施氮量對(duì)烤煙常規(guī)化學(xué)成分含量及主流煙氣中7種有害成分釋放量的影響

以烤煙豫煙12號(hào)為材料,通過(guò)田間試驗(yàn)研究不同施氮量對(duì)烤煙化學(xué)成分和主流煙氣中7種有害成分的影響。結(jié)果表明,施氮量不同不僅影響煙葉常規(guī)化學(xué)成分的含量,而且對(duì)主流煙氣中有害成分的釋放量產(chǎn)生了影響;隨施氮量增加,煙葉中蛋白質(zhì)、煙堿和總氮的含量增加,總糖和還原糖含量減少,鉀和氯含量影響不明顯;主流煙氣中7種有害成分隨施氮量的增加變化趨勢(shì)不同,隨施氮量增加,巴豆醛的釋放量減少,氨和NNK的釋放量增加;施氮量在55 kg/hm2時(shí),煙葉常規(guī)化學(xué)成分協(xié)調(diào),主流煙氣中7種有害成分的危害性指數(shù)最低,煙草質(zhì)量安全性較高。

隨著消費(fèi)者對(duì)吸煙與健康的日益關(guān)注和控?zé)熈Χ鹊牟粩嗉訌?qiáng),“減害降焦”成為煙草行業(yè)迫切需要解決的重大問(wèn)題,降低煙草危害對(duì)煙草行業(yè)發(fā)展和提升國(guó)際競(jìng)爭(zhēng)力也具有重要意義[1]。隨著科學(xué)技術(shù)的發(fā)展,煙草減害的手段和技術(shù)也越來(lái)越豐富,關(guān)于降低烤煙主流煙氣中7種有害成分的研究,多集中在煙草加工和減害添加劑領(lǐng)域[2-10],在農(nóng)業(yè)措施方面未見(jiàn)報(bào)道。而在很大程度上卷煙有害成分的釋放量是由原料決定的,顏克亮等[11]研究表明,上部煙葉中的總氮、蛋白質(zhì)及煙堿對(duì)危害性指數(shù)H值有顯著的影響,總氮含量與NH3、苯并芘呈顯著正相關(guān);昆明地區(qū)煙葉中總氮、煙堿、蛋白質(zhì)及總糖含量與巴豆醛釋放量成顯著相關(guān)性;紅河地區(qū)煙葉中煙堿含量與危害性指數(shù)H值呈極顯著正相關(guān)。吳清輝[12]研究表明,主流煙氣中苯酚含量與煙葉中總糖和還原糖含量呈顯著負(fù)相關(guān),與煙堿、總氮和氯含量呈顯著正相關(guān)。主流煙氣中7項(xiàng)有害成分與煙葉化學(xué)成分存在一定的相關(guān)性。關(guān)于施氮量不同對(duì)烤煙生長(zhǎng)發(fā)育和烤后煙葉主要化學(xué)成分有明顯影響已有較多報(bào)道[13-16],但對(duì)烤煙主流煙氣中7種有害成分的影響未見(jiàn)報(bào)到。本研究通過(guò)探討施氮量對(duì)煙葉常規(guī)化學(xué)成分及主流煙氣7種有害成分的影響,旨在為降低烤煙危害性提供煙田合理施肥的理論參考。
試驗(yàn)于2015年在河南省登封市穎陽(yáng)鎮(zhèn)進(jìn)行,試驗(yàn)地肥力中等,土壤有機(jī)質(zhì)含量13.67 g/kg,堿解氮62.56 mg/kg,速效磷15.43 mg/kg,速效鉀134.59 mg/kg??緹煿┰嚻贩N為豫煙12號(hào),固定磷、鉀肥的用量,供試肥料為煙草專用復(fù)合肥和芝麻餅肥。
試驗(yàn)采用隨機(jī)區(qū)組設(shè)計(jì),共設(shè)置4個(gè)處理,氮素用量設(shè)為T1(40 kg/hm2),T2(55 kg/hm2),T3(70 kg/hm2),T4(85 kg/hm2);除氮以外,保持各處理土壤肥力一致,用NH4NO3滿足4個(gè)氮素處理水平,每個(gè)處理面積667 m2,3次重復(fù),四周設(shè)置保護(hù)行。煙株間行距120 cm,株距50 cm,于2015年4月28日移栽,其他田間管理措施按優(yōu)質(zhì)煙栽培技術(shù)規(guī)范進(jìn)行。成熟后煙樣采用三段式烘烤工藝完成烘烤,取各處理煙樣C3F煙葉。
測(cè)定烤后煙葉中常規(guī)化學(xué)成分和單料煙主流煙氣中7種有害成分(CO、HCN、NNK、B[a]P、苯酚、巴豆醛、NH3)。
煙堿:參照YC/T160-2002;總氮:參照YC/T161-2002;蛋白質(zhì):參照YC/T166-2003;總糖:參照YC/T 159-2002;還原糖:參照YC /T 216-2007;鉀:參照YC/T217-2007;氯:參照YC/T162-2002。
將煙葉樣品進(jìn)行回潮、去梗切絲(寬度約1.0mm)備用,然后采用統(tǒng)一空煙筒用手動(dòng)卷煙機(jī)將處理過(guò)的煙絲卷制成84×24mm的煙支作為研究對(duì)象。將卷制的煙支樣品置于溫度(22±1)℃、相對(duì)濕度(60±2)%的環(huán)境下平衡48 h,根據(jù)質(zhì)量允差選取平均質(zhì)量0.90±0.02g和平均吸阻1000±50Pa的煙支為測(cè)試樣品,供試樣品的物理指標(biāo)和抽吸參數(shù)見(jiàn)表1。一氧化碳(CO)、氰化氫(HCN)、4-(甲基亞硝胺基-)1-(3-吡啶基-)1-丁酮(NNK)、苯并[a]芘(B[a]P)、苯酚、巴豆醛、氨(NH3)的測(cè)定分別按照標(biāo)準(zhǔn)方法GB/T 23356-2009[17]、YC/T 253-2008[18]、GB/T 23228-2008[19]、GB/T 21 130-2007[20]、YC/T 255-20087[21]、YC/T 254-2008[22]、YC/T 377-2010[23]進(jìn)行。

