        采自海南省的環(huán)紋炭團菌屬3個中國新記錄種

        摘""要:在形態(tài)學研究和分子系統(tǒng)發(fā)育分析的基礎上,本研究報道了采集自海南省的環(huán)紋炭團菌屬3個中國新記錄種,分別為白色環(huán)紋炭團菌(Annulohypoxylon"albidiscum)、班帕登環(huán)紋炭團菌(A."bahnphadengense)和斯普格環(huán)紋炭團菌(A."spougei)。根據國內采集標本材料,文中提供了3個新記錄種形態(tài)特征描述和圖示。基于ITS序列采用最大似然法和貝葉斯法構建系統(tǒng)發(fā)育樹,3個種均得到分子數據的支持。此外,將每種都與模式標本的特征進行比較,并討論與相似物種的鑒別特征。研究標本現存放于中國熱帶農業(yè)科學院熱帶生物技術研究所大型真菌標本館(FCATAS)。

        環(huán)紋炭團菌屬(Annulohypoxylon),隸屬于子囊菌門(Ascomycota)、糞殼菌綱(Sor dariomycetes)、炭角菌目(Xylariales)、炭團菌科(Hypoxylaceae),模式種為截形環(huán)紋炭團菌[A."truncatum"(Starb?ck)"Y.M."Ju,"J.D."Rogers"amp;"H.M."Hsieh][1]。環(huán)紋炭團菌屬是HSIEH等[2]于2005年依據肌動蛋白(α-actin,ACT)和微管蛋白(β-Tubulin,"TUB)序列將炭團菌屬(Hypoxylon)的亞屬(Annulata)提升至屬的水平,與炭團菌屬區(qū)分開來。環(huán)紋炭團菌屬(Annulohypoxylon)與炭團菌屬(Hypoxylon)的主要區(qū)別是前者有炭質層圍繞子囊殼,孔口通常具有環(huán)形圓盤圍繞,高于子座表面,子囊孢子壁在芽縫一側有增厚區(qū)域[3-4]。這些特征也得到了化學分類特征的支持[5]。環(huán)紋炭團菌屬(Annulohypoxylon)的基物主要是腐爛的木頭和植物枯死的莖、葉,部分種類能夠引起植物病害[6-9]。很多種類分離出新的次生代謝產物和酶[10-11]。該屬在全球范圍均有分布,其中以熱帶和亞熱帶居多[12-16]。根據Index"Fungorum(https://"www.speciesfungorum.org/Names/Names.asp)的記載,截至2023年8月,有89個名稱記錄。目前全球該屬共有65個種,中國有24種[17-25]。

        2020—2023年,團隊對海南熱帶雨林國家公園進行大型真菌資源調查過程中,采集到3號環(huán)紋炭團菌屬標本,鑒定發(fā)現這3號標本分別代表了3個中國新記錄種,分別是白色環(huán)紋炭團菌(A."albidiscum"J.F."Zhang,"J.K."Liu,"K.D."Hyde"amp;"Z.Y."Liu)、班帕登環(huán)紋炭團菌(A."bahnphadengense"J."Fourn."amp;"M."Stadler)和斯普格環(huán)紋炭團菌(A."spougei"Suwannasai,"M.P."Martín,"Phosri"amp;"Whalley)。

        1""材料與方法

        1.1""材料

        研究標本采集自中國海南省熱帶雨林和亞熱帶常綠闊葉林,現存放于中國熱帶農業(yè)科學院熱帶生物技術研究所大型真菌標本館(FCATAS)。

        1.2""方法

        1.2.1""形態(tài)學鑒定""采用MA等[26]、SONG等[27-28]的研究方法進行分類鑒定。在野外使用佳能G15照相機拍攝真菌生境照片。使用超景深三維顯微系統(tǒng)(VHX-6000,基恩士,日本)對環(huán)紋炭團菌的宏觀特征包括子座表面、子囊殼的形狀及大小進行觀察測量。用手術刀切下一小塊標本滴入10%"KOH溶液,觀察染色反應。在進行顯微特征觀察時,將取好的組織切片樣品分別置于水、Melzer試劑和10%"KOH溶液浮載劑中,通過激光捕獲顯微分離系統(tǒng)(Cell"Cut"Plus,MMI,德國)觀察子囊和子囊孢子的顏色、形狀、大小,子囊頂環(huán)在梅爾澤試劑中是否變藍,以及芽縫是否存在等特征,并隨機測量至少30個子囊和子囊孢子。

