        PAB—Gal4—DBD—HA—Twist重組質(zhì)粒的制備

        【摘要】 目的:構(gòu)建人Twist基因的表達(dá)載體pAB-Gal4-DBD-HA-Twist,為研究其轉(zhuǎn)錄調(diào)控機(jī)制提供實(shí)驗(yàn)工具。方法:重組克隆質(zhì)粒pcDNA-Flag-Twist用含有特異BamH Ⅰ和Hind Ⅲ酶切位點(diǎn)的引物進(jìn)行聚合酶鏈反應(yīng);對(duì)含有兩種酶切位點(diǎn)的目的Twist基因片段和pAB-Gal4-DBD-HA載體進(jìn)行酶切、分離、回收純化、連接后轉(zhuǎn)入感受態(tài)DH5α中,細(xì)菌培養(yǎng),菌落聚合酶鏈反應(yīng)、測(cè)序鑒定。結(jié)果:鑒定結(jié)果顯示,Twist基因正確克隆到PAB-Gal4-DBD-HA表達(dá)載體,并且轉(zhuǎn)入了大腸桿菌中。結(jié)論:成功構(gòu)建了人twist基因PAB-Gal4-DBD-HA-Twist表達(dá)載體,為進(jìn)一步研究其表達(dá)、調(diào)控、作用機(jī)制奠定基礎(chǔ)。

        【Abstract】 Objective:To construct PAB-Gal4-DBD-HA-twist recombinant expression plasmid,providing experimental tools to study the transcription regulation.Method:The recombinant plasmid pcDNA-Flag-Twist was detected by polymerase chain reaction (PCR) using primers containing specific BamHⅠ and Hind Ⅲ restriction enzyme site.For the purpose of the Twist gene and pAB-Gal4-DBD-HA vector containing two kinds of enzyme cutting sites were studied by enzyme digestion,separation,recovery and purification,connected and transformed into competent DH5 alpha,they were detected by bacterial culture,colony polymerase chain reaction and sequencing identification.Result:The identification results showed that the twist gene recombinant plasmid was successfully constructed and transformed in E.coli.Conclusion:Full-length twist is subcloned into pAB-Gal4-DBD-HA vector successfully,and lay the foundation to further study its expression,regulation and mechanism.

        TWIST蛋白是一種新近發(fā)現(xiàn)的轉(zhuǎn)錄因子,在胚胎發(fā)育中起關(guān)鍵調(diào)控作用[1]。隨后的研究發(fā)現(xiàn),其能與靶基因上的特異DNA序列結(jié)合,廣泛參與調(diào)控器官生長(zhǎng)發(fā)育、細(xì)胞的凋亡、腫瘤發(fā)生、細(xì)胞增殖分化等過程[2]。Twist基因是一個(gè)候選癌基因,能通過干擾p53相關(guān)的通路、抑制p21wafl、干擾NF-KB通路、直接抑制p65,干擾NF-KB的轉(zhuǎn)錄激活作用等多種途徑抑制細(xì)胞凋亡[3-8]。Sosic等[7]研究發(fā)現(xiàn),在生理情況下,TWIST蛋白不但在胚胎發(fā)育上扮演重要的角色,而且可以通過活化NF-kB途徑抑制細(xì)胞因刺激而產(chǎn)生更多的細(xì)胞因子而造成對(duì)細(xì)胞的損傷[1]。同時(shí),此基因的高表達(dá)可以使E-鈣黏蛋白(E-cadherin)表達(dá)缺失,促進(jìn)間質(zhì)-上皮轉(zhuǎn)變(MET)而使惡性腫瘤獲得或浸潤(rùn)、轉(zhuǎn)移力增強(qiáng)[2]。由此可見,對(duì)于twist基因轉(zhuǎn)錄調(diào)控作用的研究是揭示其眾多生理、病理功能的重要基礎(chǔ)。目前,研究基因調(diào)控功能的方法之一就是構(gòu)建待研究基因的表達(dá)載體。利用此研究思路,筆者嘗試構(gòu)建含有twist基因與Gal4-DNA結(jié)合區(qū)序列的PAB-Gal4-DBD-HA-Twist表達(dá)載體,來研究此基因表達(dá)調(diào)控機(jī)制。

