珠子草(PhyllanthusniriruL.) 為大戟科葉下珠屬植物,是我國和印度的一種常用傳統(tǒng)民間草藥[1]。珠子草傣語稱為芽害巴,入風、水塔,具有清熱解毒、利濕通淋的功能,臨床用于黃疸肋痛、瀉利水腫、小便不利的治療,是傣藥國藥準字號品種珠子肝泰膠囊的主要組方藥材[2]。近年來,國內(nèi)外學者對珠子草的化學成分和藥理活性開展了大量研究[3]。根據(jù)文獻報道,珠子草的化學成分包括生物堿、花色苷、綠原酸、香豆素、黃酮、木脂素、酚酸、單寧、三萜、皂苷等多種類型;現(xiàn)代藥理學研究顯示,珠子草具有抗乙肝病毒、降血糖、排泌尿系統(tǒng)結(jié)石、抗腫瘤、肝保護、降血脂、抑菌、止痛、抗?jié)兒臀副Wo作用、抗瘧疾、腎保護作用等多種活性。
傣醫(yī)將珠子草打粉或提取,全株入藥。結(jié)合臨床應用,為了尋找珠子草抗乙肝病毒的活性成分,本文根據(jù)傣醫(yī)的臨床使用方法,對珠子草提取物的各個極性部位進行了系統(tǒng)的化學成分研究,共分離鑒定了14個單體化合物,根據(jù)波譜數(shù)據(jù)和質(zhì)譜數(shù)據(jù),查閱文獻,鑒定了他們的結(jié)構(gòu)(圖1)。并根據(jù)不同類型化合物的文獻報道,推測珠子草抗乙肝病毒活性可能與木脂素類成分和多酚類成分有關。

Bruker 600M AVACE Ⅲ核磁共振儀(TMS 為內(nèi)標)(德國Bruker公司);Agilent 1260-6120LC/MS (美國Agilent科技公司);R210型旋轉(zhuǎn)蒸發(fā)儀(瑞士Buchi公司);C18反相硅膠(YMC ODS-A,50μm)(日本YMC公司);正相硅膠板(GF254)(青島海洋化工廠);反相硅膠板(TLC Silica Gel RP-18 F254s)(德國Merk公司);氘代試劑(Sigma公司);Waters Prep150 型高效液相色譜儀(檢測器2489UV / Visible Detector,沃特世科技有限公司);制備色譜柱YMC ODS-A柱(250mm×10mm,5μm,YMC科技有限公司);柱色譜硅膠(100~200目,200~300 目,青島海洋化工有限公司),薄層色譜硅膠(GF-254,青島海洋化工有限公司); Sephadex LH-20(北京慧德義科技發(fā)展有限公司)。
珠子草藥材采自西雙版納版納藥業(yè)有限公司栽培基地,經(jīng)公司王劍總經(jīng)理鑒定為珠子草PhyllanthusniruriL.(芽害巴)。
藥材自然干燥,切2~3cm小段,取5.0kg,放置適當容器中,加8倍量95%乙醇室溫冷浸提取3次,每次24h,合并提取液并減壓濃縮,得到浸膏約0.8kg。浸膏加適量水懸浮,分別用石油醚、乙酸乙酯和正丁醇萃取,每次萃取至顏色較淡。合并各萃取部分,減壓濃縮至浸膏,得到石油醚部位(101g)、乙酸乙酯部位(285g)、正丁醇部位(320g)和水部位。
石油醚部位采用硅膠柱色譜分離,石油醚-乙酸乙酯梯度洗脫(10∶0~1∶1),收集流分,每份500mL,薄層色譜檢測合并主點相同部位,共得到10個組分(Fr11~10);乙酸乙酯部位用硅膠柱色譜進行粗分,氯仿-甲醇梯度洗脫(10∶0~1∶1),收集流分,每份500mL,薄層色譜檢測合并主點相同部位,得到8個組分(Fr21~8);正丁醇部位用硅膠柱色譜進行粗分,二氯甲烷-甲醇梯度洗脫(10∶0~1∶1),收集流分,每份500mL,薄層色譜檢測合并主點相同部位,共10個組分(Fr31~10)。選取提取物組分中主點明確的部分,采用Sephadex LH-20凝膠色譜柱(甲醇洗脫)、ODS柱色譜和制備液相色譜(甲醇-水系統(tǒng)),結(jié)合重結(jié)晶等方法,從石油醚部位分離得到化合物β-谷甾醇(1)、木栓酮(2)、珠子草脂素(3)、珠子草次素(4)、珠子草素(5),乙酸乙酯部位分離得到化合物β-胡蘿卜苷(6)、羽扇豆醇(7)、蘆丁(8)、山奈酚-3-O-蕓香糖苷(9)、短葉蘇木酚(10);正丁醇部位分離得到化合物沒食子酸(11)、鞣花酸(12)、柯里拉京(13)、老鸛草素(14)等4個多酚類成分。
化合物(1): 白色結(jié)晶(甲醇),薄層色譜濃硫酸顯色呈紫紅色。ESI-MS m/z 415 [M+H]+,分子式C29H50O。1H-NMR (500 MHz,CDCl3) δ :0.65 (3H,s,H-18),1.00 (3H,s,H-19),0.90 (3H,m,H-21),0.82 (3H,m,H-26),0.84 (3H,s,H-27),0.89 (3H,s,H-29),3.50 (1H,m,H-3),5.33 (1H,d,J=2.4 Hz,H-6)。13C-NMR (150 MHz,CDCl3) δ:37.8 (C-1),31.6 (C-2),72.0 (C-3),42.1 (C-4),141.0 (C-5),121.8 (C-6),32.4 (C-7),32.8 (C-8),50.5 (C-9),38.0 (C-10),22.0 (C-11),42.3 (C-12),42.7 (C-13),59.4 (C-14),25.3 (C-15),30.0 (C-16),55.6 (C-17),11.5 (C-18),20.2 (C-19),36.7 (C-20),18.8 (C-21),36.2 (C-22),27.3 (C-23),44.8 (C-24),29.9 (C-25),17.6 (C-26),19.6 (C-27),22.9 (C-28),11.3 (C-29)。以上數(shù)據(jù)與文獻報道[4-5]基本-致,因此鑒定化合物1為β-谷甾醇(β-sitosterol)。

