摘 要:天然產(chǎn)物Maoecrystal V是云南昆明植物所的孫漢董院士研究小組從香茶菜屬植物疏花毛萼香茶菜中分離得到的二萜化合物,它具有獨(dú)特的五環(huán)骨架,含有6個(gè)手性中心,其中包括4個(gè)連續(xù)手性中心,3個(gè)全碳季碳中心。天然產(chǎn)物Maoecrystal V具有重要的生理活性,體外實(shí)驗(yàn)表明它對(duì)HeLa細(xì)胞系有顯著的選擇性抑制作用。由于其復(fù)雜的結(jié)構(gòu)和重要的生物活性,在全世界范圍內(nèi)有多個(gè)著名有機(jī)合成研究小組開展了對(duì)Maoecrystal V的全合成研究。該研究于2010年完成了對(duì)該天然產(chǎn)物的首次全合成。 該論文對(duì)Maoecrystal V全合成的研究工作主要包括以下兩個(gè)方面:(1)以Wessely氧化去芳構(gòu)化和分子內(nèi)Diels-Alder反應(yīng)為關(guān)鍵反應(yīng)構(gòu)建了Maoecrystal V的四環(huán)結(jié)構(gòu)模型;(2)在模型研究的基礎(chǔ)上,嘗試了多種合成策略,最終完成了天然產(chǎn)物Maoecrystal V的首次全合成研究。

Abstract:Maoecrystal V is a novel diterpenoid which was isolated from the leaves of a Chinese medicinal herb called Isodon eriocalyx by Sun and co-workers. It has noval pentacyclic ring system and six chiral centers, including 4 continuous chiral centers,3 allcarbon quaternary centers. It shows potent selective inhibitory activity against HeLa cells. Because of the complexity of the structure and importance of its bio-acitivity, many synthetic groups carry out the total synthesis toward maoecrystal V. The first total synthesis was completed by our group in 2010. Our synthetic study towards this natural product mainly focuses on two aspects:(1)Using Wessly oxidative dearomatization and intramolecular Diels-Alder to construct the tetracyclic model system of Maoecrystal V;(2)Based on the model study, we accomplished the first total synthesis of natural product Maoecrystal V.