        1 材料與方法

        1.1 材料 pcDNA-Flag-twist和pAB-Gal4-DBD-HA由美國(guó)貝勒醫(yī)學(xué)院徐建明博士贈(zèng)予;DH5α由瀘州醫(yī)學(xué)院醫(yī)學(xué)分子生物學(xué)實(shí)驗(yàn)室保存。BamHⅠ和HindⅢ、DNA mark(DL2000、100 bp DNA Ladder)、TaKaRa LA Taq with GC Buffer PCR試劑盒、質(zhì)粒小量提取試劑盒、凝膠回收試劑盒購自大連寶生物公司;T4 DNA連接酶購自普洛麥格(北京)生物技術(shù)公司;溴化乙錠(EB)購自Sigma公司,酵母提取物、胰蛋白胨、瓊脂糖購自上海生物工程有限公司。

        1.2 方法

        1.2.1 目的基因擴(kuò)增

        1.2.1.1 帶有酶切位點(diǎn)BamHⅠ和Hind Ⅲ的TWIST基因片段的獲得 (1)擴(kuò)增模版:pcDNA-Flag-Twist質(zhì)粒。(2)引物設(shè)計(jì):上游引物P1:5-GTTGGATCCAATGATGCAGGACGTGTCC-3;下游引物P2:5-TTAAAGCTTGTGGGACGCGGACATGGA-3。P1中GTT為保護(hù)堿基,GGATCC為BamHⅠ酶切位點(diǎn),P2中TTA為保護(hù)堿基,AAGCTT為Hind Ⅲ酶切位點(diǎn)。擴(kuò)增片段長(zhǎng)度為650 bp(不含引入的酶切位點(diǎn)和保護(hù)堿基)。引物經(jīng)BLAST分析,與Genebank數(shù)據(jù)庫中其他基因沒顯著同源性,最后送上海生物工程有限公司合成。endprint

        1.2.1.2 PCR擴(kuò)增 反應(yīng)體系:2×GCBufferⅠ 25 μL,dNTP mixture 8 μL,引物P1和P2各1 μL,TaKaRa LA Taq 0.5 μL,ddH2O 13.5 μL。在PCR擴(kuò)增儀上進(jìn)行反應(yīng):94 ℃條件下進(jìn)行預(yù)變性 1 min,然后按94 ℃ 30 s,64 ℃ 30 s,72 ℃ 45 s,反應(yīng)30個(gè)循環(huán)后,72 ℃延伸10 min,4 ℃保存。1%的瓊脂糖凝膠電泳檢測(cè)。凝膠回收目的條帶。

        1.2.2 重組表達(dá)載體的構(gòu)建 PCR產(chǎn)物純化后與質(zhì)粒pAB-Gal4-DBD-HA用BamH Ⅰ和Hind Ⅲ于37 ℃ 2 h分別進(jìn)行雙酶切,酶切產(chǎn)物經(jīng)凝膠電泳后回收純化,按照T4 DNA連接系統(tǒng)說明進(jìn)行連接反應(yīng)。反應(yīng)體系如下:10×biolabs buffer 2 μL,Twist基因0.5μL,線性pAB-Gal4-DBD-HA 0.7 μL,T4連接酶0.5 μL,ddH2O 16.3 μL,總體系20 μL,將上述反應(yīng)體系室溫放置15 min。將連接產(chǎn)物通過熱休克法轉(zhuǎn)化至感受態(tài)DH5α,37 ℃150 rpm搖床復(fù)蘇培養(yǎng)45 min。取菌液均勻涂布于含終濃度為50 μg/mL的氨芐青霉素LB瓊脂板上,37 ℃恒溫箱中倒置培養(yǎng)過夜。

        1.2.3 重組體的篩選和鑒定

        1.2.3.1 菌落PCR篩選重組體 隨機(jī)挑取數(shù)個(gè)菌落,做好標(biāo)記,菌落聚合酶鏈法篩選重組體,按上述體系和條件反應(yīng);根據(jù)結(jié)果,挑取陽性克隆的另半個(gè)菌落至LB培養(yǎng)液中,37 ℃培養(yǎng)過夜。

        1.2.3.2 質(zhì)粒的提取酶切鑒定 按照質(zhì)粒的提取試劑盒提取質(zhì)粒,進(jìn)行雙酶切鑒定。Twist基因通過BamHⅠ和HindⅢ酶切位點(diǎn)進(jìn)入載體pAB-Gal4-DBD-HA,雙酶切后將會(huì)有約4500 bp、650 bp兩個(gè)大、小片段。酶切體系如下:10×NE buffer 1μL,10×BSA 1μL,pAB-Gal4-DBD-HA-Twist 3μL,BamH Ⅰ 0.25 ?L,Hind Ⅲ 0.25 μL,雙蒸水 4.5μL??偡磻?yīng)體系為10 ?L,37 ℃培養(yǎng)孵育1 h。

