顯柱南蛇藤(Celastrus stylosusWall)系衛(wèi)矛科(Celastraceae)南蛇藤屬(Celastrus)植物,該屬植物包含50 多個種,分布于世界各地,常見的有燈油藤、獨子藤、苦皮藤、顯柱南蛇藤等,中國約有30 種分布于各地[1]. 目前研究發(fā)現(xiàn),南蛇藤屬植物萜類成分較為豐富,從其屬植物的果實和根皮中共發(fā)現(xiàn)倍半萜類70 個,三萜類67 個和二萜類化合物2 個[2],其中倍半萜主要以β-二氫沉香呋喃型為主,三萜類化合物大多為五環(huán)三萜,如木栓烷型、齊墩果烷型、羽扇豆烷型等. 南蛇藤屬植物的葉子中還含有大豆素、山奈酚、槲皮素等常見黃酮類化合物[3]. 此外,南蛇藤屬植物還含有一些常見的脂肪酸、酚酸類化合物,如棕櫚酸、肉桂酸等[4]. 現(xiàn)代藥理學(xué)研究發(fā)現(xiàn),該屬植物具有良好的抗腫瘤、降壓、抗炎、抗氧化、抗菌、抗病毒[5]等藥理活性. 為進(jìn)一步開發(fā)和利用顯柱南蛇藤,闡明其藥效發(fā)揮的物質(zhì)基礎(chǔ),本實驗對顯柱南蛇藤根皮85%乙醇提取物氯仿部位進(jìn)行化學(xué)成分研究,并采用多種分離技術(shù)和波譜學(xué)鑒定方法,從其根皮氯仿部位分離鑒定15 個化合物(如圖1),所有化合物均為首次從該種植物中分離得到.

核磁共振儀(Bruker AV-400 型,德國Bruker 公司),高效液相色譜-質(zhì)譜聯(lián)用(Agilent 公司),中壓制備液相色譜 (蘇州匯通),柱層析硅膠(0.054~0.077 mm,200~300 目,青島海洋化工廠),制備薄層板(煙臺江友硅膠開發(fā)有限公司),Sephadex LH-20葡聚糖凝膠(吉泰科技生物),ODS 柱色譜填料(C18, 15 μm, Merck 公司).
試劑:石油醚(60~90 ℃),分析純;乙酸乙酯,分析純;氯仿,分析純;甲醇,色譜純;乙醇,分析純.以上試劑均為國藥集團(tuán)化學(xué)試劑有限公司生產(chǎn).
顯柱南蛇藤根皮采收于湖北省恩施市,由湖北民族大學(xué)王發(fā)松教授鑒定為顯柱南蛇藤(Celastrus stylosusWall).
顯柱南蛇藤干燥根皮15.0 kg 粉碎過篩,85%乙醇60 ℃(50 L × 3)超聲提取,合并濃縮得1.1 kg總浸膏. 總浸膏用60 ℃水(10 L)分散制成混懸液,分別用石油醚、氯仿和正丁醇萃取. 合并氯仿萃取液,濃縮得氯仿部位浸膏160 g. 氯仿部位濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯梯度洗脫. 經(jīng)TLC 檢識得4 個組分Fr.1~ Fr.4.
Fr.1(20 g)濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯(體積比50∶1→10∶1)梯度洗脫,TLC檢識合并得3 個亞組分Fr.1-1~ Fr.1-3. 其中,F(xiàn)r.1-1、Fr.1-3 靜置有晶體析出,反復(fù)重結(jié)晶得化合物1(11 mg),化合物2 (16 mg). Fr.1-2 經(jīng) Sephadex LH-20 凝膠柱層析(甲醇-氯仿,體積比1∶1)分離,TLC 檢識合并3 個亞組分Fr.1-2-1~Fr.1-2-3. Fr.1-2-2靜置有晶體析出,反復(fù)重結(jié)晶得化合物3 (15 mg).Fr.1-2-3 經(jīng)制備薄層板分離,得化合物4(10 mg).
