        核桃青皮正丁醇部位化學(xué)成分的研究

        核桃青皮為胡桃科胡桃屬植物胡桃楸JuglansmandshuricaMaxim.的未成熟青果皮,別名青胡桃皮、青龍衣,主要分布在我國(guó)的東北地區(qū),在內(nèi)蒙古、河北、山東等地區(qū)也有少許分布,資源豐富,為我國(guó)民間常用中藥,其味苦、澀,性平,具有清熱解毒、止痢、療癬等功效,可用于治療皮膚瘙癢、發(fā)熱咳嗽、水痢等[1-2]。目前,從核桃青皮中主要分離得到三萜類、黃酮類、萘醌類等成分。現(xiàn)代藥理研究表明核桃青皮具有抗腫瘤、抗氧化、抗菌等作用[3]。近年來(lái)胡桃屬的藥用價(jià)值引起了人們的關(guān)注,為了進(jìn)一步研究其藥效物質(zhì)基礎(chǔ),本實(shí)驗(yàn)對(duì)其化學(xué)成分進(jìn)行系統(tǒng)研究,共分離得到14個(gè)化合物,其中3個(gè)黃酮類、7個(gè)黃酮苷類、4個(gè)萘苷類。

        1 材料

        Bruker-400型超導(dǎo)核磁共振光譜儀(瑞士Bruker公司);Finnigan LCQ質(zhì)譜儀;Heidolph-Laborota 4000型旋轉(zhuǎn)蒸發(fā)儀(德國(guó)海道夫公司);柱層析硅膠(青島海洋化工廠);柱色譜ODS (日本YMC公司);其他試劑均為分析純。

        藥材于2019年7月采自吉林省長(zhǎng)白山地區(qū)。經(jīng)黑龍江中醫(yī)藥大學(xué)藥學(xué)院楊炳友教授鑒定為胡桃科植物核桃楸JulansmandshuricaMaxim未成熟外果皮,標(biāo)本(20190813)保存于黑龍江中醫(yī)藥大學(xué)中藥化學(xué)教研室。

        2 提取與分離

        取干燥的核桃青皮粉末10.0 kg,用甲醇浸泡1 d,回流提取3次,減壓濃縮得到提取物。以適量蒸餾水添加并分散所得提取物,依次用二氯甲烷、乙酸乙酯及水飽和正丁醇進(jìn)行多次萃取,萃取液經(jīng)減壓濃縮得各部位浸膏。

        取正丁醇浸膏(208 g),經(jīng)硅膠柱,以二氯甲烷-甲醇系統(tǒng)梯度洗脫,得到8個(gè)組分Fr.1~Fr.8。Fr.1經(jīng)硅膠柱,以二氯甲烷-甲醇(15∶1~0∶1)梯度洗脫,得化合物12(6.8 mg)、13(5.4 mg)、14(7.2 mg);Fr.6經(jīng)硅膠柱以二氯甲烷-甲醇(15∶1~0∶1)梯度洗脫后,得7個(gè)組分Fr.6.1~Fr.6.7,F(xiàn)r.6.4經(jīng)硅膠柱以二氯甲烷-甲醇(20∶1~0∶1)系統(tǒng)梯度洗脫后,得到化合物2(3.6 mg)、5(6.3 mg)、7(4.7 mg);Fr.6.6經(jīng)硅膠柱以二氯甲烷-甲醇(10∶1~0∶1)系統(tǒng)梯度洗脫后,得化合物4(5.8 mg)、6(5.6 mg)、8(7.2 mg)、10(5.7 mg);Fr.6.7經(jīng)硅膠柱以二氯甲烷-甲醇(15∶1~0∶1)系統(tǒng)梯度洗脫后,得到化合物1(4.2 mg)、11(4.8 mg);Fr.8經(jīng)硅膠柱色譜二氯甲烷-甲醇(10∶1~0∶1)與ODS硅膠柱甲醇-水(40∶60~100∶0)反復(fù)梯度洗脫得化合物3(3.3 mg)、9(5.1 mg)。

        3 結(jié)構(gòu)鑒定

        4 討論

        核桃青皮是我國(guó)傳統(tǒng)中藥材,有著悠久的藥用歷史,近年來(lái),隨著現(xiàn)代科學(xué)技術(shù)的發(fā)展,對(duì)于核桃青皮化學(xué)成分的研究日益深入,但是就目前而言,對(duì)其正丁醇部位的研究報(bào)道較少。本實(shí)驗(yàn)通過(guò)現(xiàn)代提取分離技術(shù),對(duì)核桃青皮正丁醇部位進(jìn)行系統(tǒng)研究,共分離得到14個(gè)化合物,包括萘醌苷類、黃酮及其苷類,其中化合物1~5、7~11、13為首次從該植物中分離得到,化合物5、9為首次從胡桃屬中分離得到。本研究在一定程度上豐富了核桃青皮的化學(xué)成分庫(kù),完善了核桃青皮正丁醇部位化學(xué)成分的研究,以期為核桃青皮正丁醇部位的藥理活性研究提供物質(zhì)基礎(chǔ),對(duì)今后核桃青皮的合理開發(fā)與利用具有指導(dǎo)意義。
