松針多糖對(duì)肉雞生產(chǎn)性能和免疫功能的影響

本研究旨在探討松針多糖對(duì)肉雞生長(zhǎng)性能和免疫功能的影響。選取240只1日齡的815肉雞,隨機(jī)分為4個(gè)處理,每個(gè)處理4個(gè)重復(fù),每個(gè)重復(fù)15只雞,分別在基礎(chǔ)飼糧中添加0(對(duì)照)、0.05%、0.10%和0.15%的松針多糖提取物。試驗(yàn)期為42 d,測(cè)定處理組生長(zhǎng)性能、免疫器官指標(biāo)、血清免疫指標(biāo)。結(jié)果顯示,在生長(zhǎng)前期(1-21 d),松針多糖對(duì)肉雞的生產(chǎn)性能沒有影響(P>0.05),0.05%多糖組白細(xì)胞介素2的濃度極顯著高于對(duì)照組(P<0.01),對(duì)腫瘤壞死因子、胸腺素及免疫器官指標(biāo)影響不顯著(P>0.05)。在生長(zhǎng)后期(22-42 d),0.10%多糖組平均日增重顯著高于對(duì)照組(P<0.05),對(duì)平均體重及料重比無顯著影響(P>0.05),0.05%多糖組腫瘤壞死因子濃度顯著高于對(duì)照組(P<0.05),而對(duì)白細(xì)胞介素2、胸腺素及免疫器官指標(biāo)影響不顯著(P>0.05)。結(jié)果表明,飼糧中添加適量的松針多糖在一定程度上可以提高肉雞的免疫功能和生產(chǎn)性能,0.10%的松針多糖添加量較為適宜。
松針多糖;肉雞;生長(zhǎng)性能;免疫功能
松針為松科(Pinaceae)松屬(Pinus)植物的針狀葉,在我國(guó)資源較為豐富[1]。研究發(fā)現(xiàn),松針提取物中富含蛋白質(zhì)、粗脂肪、多糖、多種氨基酸及微量礦物質(zhì)元素、維生素等多種營(yíng)養(yǎng)成分,具有促進(jìn)血液循環(huán)、提高機(jī)體免疫力等功效[2-3]。在畜牧業(yè)生產(chǎn)中,因飼用抗生素普遍使用而引起的藥物殘留不僅危害人類健康,而且造成環(huán)境污染。為此,眾多研究者致力于尋找天然低毒的飼料添加劑以替代抗生素產(chǎn)品。大量研究表明,植物多糖具有抗炎、抗菌、抗病毒等作用,且低毒或無毒,殘留少,是一種天然的綠色飼料添加劑[4]。此外,植物多糖還具有調(diào)節(jié)細(xì)胞的生長(zhǎng)和衰老、增強(qiáng)淋巴細(xì)胞活性、促進(jìn)抗體生成等功能,可作為免疫增強(qiáng)劑使用[5]。目前已開展研究的多糖種類豐富,部分已在家禽生產(chǎn)中推廣應(yīng)用,如黃芩多糖[6]、黃芪多糖[7]和苜蓿多糖[8]都能提高肉雞血清中免疫球蛋白水平,調(diào)節(jié)肉雞免疫能力,提高肉雞生產(chǎn)性能。松針多糖是松針的一種有效成分,含有鼠李糖、葡萄糖及果糖等多種可溶性糖類[9]。目前國(guó)內(nèi)松針多糖的研究大多還停留在提取純化的階段,在動(dòng)物營(yíng)養(yǎng)中研究應(yīng)用較少。815肉雞是江西五鳳家禽公司培育的以AA肉雞為父本、本地黃雞為母本、飼養(yǎng)周期相對(duì)較長(zhǎng)、肉質(zhì)口感較好的一種小型肉用雞品種。本研究通過在日糧中添加松針多糖后測(cè)定815肉雞的生長(zhǎng)性能、免疫器官指數(shù)和血清免疫指標(biāo),探討松針多糖對(duì)肉雞生長(zhǎng)性能和免疫機(jī)能的影響,旨為進(jìn)一步開發(fā)利用松針多糖飼料添加劑提供理論依據(jù)。
1.1試驗(yàn)材料
松針多糖購(gòu)自陜西西安文竹生物有限公司(批號(hào):479-30-6),為黃色粉末,經(jīng)實(shí)驗(yàn)室采用蒽酮硫酸比色法測(cè)定多糖的含量為69%。
1.2試驗(yàn)設(shè)計(jì)與飼糧組成
選取體重相近的1日齡健康的815肉仔雞240只,采用單因素試驗(yàn)設(shè)計(jì),隨機(jī)分成4個(gè)處理,每個(gè)處理4個(gè)重復(fù),每個(gè)重復(fù)15只。以基礎(chǔ)日糧為對(duì)照組,各試驗(yàn)處理組分別在基礎(chǔ)飼糧中添加0.05%、0.10%和0.15%的松針多糖。飼養(yǎng)期42 d,分為前期(1-21 d)和后期(22-42 d)兩個(gè)階段?;A(chǔ)日糧參考美國(guó)國(guó)家研究委員會(huì)(NRC)《家禽營(yíng)養(yǎng)需要》(1994)和中國(guó)雞飼養(yǎng)標(biāo)準(zhǔn)(2004)黃羽肉雞仔雞的營(yíng)養(yǎng)需要配制,日糧配方與營(yíng)養(yǎng)水平見表1。
