        Centella asiatica(Linn.)belongs to the family of Apiaceae (Umbeliferae).It has been widely used for traditional medicine,especially for wound healing,anti-infammatory, antipsoriatic,antiulcer,antibacterial and antifungal.The biologically active ingredients of Centella asiatica are believed to be triterpenes,particularly asiatic acid,madecassic acid, asiaticoside and madecassoside(Fig.1).Due to its ability to stimulate collagen,Centella has been used in skin care products for restoring skin frmness and elasticity and improving skin appearance[1].The topical formulations of Centella asiatica extract have been developed as scaffolds[2]and transdermal patch[3].Interestingly,there have been no reports on the preparation of Centella asiatica extract topical spray for the treatment of fresh wounds such as heat burns or accidental injuries.

        To prepare the experiment,the topical spray containing Centella asiatica extract had to include hydroxypropyl-βcyclodextrin(HP-β-CD)and was formulated with other excipients,including Eudragit E100,glycerine,PEG400, copovidone,ethanol and purifed water in order to obtain clear yellowish solution(F1-F8).Physical properties and excision wounds in rats were investigated for 14 days.The assay content of triterpenes of the extract was found having 0.12%of asiatic acid,0.54%of madecassic acid,0.25%of asiaticoside and 1.02% of madecassoside respectively.The physical properties of the topical spray showed pH of the solution at about 5.5–6.0.Viscosity was in the range of 20–60 cps and the surface tension was 20.3–24.6 mN/m.In addition,the contact angle was less than 20°.The triterpenes results in the topical formulation revealed that the contents were closed to 100%of the triterpenes in extract.In this study,we selected the formulation F8 due to the medium level of the viscosity and highest score of satisfactory evaluation by the volunteers.To evaluate the effcacyof the extract topical spray,male Sprague–Dawley rats weighing 250–300 g were randomly divided in 3 groups(n=6 of each group).Group 1 was an untreated and controlled group.In Group 2 the animals received povidone iodine solution(Betadine?) treatment,while in Group 3 the animals received Centella asiatica extract topical spray formulation F8.The excision wound fromthe marked area was excised to produce wound measuring around 200 mm2.The test substances were applied topically once a day.The wound contraction was measured at interval of 2 days for 14 days.No other topical or systemic therapy was given to the animals during the course of this study.A skin irritation study indicated that the formulation was nonirritating in a rat model.The results showed that the wound excision received the signifcantly completed wound healing recovery after the 14th day.However,Group 2 treated with conventional treatment of wound by povidone iodine solution showed the highest wound healing activity due to antimicrobial activity of povidone iodine.Formulation F8 containing 1% Centella asiatica extract showed high effcacy to treatment of fresh wound.To summarize,Centella asiatica extract–HP-β-CD complex formulated as topical spray enhanced wound healing.