        1.2.2""分子生物學鑒定""采用改良的CTAB法提取子實體的基因組DNA。取少量標本,使用植物DNA快提試劑盒(Magen),依據其操作說明書的步驟進行真菌DNA提取。采用通用引物ITS5/ITS4,PCR擴增內部轉錄間隔區(qū)(ITS)序列[29]。擴增體系50"μL:3"μL模板DNA,引物各1"μL,25"μL"2×Taq"Master"Mix(東盛生物科技有限公司),最后用ddH2O補齊。PCR程序:94"℃預變性4"min;94"℃變性40"s,55"℃退火50"s,72"℃延伸1"min,計35個循環(huán);72"℃延伸10"min,4"℃保存。PCR擴增產物送往華大基因(廣州)股份有限公司進行測序。隨后序列提交至GenBank,ITS測序結果通過NCBI網站(https://www.ncbi."nlm.nih.gov/)進行比對,分析序列的相似性,初步確定分類地位。

        選取同源性較高的序列(表1),利用在線分析網站mafft(MAFFT"alignment"and"NJ/"UPGMA"phylogeny)將序列比對后對齊。使用軟件trimAI對序列進行剪切,將剪切后的序列通過clustalx"1.83轉換為nxs格式。通過RAxML"GUI"1.3.1軟件,構建最大似然法(ML)系統(tǒng)發(fā)育樹。采用GTRGAMMA模型,ML+rapid"bootstrap程序,運行1000次重復,以評估各分支的可信度。通過MrBayes"v.3.2.6軟件,構建貝葉斯(BI)系統(tǒng)發(fā)育樹。采用invgamma模型,運行100萬代,每100代采樣1次。使用FigTree"v1.4.3軟件進行系統(tǒng)發(fā)育樹美化處理。選取Biscogniauxia"petrensis"Z.F."Zhang,"F."Liu"amp;"L."Cai作為外類群構建系統(tǒng)發(fā)育樹,BSgt;70%(左)和PPgt;0.8(右)的值在系統(tǒng)樹中標注,自測序列均用粗體標出。

        2""結果與分析

        2.1""白色環(huán)紋炭團菌(Annulohypoxylon"albidiscum"J.F."Zhang,"J.K."Liu,"K.D."Hyde"amp;"Z.Y."Liu)(圖1)

        形態(tài)特征:子座墊狀,厚0.6"mm,子囊殼突起明顯,表面黑色,初期伴有褐色絨毛層,后期黑色發(fā)亮;子座表層下顆粒黑色,炭質;在KOH溶液中組織顏色變?yōu)殚蠙炀G色;子囊殼下層組織不明顯,褐色,厚0.1"mm。子囊殼近球形至倒卵球形,直徑0.31~0.41"mm,孔口乳突狀,被truncatum型圓盤環(huán)繞,圓盤直徑0.14~0.24"mm。子囊圓柱形,單囊壁,內含8個子囊孢子,單列排列,全長68~"108"μm,有孢子部分長50~76"μm,寬4.1~5.6"μm,柄長12~39"μm;頂環(huán)在Melzer試劑中呈藍色,盤狀,高0.8~1.0"μm,寬1.7~1.8"μm。子囊孢子褐色至深褐色,單胞,不等邊橢圓形,兩端窄圓,7.4~8.9"μm×3.2~4.2"μm(=8.2"μm×3.8"μm,n="57),芽縫直,與孢子等長;在10%"KOH溶液中孢子壁開裂,光滑;孢子周壁光滑。

        討論:該種的模式標本采集于泰國,除中國標本較泰國標本子囊殼直徑略小,白色環(huán)紋褪去,其他特征均十分吻合[35]。基于ITS序列系統(tǒng)發(fā)育樹顯示,中國標本FCATAS"2766(GenBank"No."ON740669)與泰國標本MFLUCC"15-0645(Gen Bank"No."KU852741)聚為一支(圖2),BS=99%,PP=1.0,可以與形態(tài)特征相互印證,從而支持將其鑒定為A."albidiscum。此種與永加斯環(huán)紋炭團菌(Annulohypoxylon"yungensis"Sir,"Kuhnert,"Hladki"amp;"A.I."Romero)是近似種,其主要區(qū)別為后者子囊孢子較大,為(8.0~9.2)μm×(3.6~4.3)μm[5]。

        2.2""班帕登環(huán)紋炭團菌(Annulohypoxylon"bahnphadengense"J."Fourn."amp;"M."Stadler)(圖3)

        形態(tài)特征:子座墊狀,厚1.4"mm,子囊殼突起明顯,表面黑色,最外層被褐色覆蓋,后期脫落;子座表層下顆粒黑色,炭質;在KOH溶液中組織顏色變?yōu)闇\褐黑色;子囊殼下層組織明顯,黑褐色。子囊殼倒卵球形,高0.41~0.61"mm,寬0.38~"0.59"mm,孔口乳突狀,被truncatum型圓盤環(huán)繞,圓盤直徑0.24~0.31"mm。子囊圓柱形,單囊壁,內含8個子囊孢子,單列排列,全長84~122"μm,有孢子部分長53~76"μm,寬3.7~5.6"μm,柄長19~56"μm;頂環(huán)在Melzer試劑中呈藍色,盤狀,高0.5~0.7"μm,寬0.9~1.5"μm。子囊孢子褐色,單胞,不等邊橢圓形,兩端窄圓至闊圓,(6.7~"8.2)μm×(2.8~3.6)μm(=7.5"μm×3.2"μm,"n=51),芽縫直,與孢子等長;在10%"KOH溶液中孢子壁開裂,光滑;孢子周壁光滑。