1.3飼養(yǎng)管理
4個(gè)處理組的肉雞全期采用籠養(yǎng)的方式,雛雞自由采食與飲水,雞舍內(nèi)光照、溫度和濕度嚴(yán)格按照常規(guī)飼養(yǎng)管理要求進(jìn)行,育雛期輔以人工保溫。免疫程序一致,分別在7、14、21 d早上08:00對(duì)雞進(jìn)行新城疫與法氏囊免疫。采用飲水免疫,嚴(yán)格按照說明書推薦量添加,接種疫苗前一天24:00斷水。每日觀察試驗(yàn)雞的精神狀態(tài)、采食及糞便情況,記錄死亡只數(shù)。
1.4測(cè)定指標(biāo)與方法
1.4.1生長(zhǎng)性能指標(biāo)試驗(yàn)分別于第21、42天早晨逐只空腹稱重,稱重前一天20:00禁食,自由飲水。于次日08:00逐只稱重,記錄飼料消耗量。
試驗(yàn)過程中詳細(xì)記錄各組雞的健康狀況和各階段的采食量,并觀察雞群的變化。計(jì)算平均日采食量(ADFI)、平均體重(ABW)、平均日增重(ADG)和料重比(F/G)。
1.4.2免疫器官指數(shù)和血清免疫指標(biāo)試驗(yàn)分別在第21、42天選取每個(gè)重復(fù)兩只接近平均體重的雞進(jìn)行屠宰,屠宰時(shí)斷頸靜脈采血,制備血清樣品于-20℃保存。摘取脾臟、胸腺、法氏囊,剔除脂肪后稱鮮重。然后計(jì)算脾臟指數(shù)、胸腺指數(shù)、法氏囊指數(shù)。血清中白細(xì)胞介素2(IL-2)、腫瘤壞死因子(TNF-α)和胸腺素的含量采用酶聯(lián)免疫法[10-11]測(cè)定。白細(xì)胞介素2試劑盒購(gòu)自南京建成生物公司(批號(hào)20141048457),腫瘤壞死因子試劑盒購(gòu)自南京建成生物公司(批號(hào)20141049025),胸腺素試劑盒購(gòu)自南京建成生物公司(批號(hào)20141053224)。

免疫器官指數(shù)=免疫器官重量/活體重×100%。
1.5數(shù)據(jù)處理
采用SPSS 17.0統(tǒng)計(jì)軟件對(duì)各數(shù)據(jù)進(jìn)行單因素方差分析(One-way ANOVA)和多重比較(Duncan’s法),試驗(yàn)數(shù)據(jù)以“平均值±標(biāo)準(zhǔn)差”來表示。
2.1松針多糖對(duì)肉雞生長(zhǎng)性能的影響
松針多糖對(duì)肉雞的平均體重?zé)o顯著影響(P>0.05);但在生長(zhǎng)后期,多糖組的平均體重都高于對(duì)照組,其中0.10%多糖組平均體重最高(表2)。在前期,對(duì)照組的平均日增重最高;到了生長(zhǎng)后期,多糖組的平均日增重都高于對(duì)照組,0.10%多糖組最高,顯著高于對(duì)照組(P<0.05)。在前期,松針多糖組的平均日采食量均高于對(duì)照組;但在后期則低于對(duì)照組,但各組之間采食量差異不顯著(P>0.05)。松針多糖組料重比前期高于對(duì)照組(P>0.05),后期低于對(duì)照組(P>0.05)。從全期來看,0.10%松針多糖組的平均日增重最高,料重比最低。
2.2松針多糖對(duì)肉雞脾臟、胸腺、法氏囊指數(shù)的影響
所有屠宰的肉仔雞免疫器官外觀未見異常(表3)。21日齡時(shí),隨著多糖添加水平的提高,多糖組的脾臟指數(shù)也有緩慢的提高,但差異不顯著(P>0.05)。在42日齡時(shí),0.10%多糖組的脾臟指數(shù)最高,而0.05%多糖組和0.15%多糖組均低于對(duì)照組(P>0.05)。松針多糖對(duì)肉雞的各組胸腺指數(shù)均無顯著影響(P>0.05)。21日齡時(shí),0.10%多糖組的胸腺指數(shù)最高;在42日齡時(shí),試驗(yàn)組的胸腺指數(shù)均低于對(duì)照組(P>0.05),以0.10%多糖組的胸腺指數(shù)與對(duì)照組的最為接近。隨著飼養(yǎng)日齡的增加,法氏囊指數(shù)呈現(xiàn)逐漸減少的趨勢(shì),并且各組法氏囊指數(shù)均無顯著差異(P>0.05)。在21日齡時(shí),多糖組中0.10%多糖組的法氏囊指數(shù)最高,0.05%多糖組次之;42日齡時(shí),0.05%多糖組的法氏囊指數(shù)最高,而0.10%多糖組和0.15%多糖組的法氏囊指數(shù)均低于對(duì)照組。
2.3松針多糖對(duì)肉雞血清IL-2、TNF-α和胸腺素含量的影響
