摘 要:該研究計(jì)劃通過分子生物學(xué)技術(shù)、神經(jīng)電生理技術(shù)、組織化學(xué)與免疫組化方法以及視覺行為學(xué)檢測(cè)方法,研究視神經(jīng)損傷的病理及修復(fù)機(jī)制,探討納米材料、神經(jīng)營(yíng)養(yǎng)因子、新型Nogo受體抑制劑及抗炎因子等對(duì)視神經(jīng)損傷的修復(fù)與保護(hù)機(jī)制,防治視神經(jīng)及節(jié)細(xì)胞的損傷,為視神經(jīng)假體的成功應(yīng)用奠定基礎(chǔ)。該年度主要在以下幾個(gè)方面開展了研究:(1)探索微電極刺激視神經(jīng)的安全電流強(qiáng)度范圍、刺激時(shí)間時(shí)程;(2)測(cè)試電極假體插入視神經(jīng)后的缺血狀態(tài)以及缺血對(duì)視網(wǎng)膜的影響;(3)探索3,4-二羥基苯甲酸甲酯(MDHB)對(duì)氧化應(yīng)激所致RGC-5細(xì)胞凋亡的拮抗作用及其作用機(jī)制;(4)探索原花青素對(duì)視網(wǎng)膜節(jié)細(xì)胞的保護(hù)作用及其作用機(jī)制;(5)探索EphA2基因在視網(wǎng)膜神經(jīng)節(jié)細(xì)胞損傷中的作用。

Abstract:This project plans to explore the repair and protection of optic nerve after its damage, through molecular, electrophysiological, histological, immunological and behavioral test. The nana-material, neural nutrition factors, new Nogo receptor inhibitors and anti-inflammation factors will be applied to test their effects on the protection of optic nerve, in order to prevent the optic nerve damage and the death of retinal ganglion cells. The ultimate object is to provide foundation for the clinical application of optical nerve prosthesis. In this year, we developed the following researches:(1)Explored the safe range of the electric intensity and stimulating duration of micro-electrode inserted into the optic nerve.(2)Explored whether the insertion of micro-electrode would cause ischemia in optic nerve and how temporal ischemia in the brain affected the histology and function of retinal neurons and the visual behavior of the animal.(3)Explored the protection effect of MDHB on oxidative-stress injured RGC-5 cells and its mechanism.(4)Explored the protection effect of oligomeric proanthocyanidin on retinal ganglion cells against oxidative stress induced apoptosis in vitro.(5)Explored the function of EphA2 in the protection of retinal ganglion cells.