        生境:生于熱帶雨林無皮枯木頭上。

        世界分布:中國、泰國[13]。

        討論:班帕登環(huán)紋炭團菌(A."bahnphadengense)的模式標本發(fā)現于泰國清邁府班帕登。中國尖峰嶺標本在形態(tài)上與該種模式標本的描述基本一致[13]。基于ITS序列系統(tǒng)發(fā)育樹分析,中國標本FCATAS"2762(GenBank"No."OR416117)與泰國標本STMA13115(GenBank"No."KX376338)聚為一支(圖2),BS=100%,PP=1.0,可以與形態(tài)特征相互印證,從而支持將其鑒定為A.bahn phadengense。此種與努里格環(huán)紋炭團菌(Annu lohypoxylon"nouraguense"J."Fourn."amp;"Lechat)是在形態(tài)上較近似,其主要區(qū)別是后者KOH染色為紫色[36]。此種與近亮環(huán)紋炭團菌(Annulohy poxylon"subnitens"J."Fourn."amp;"Lechat)是在宏觀形態(tài)上也較相似,其主要區(qū)別是后者子囊殼大,直徑0.6~0.7"mm[36]。

        2.3""斯普格環(huán)紋炭團菌(Annulohypoxylon"spougei"Suwannasai,"M.P."Martín,"Phosri"amp;"Whalley)(圖4)

        形態(tài)特征:子座墊狀至半球狀,通常匯合,長0.3~1.5"cm,寬0.3~0.9"cm,子囊殼突起明顯,表面黑色;子座表層下顆粒黑色,炭質;在KOH溶液中組織顏色變?yōu)殚蠙炀G色。子囊殼球形至倒卵球形,高0.48~0.73"mm,寬0.44~0.59"mm,孔口乳突狀,被bovei型圓盤環(huán)繞,圓盤直徑0.30~0.43"mm。子囊圓柱形,單囊壁,內含8個子囊孢子,單列排列,全長77~106"μm,有孢子部分長59~77"μm,寬4.8~6.6"μm,柄長15~34"μm。子囊孢子褐色至深褐色,單胞,不等邊橢圓形,(7.5~9.7)μm×(3.1~5.0)μm(=8.4"μm×4.0"μm,n="68),芽縫直,與孢子等長;在10%"KOH溶液中孢子壁開裂,光滑;孢子周壁光滑。

        生境:生于亞熱帶常綠闊葉林枯木樹皮上。

        研究標本:中國海南省??谑旋埲A區(qū)金牛嶺公園,海拔8"m,2022年10月3日,馬海霞,FCATAS"5413。

        世界分布:中國、泰國[33]。

        討論:斯普格環(huán)紋炭團菌(A."spougei)的模式標本發(fā)現于泰國,中國海南的標本在形態(tài)特征上與該種的原始描述一致[33]?;贗TS序列系統(tǒng)發(fā)育樹分析顯示,中國標本FCATAS"5413(Gen Bank"No."OR417347)與泰國標本SWUFH099(GenBank"No."FN252419)聚為一支(圖2),BS=100%,PP=1.0,可以與形態(tài)特征相互印證,從而支持將其鑒定為A."spougei。

        3""討論

        海南省地處我國最南端,擁有我國分布最集中、類型最多樣、保存最完好、連片面積最大的大陸性島嶼型熱帶雨林,獨特的氣候和地形為真菌的生長提供了極其適宜的條件。炭團菌是子囊菌中的重要類群,可寄生在植物上,引起林木病害,有些種類能夠產生多種次級代謝產物,具有重要的生態(tài)價值和經濟價值。在我國相繼發(fā)現了大量的新分類單元,特別是對熱帶和亞熱帶不同區(qū)域有較多報道[26-28,"37-39],但對于環(huán)紋炭團菌屬的研究相對較少[17-25]。本研究所報道的環(huán)紋炭團菌3個新記錄種,其中有2種采自海南熱帶雨林,1種采自亞熱帶常綠闊葉林,是對中國大型子囊菌的補充,其新分布的發(fā)現對于炭團菌科真菌物種起源、區(qū)系研究具有重要意義。同時,也說明我國熱帶和亞熱帶地區(qū)大型真菌多樣性極其豐富,隨著調查的深入,將會有更多新的分類單元被發(fā)現。