血清中的IL-2的濃度后期大于前期,0.05%多糖組IL-2最高,21日齡時(shí),0.05%多糖組IL-2的濃度極顯著高于對(duì)照組(P<0.01)(表4)。42日齡時(shí),各組無顯著差異(P>0.05),其中0.15%多糖組IL-2的濃度高于其它各組。TNF-α的濃度前期大于后期。在21日齡時(shí),各組TNF-α的濃度無顯著差異(P>0.05),0.05%多糖組TNF-α的濃度最高。42日齡時(shí),0.05%多糖組TNF-α濃度顯著高于對(duì)照組(P<0.05)。在21日齡時(shí),各組胸腺素濃度無顯著差異(P>0.05),0.05%多糖組胸腺素濃度最高。42日齡時(shí),試驗(yàn)組的胸腺素濃度均高于對(duì)照組,0.05%多糖組的胸腺素濃度最高,比對(duì)照組提高了49.50%。

3.1松針多糖對(duì)肉雞生長(zhǎng)性能的影響
有研究表明,在肉雞飼糧中添加松針粉可顯著降低料重比,提高平均日增重,對(duì)肉雞生產(chǎn)有促進(jìn)作用[12-13]。這可能與松針粉中含有植物多糖活性成分密切相關(guān)。多糖類物質(zhì)能夠促進(jìn)核酸和蛋白質(zhì)的合成,對(duì)機(jī)體特異性免疫與非特異性免疫、細(xì)胞免疫與體液免疫均有重要影響,具有重要的生物學(xué)和生理學(xué)功能,尤以對(duì)動(dòng)物免疫調(diào)控作用突出[14]。日糧中添加2%的紅酒香菇多糖能顯著提高肉雞的生長(zhǎng)性能和飼料轉(zhuǎn)化效率[15]。在飼糧中添加1%的黃芪多糖可提高肉雞平均日增重,平均日采食量,降低料重比[16]。植物多糖改善動(dòng)物生長(zhǎng)性能的機(jī)制可能是通過增強(qiáng)機(jī)體的免疫力,提高腸道內(nèi)有益菌群的數(shù)量,使?fàn)I養(yǎng)物質(zhì)容易被吸收利用[17]。本研究結(jié)果表明,在飼糧中添加0.10%的松針多糖組,能提高肉仔雞的體重和日增重,降低料重比,對(duì)肉雞生產(chǎn)有促進(jìn)作用。但從整體來看,松針多糖組的平均日采食量要低于對(duì)照組,這可能與多糖提取物對(duì)肉雞采食量有一定程度的抑制作用相關(guān)。有些研究證實(shí),多糖提取物對(duì)動(dòng)物采食量及飼料轉(zhuǎn)化率有一定的負(fù)面影響,其原因是多糖通過增加食糜黏度,妨礙消化酶與營(yíng)養(yǎng)物質(zhì)的接觸而降低消化利用率[18];較高劑量的多糖因與脂肪絡(luò)合抑制脂肪吸收,干擾了機(jī)體的正常功能,導(dǎo)致營(yíng)養(yǎng)物質(zhì)供給不足,進(jìn)而影響機(jī)體的生長(zhǎng),降低了動(dòng)物的生長(zhǎng)性能[19]。目前研究可見,關(guān)于多糖對(duì)肉雞生長(zhǎng)性能的影響存在爭(zhēng)議,還有待于進(jìn)一步研究探索。
3.2松針多糖對(duì)肉雞免疫器官指數(shù)的影響
免疫器官的重量與動(dòng)物免疫功能密切相關(guān)[20]。家禽的免疫器官主要為胸腺、脾臟和法氏囊等[21]。胸腺、脾臟和法氏囊的重量可用于評(píng)價(jià)雞的免疫狀況[22-24]。多糖可影響免疫器官的發(fā)育。飼糧中添加黃芩多糖能提高肉雞的胸腺指數(shù)、脾臟指數(shù)和法氏囊指數(shù),尤其是200 mg·g-1添加組對(duì)各項(xiàng)免疫器官指數(shù)的的效果最好[6]。肉雞4周齡、7周齡時(shí),添加1%、3%和5%的松針粉能有效促進(jìn)肉雞脾臟、胸腺和法氏囊免疫器官的發(fā)育,尤其是添加 5% 松針粉組效果明顯,7周齡時(shí)添加3%松針粉組效果較明顯[25]。本研究結(jié)果表明,0.10%的松針多糖能夠提高肉仔雞的免疫器官指數(shù),但多糖對(duì)免疫器官指數(shù)的影響受生長(zhǎng)時(shí)期、添加劑量和不同器官的影響。
3.3松針多糖對(duì)肉雞IL-2、TNF-α和胸腺素含量的影響
IL-2、TNF-α和胸腺素是參與肉雞體液免疫的細(xì)胞因子,它們?cè)谘逯泻康亩嗌伲梢蚤g接影響動(dòng)物的細(xì)胞免疫和體液免疫。黃芪多糖復(fù)方雞可以通過調(diào)節(jié)細(xì)胞因子IL-10和IFN-γ的分泌來提高機(jī)體的免疫功能[26]。黃芪多糖還可以提高血清細(xì)胞因子IL-1、IL-2和TNF-α的含量,但對(duì)其影響呈劑量依賴關(guān)系,血清IL-1 、IL-2和TNF-α黃芪多糖添加劑量的增加而增加,當(dāng)添加量提高到2.0%時(shí),各試驗(yàn)階段肉仔雞血清IL-1、IL-2和TNF-α含量均有不同程度的降低[27]。本研究表明,0.05%松針多糖能夠提高生長(zhǎng)前期IL-2和生長(zhǎng)后期TNF-α的濃度,但再增加劑量效果反而下降。