        1.2.3.3 測(cè)序鑒定 初步鑒定陽性的克隆質(zhì)粒送上海生工進(jìn)行序列測(cè)定。

        2 結(jié)果

        2.1 pcDNA-Flag-Twist質(zhì)粒PCR擴(kuò)增結(jié)果 以pcDNA-Flag-Twist為模板,用特異性引物經(jīng)聚合酶鏈擴(kuò)增后,電泳檢測(cè)為大小約650 bp的片段,與目的片段大小相符(圖1)。

        2.2 菌落PCR法鑒定pAB-Gal4-DBD-HA-Twist結(jié)果 pAB-Gal4-DBD-HA-Twist連接產(chǎn)物轉(zhuǎn)化至DH5α,菌落PCR法擴(kuò)增表達(dá)質(zhì)粒,經(jīng)凝膠電泳之后,結(jié)果與預(yù)期結(jié)果一致(圖2)。

        2.3 重組表達(dá)載體的雙酶切鑒定 構(gòu)建的pAB-Gal4-DBD-HA-Twist用BamH Ⅰ和Hind Ⅲ酶切后出現(xiàn)兩條片段,分別為pAB-Gal4-DBD-HA載體和twist片段,可初步鑒定此克隆為陽性(圖3)。

        2.4 表達(dá)質(zhì)粒pAB-Gal4-DBD-HA-Twist測(cè)序結(jié)果與分析 初步鑒定為陽性克隆的pAB-Gal4-DBD-HA-Twist送上海生工公司測(cè)序,結(jié)果分析顯示,重組質(zhì)粒中的twist基因序列與基因庫中公布的已知序列(NM_000474)完全匹配(圖4)。表明目的基因序列按照預(yù)期方式定向插入表達(dá)載體,成功構(gòu)建了重組表達(dá)載體。

        3 討論

        Twist基因首先在果蠅中被發(fā)現(xiàn),隨后相繼在其他脊椎動(dòng)物中發(fā)現(xiàn),脊椎動(dòng)物的twist基因具有84%~100%的同源性[9-10]。其在幼稚中胚層細(xì)胞和中胚層起源器官中表達(dá)最多,而在出生后降至低水平,廣泛參與眾多生理、病理過程,它的信號(hào)通路是一個(gè)復(fù)雜、多途徑的網(wǎng)絡(luò)系統(tǒng),是多種與腫瘤發(fā)生發(fā)展相關(guān)途徑的中心環(huán)節(jié)[1,7,11]。Twist基因的轉(zhuǎn)錄水平與腫瘤的惡性程度、臨床病理分期分級(jí)和轉(zhuǎn)移能力有關(guān),Twist基因促進(jìn)了EMT的進(jìn)程,增加了惡性腫瘤的轉(zhuǎn)移與侵襲力,其上調(diào)表達(dá)與紫杉醇和/或長(zhǎng)春新堿的耐藥性有關(guān)[2,11-13]。

        在本研究中,筆者采用HA作為目的基因的連接蛋白,其不會(huì)干擾標(biāo)記蛋白的正常功能,而且可以任意加到目標(biāo)蛋白的C-或N-末端。含有twist與Gal4-DNA結(jié)合區(qū)序列融合形成嵌合基因的重組表達(dá)質(zhì)粒構(gòu)建路線如圖5所示。該質(zhì)??梢栽诩?xì)胞內(nèi)表達(dá)出TWIST-Gal4-DBD融合蛋白,利用該蛋白的Gal4-DBD可以與報(bào)告基因的啟動(dòng)子結(jié)合的特性,使TWIST蛋白能夠結(jié)合于報(bào)告基因的附近,對(duì)下游靶基因發(fā)揮轉(zhuǎn)錄調(diào)控作用。pAB-Gal4-DBD-HA具有很強(qiáng)的復(fù)制能力,可以滿足宿主細(xì)胞分裂時(shí)跟隨胞漿穩(wěn)定的遺傳給子代細(xì)胞,保證了目的基因的表達(dá),此載體還含有高效SV40增強(qiáng)子,能順式調(diào)控基因轉(zhuǎn)錄活性。

        因此,該重組質(zhì)粒的成功構(gòu)建可以用于在活細(xì)胞自然狀態(tài)下觀察Twist的基因表達(dá)功能,為后續(xù)的其功能和臨床的研究奠定了實(shí)驗(yàn)基礎(chǔ)。