化合物(11): 白色針狀結(jié)晶(甲醇)。ESI-MS m/z 169 [M-H]-,分子式C7H6O5。1H-NMR (500 MHz,CD3OD) δ: 7.03 (2H,s,H-2,6);13C-NMR (125MHz,CD3OD) δ:121.8 (C-1),109.2 (C-2,6),145.4(C-3,5),138.5 (C-4),169.7 (C-7)。以上數(shù)據(jù)與文獻[20-21]報道基本一致,鑒定化合物11為沒食子酸(Gallic acid)。
化合物(12): 白色芳香粉末(乙酸乙酯)。ESI-MS m/z 301 [M-H]-,分子式C14H6O8。1H-NMR(500MHz,DMSO) δ: 7.51 (2H,s,H-5,5’);13C-NMR (125MHz,DMSO) δ: 106.4 (C-1,1’),136.1 (C-2,2’),140.3 (C-3,3’),148.0 (C-4,4’),109.6 (C-5,5’),113.6 (C-6,6’),160.0(C-7,7’)。波譜和質(zhì)譜檢測數(shù)據(jù)與文獻[21]報道基本一致,鑒定該化合物12為鞣花酸(Ellagic acid)。

通過系統(tǒng)分離,從傣藥珠子草中共分離鑒定出了14個單體化合物,分為甾體和三萜類化合物、木脂素類、黃酮類和多酚類等。根據(jù)文獻報道,珠子草具有肝保護活性的研究主要采用全草水提取物、鮮葉汁和甲醇提取物,開展肝保護活性研究的單體化合物主要有葉下珠脂素、葉下珠次素和珠子草素等[3],而抗病毒活性以珠子草的甲醇提取物活性最強[25-26]。通過對珠子草系統(tǒng)的化學成分研究,傣醫(yī)用其治療肝臟類疾病,主要是通過其含有的木脂素類成分發(fā)揮保肝作用,通過多酚類成分發(fā)揮抗病毒作用。
傣藥珠子草是西雙版納地區(qū)常用的藥材之一,資源豐富,臨床使用廣泛,尤其是在乙型肝炎的治療中屬于最常用的傣藥材之一。其化學成分和藥理活性已有一定的研究基礎,且有研究表明其提取物毒性低。根據(jù)國內(nèi)外文獻報道,珠子草除了用于肝病的治療,還有多種活性,因此具有廣闊的研究開發(fā)前景。