飼糧中添加適量松針多糖可在一定程度上提高肉仔雞血清中白細(xì)胞介素2和腫瘤壞死因子濃度,加強(qiáng)肉雞免疫機(jī)能,促進(jìn)肉雞的生長(zhǎng),提高肉雞的體重和日增重,降低料重比。綜合考慮,以0.10%的松針多糖添加量效果較好。

Effects of pine needles polysaccharide on growth performance and immune functions of broilers

This experiment was conducted to investigate the effects of pine needles polysaccharide on growth performance and immune functions of broilers. Two hundred and forty one-day-old 815 broilers were randomly divided into 4 groups with 4 replicates in each group and 15 broilers in each replicate. The basal diets were supplemented with 0, 0.05%, 0.10% and 0.15% pine needles polysaccharide, respectively. The experiment lasted for 42 days. Growth performance, immune organomdocws indices, serum immune indices were determined in different periods. The results showed as follows: in the early stage (1-21 d), there was no significant difference among all groups in the growth performance (P>0.05). The concentration of IL2 in the 0.05% polysaccharide group was extremely significantly higher than that in control group (P<0.01), while pine needles polysaccharide took no effect on the TNFα, thymosin concentration and immune organ indices (P>0.05). In the later stage (22-42 d), ADG (average daily gain) of the broilers in the 0.10% polysaccharide group was significantly higher than that in control group (P<0.05). There was no difference in ABG (average body gain) and F/G (feed/gain) in all the groups (P>0.05). The concentration of TNFα in 0.05% polysaccharide group was significantly higher than that in control group (P<0.05). In contrast, there was no significant difference in the IL2, thymosin concentration and immune organ indices in all groups (P>0.05). In conclusion, pine needles polysaccharide could improve the growth performance and immune functions of broilers. The adding amount of 0.05%~0.10% pine needles polysaccharide was appropriate.