卷煙危害性評(píng)價(jià)指數(shù)計(jì)算公式[24]為:
其中:A為CO釋放量(mg/支),B為NNK釋放量(ng/支),C為B[a]P釋放量(ng/支),D為巴豆醛釋放量(μg/支),E為苯酚釋放量(μg/支),F(xiàn)為NH3釋放量(μg/支),G為HCN釋放量(μg/支)。
采用SPSS 21.0和Microsoft Excel 2010 軟件進(jìn)行統(tǒng)計(jì)分析,對(duì)數(shù)據(jù)進(jìn)行差異顯著性分析,顯著性水平P <0 .05,以a、b、c等表示其差異性,數(shù)字后不含有相同字母表示差異有統(tǒng)計(jì)學(xué)意義。
施氮量不同對(duì)烤煙煙葉常規(guī)化學(xué)成分含量的影響見(jiàn)表2,由表2可以看出,總糖、還原糖含量和糖堿比隨施氮量的增加而減少,蛋白質(zhì)、煙堿和總氮含量隨施氮量的增加而增加;總糖含量在40 kg/hm2與55 kg/hm2之間差異沒(méi)有統(tǒng)計(jì)學(xué)意義,其他處理間均達(dá)到差異有統(tǒng)計(jì)學(xué)意義;還原糖、蛋白質(zhì)含量和糖堿比在各處理間差異均達(dá)到有統(tǒng)計(jì)學(xué)意義;煙堿含量在55 kg/hm2與70 kg/hm2之間差異沒(méi)有統(tǒng)計(jì)學(xué)意義,其他處理間均差異有統(tǒng)計(jì)學(xué)意義;總氮含量在40 kg/hm2與85 kg/hm2之間差異有統(tǒng)計(jì)學(xué)意義,其他處理間均未達(dá)到有統(tǒng)計(jì)學(xué)意義;氮堿比在85 kg/hm2時(shí)顯著低于其他處理,而其他處理間差異均未達(dá)到有統(tǒng)計(jì)學(xué)意義;鉀、氯含量和鉀氯比隨施氮量的增加在各處理間差異沒(méi)有統(tǒng)計(jì)學(xué)意義,其含量變化隨施氮量的影響較小。
不同施氮量對(duì)主流煙氣中有害成分釋放量及危害性指數(shù)的影響見(jiàn)表3。由表3可以看出,施氮量對(duì)主流煙氣不同有害成分釋放量的影響趨勢(shì)不同,CO的釋放量隨施氮量的增加無(wú)增減趨勢(shì),其釋放量在各處理間差異沒(méi)有統(tǒng)計(jì)學(xué)意義;HCN的釋放量隨施氮量大體呈低氮量(40~55 kg/hm2)時(shí)釋放量少,高氮量(70~85 kg/hm2)時(shí)釋放量多,但在各處理間其釋放量差異沒(méi)有統(tǒng)計(jì)學(xué)意義;苯酚在施氮量為40 kg/hm2和85 kg/hm2時(shí)其釋放量較高,施氮量為55~70 kg/hm2時(shí)其釋放量較低,且T1、T4的釋放量顯著低于T2、T3;氨和NNK的釋放量隨施氮量的增加而增加,施氮量在85 kg/hm2時(shí),氨的釋放量與其他處理之間差異有統(tǒng)計(jì)學(xué)意義,其他處理間釋放量的差異氨沒(méi)有統(tǒng)計(jì)學(xué)意義;NNK的釋放量T1與T2,T3與T4處理間差異沒(méi)有統(tǒng)計(jì)學(xué)意義,其他處理間差異有統(tǒng)計(jì)學(xué)意義;巴豆醛的釋放量隨施氮量的增加而減少,T1與T2,T3與T4沒(méi)有統(tǒng)計(jì)學(xué)意義,其他處理間差異有統(tǒng)計(jì)學(xué)意義;苯并芘的釋放量隨施氮量的增加呈先減少后增加的趨勢(shì),T1(40 kg/hm2)含量最高,T3(70 kg/hm2)含量最低,T1與T3間差異有統(tǒng)計(jì)學(xué)意義,其他處理間差異沒(méi)有統(tǒng)計(jì)學(xué)意義;危害性評(píng)價(jià)指數(shù)H值隨施氮量的增加呈先減少后增加的趨勢(shì),施氮量在55 kg/hm2(T2)時(shí),危害性指數(shù)(H值)最低,施氮量在85 kg/hm2(T4)時(shí),危害性指數(shù)最高,T4處理顯著高于其他處理,而其他處理間差異沒(méi)有統(tǒng)計(jì)學(xué)意義。

