulative contribution rate was more than 85%. The comprehensive value () of phenotypic traits of core collections averaged 1.740. The highestvalue (2.302) was from Australia’s N74-250, and the lowestvalue (0.624) was from Liaoyang green cotton. Correlation analysis between 17 phenotypic traits and theirvalues illuminated that 14 phenotypic traits including boll weight, lint percentage, fiber strength, upper half mean length, etc. were very significantly correlated with thevalues, except micronaire, seed index and yellow givers. Finally, the regression equation was constructed for comprehensively evaluating the core collection with 8 phenotypic traits as the independent variables including boll opening stage, boll weight, elongation, flowering, micronaire, plant height, number of fruit branches, and spinning consistent index. 【Conclusion】The core collection of upland cotton from China has a rich genetic diversity. The difference in genetic variation of the germplasms with different geographical origins is great. The accessions from different ecological zones have unique characteristics.