Fr.2(23 g)組分濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯(體積比40∶1→1∶1)梯度洗脫,TLC 檢識合并得3 個亞組分Fr.2-1~ Fr.2-3. 其中,F(xiàn)r.2-1、Fr.2-2 靜置有晶體析出,反復(fù)重結(jié)晶得化合物5(14 mg),化合物6(17 mg). Fr.2-3 經(jīng)Sephadex LH-20 凝膠柱層析(甲醇-氯仿,體積比1∶1)分離純化,得化合物7(13 mg).
Fr.3(13 g)組分濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯(體積比30∶1→0∶1)梯度洗脫,TLC 檢識合并得4 個亞組分Fr.3-1~Fr.3-4. Fr.3-1濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯(體積比30∶1)等度洗脫,得化合物8 (33 mg). Fr.3-2濕法上樣,減壓硅膠柱層析,石油醚-乙酸乙酯(體積比25∶1)等度洗脫,得化合物9 (26 mg). Fr.3-3靜置有晶體析出,反復(fù)重結(jié)晶得化合物10 (14 mg).Fr.3-4 經(jīng)Sephadex LH-20 凝膠柱層析(甲醇-氯仿,體積比1∶1)分離純化,得化合物11 (13 mg).
Fr.4(8 g)組分經(jīng)中壓制備色譜(甲醇-水,體積比85∶15→65∶35)梯度洗脫,得4 個亞組分Fr.4-1~Fr.4-4. Fr.4-1 進(jìn)行中壓制備色譜等度洗脫(甲醇-水,體積比80∶20),得化合物12 (9 mg).Fr. 4-2 進(jìn)行中壓制備色譜等度洗脫(乙腈-水,體積比80∶20),制備薄層板分離,得化合物13 (17 mg).Fr.4-3 進(jìn)行Sephadex LH-20 凝膠(甲醇-氯仿,體積比1∶1)柱色譜分離,反復(fù)重結(jié)晶,得化合物14 (14 mg).Fr.4-4 進(jìn)行中壓制備色譜(甲醇-水,體積比3∶1)梯度洗脫,經(jīng)Sephadex LH-20 凝膠(甲醇-氯仿,體積比1∶1)柱色譜分離,反復(fù)重結(jié)晶,得化合物14(17 mg)、15 (12 mg).

查詢國內(nèi)外文獻(xiàn),發(fā)現(xiàn)目前只有對顯柱南蛇藤果實石油醚部位化學(xué)成分的研究報道[2]. 顯柱南蛇藤作為傳統(tǒng)中藥,其根皮化學(xué)成分未詳細(xì)研究. 本研究從顯柱南蛇藤根皮氯仿部位分離鑒定15 個化合物,包括2 個有機(jī)酸(1, 2)、2 個甾醇(3, 4)、1 個倍半萜(5),10 個三萜(6~15). 對比發(fā)現(xiàn),根皮與果實化學(xué)成分有顯著差異,果實部位主要以倍半萜類化合物為主,根皮部位主要以三萜類化合物為主. 南蛇藤屬植物的活性報道大多集中在抗腫瘤抗菌等方面,李嘉琪[21]等研究發(fā)現(xiàn)南蛇藤素(化合物5)能呈濃度依賴性抑制乳腺癌細(xì)胞的增殖、遷移,促進(jìn)細(xì)胞凋亡. 姚正盛[22]研究發(fā)現(xiàn)扁蒴藤素(化合物6)對鼻咽癌細(xì)胞具有增殖抑制及誘導(dǎo)凋亡的作用. 周永林[23]研究發(fā)現(xiàn)齊墩果酸(化合物14)可顯著降低由細(xì)菌性溶血素對機(jī)體造成的損傷和顯著恢復(fù)β-內(nèi)酰胺類抗生素的體內(nèi)外抗菌活性.Vonshak 等[24]發(fā)現(xiàn)南蛇藤提取物對紅色毛癬菌具有抑制作用. 丁麗娜[25]研究發(fā)現(xiàn),短柄南蛇藤果殼、假種皮、種子的部分提取物對大腸埃希菌有抑制作用. 南蛇藤屬植物化學(xué)成分豐富,但活性研究大都只停留在初步的活性篩選中,特別是抗菌的作用機(jī)制還需要更加深入研究. 本研究所得化合物可用于進(jìn)一步相關(guān)活性篩選,以期進(jìn)一步闡明顯柱南蛇藤根皮的藥效物質(zhì)基礎(chǔ).