本試驗(yàn)結(jié)果表明,隨施氮量增加,煙葉中蛋白質(zhì)、煙堿和總氮的含量呈增加趨勢(shì),總糖和還原糖含量呈減少趨勢(shì),這與崔保偉等[25]研究施氮量對(duì)烤煙化學(xué)品質(zhì)的影響結(jié)果相同,說(shuō)明氮素對(duì)煙株常規(guī)化學(xué)成分有顯著作用。主流煙氣中7種有害成分隨施氮量的增加變化趨勢(shì)不同,隨施氮量的增加,氨和NNK的釋放量隨之增加,巴豆醛的釋放量隨之減少,苯并芘釋放量呈先減少后增加的趨勢(shì),CO和HCN的釋放量在各處理間均未達(dá)到顯著差異水平;苯酚在施氮量為55~70 kg/hm2范圍內(nèi)釋放量較低,并顯著低于在40 kg/hm2(較低施氮量)和85 kg/hm2(較高施氮量)條件下的釋放量。由危害性指數(shù)H值評(píng)價(jià)不同施氮水平下烤煙的總危害性可知,施氮量在85kg/hm2時(shí),危害性指數(shù)H值顯著高于其他處理;在40~70 kg/hm2時(shí),其危害性指數(shù)H值無(wú)顯著性差異,但施氮量在55 kg/hm2時(shí),危害指數(shù)H值最低。
煙草對(duì)氮素營(yíng)養(yǎng)十分敏感,土壤中氮肥施用量的高低對(duì)烤煙碳氮代謝平衡影響不同,協(xié)調(diào)影響烤后煙葉化學(xué)成分,對(duì)煙葉常規(guī)化學(xué)成分與主流煙氣中7種有害成分的相關(guān)性研究已有諸多報(bào)道[11-12,26]??偟焿A、蛋白質(zhì)與氨、苯并芘成正相關(guān),與巴豆醛呈負(fù)相關(guān);總糖、還原糖與巴豆醛呈正相關(guān),與苯酚呈負(fù)相關(guān)。本試驗(yàn)表明,施氮量不同影響煙葉常規(guī)化學(xué)成分的含量,對(duì)主流煙氣中有害成分的釋放量進(jìn)而產(chǎn)生了影響,施氮量過(guò)高或過(guò)低煙葉主流煙氣中有害成分的釋放量均較高,合理施用氮肥可降低主流煙氣中有害成分的釋放量,提高卷煙安全性。本試驗(yàn)僅對(duì)一個(gè)單一的施氮肥水平進(jìn)行了初步研究,施氮量對(duì)鉀和氯含量影響不明顯,所以在主流煙氣中7種有害成分與其相關(guān)性有待進(jìn)一步探索。

E ff ect of nitrogen application rate on routine chemical components in fl ue-cured tobacco and delivery of seven harmful components in mainstream smoke

Field experiments with tobacco cultivar Yuyan No.12 were carried out to study e ff ects of di ff erent nitrogen fertilizer rates on chemical components and 7 harmful components delivery in mainstream smoke. Results showed that different nitrogen fertilizer rates a ff ected content of routine chemical compositions, and had in fl uence on delivery of harmful components in mainstream smoke. Contents of protein, nicotine and total nitrogen increased, while total sugar and reducing sugar decreased with the increase of nitrogen fertilizer rate,while its e ff ect on e ff ect on potassium and chlorine content were not obvious. Nitrogen fertilizer rate had di ff erent e ff ects on 7 harmful components in mainstream smoke. Crotonaldehyde decreased while ammonia and N-nitrosamines increased along with the increase of nitrogen fertilizer rate. When treated with 55 kg/hm2nitrogen fertilizer, conventional chemical composition was coordinate, hazardous index of seven harmful components in mainstream smoke was lowest.
