[摘要] 目的 篩選人參皂苷及多糖最優(yōu)化微波提取工藝。 方法 通過單因素實驗和響應曲面法確定人參皂苷及多糖的微波輔助提取最佳工藝條件。 結果 人參皂苷的最佳提取條件為:固液比為1∶40,乙醇濃度為55%,微波提取時間為4 min,微波提取溫度為60℃;人參多糖的最佳提取條件為:固液比為1∶50,微波提取時間為2 min,微波提取溫度為90℃。按照優(yōu)化后工藝條件實驗,人參皂苷及多糖提取率分別為12.71%和23.32%。 結論 優(yōu)化后的工藝穩(wěn)定,微波輔助提取人參皂苷及多糖提取時間短,提取率高。

Optimization of microwave assisted extraction of Ginsenosides and Polysaccharide by respone surface methodology

[Abstract] Objective To screen optimal extraction process of ginsenosides and polysaccharides. Methods Single factor experiment and response surface method were applied and the optimum conditions were finally determined. Results The optimum extraction conditions of ginsenosides were: solid-liquid ratio was 1:40, ethanol concentration was 55%, microwave time was 4 min, microwave temperature was 60℃; the optimum extraction conditions of polysaccharides were: the solid-liquid ratio was 1∶50, extraction time was 2 min, the extraction temperature was 90℃. According to the optimized experimental conditions, ginsenosides and polysaccharide extraction rates were 12.71% and 23.32% respectively. Conclusion The optimized process is stable, microwave-assisted extraction of ginsenosides and polysaccharide are showed less time needed and the higher extraction rate.

人參為五加科多年生草本植物人參(Panax ginseng C. A.Mey.)的干燥根,為中華民族傳統(tǒng)中藥,有著4000多年的應用歷史,是東北三寶之一。在悠久的中醫(yī)藥文化歷史中,《神農(nóng)本草經(jīng)》首先記載了這一珍貴藥材,堪稱上品。據(jù)記載,人參具有“補五臟、安精神、定魂魄、止驚悸、除邪氣、明目開心益智、久服輕身延年”的功效[1]。人參的主要有效成分人參皂苷具有清熱解毒,促進腫瘤細胞的凋亡和分化[2],延緩衰老[3],調節(jié)中樞神經(jīng)系統(tǒng)[4],提高機體免疫力[5],改善心腦血管供血不足等作用。人參多糖可明顯增強免疫系統(tǒng)的功能并有抗腫瘤活性[6]。常用的中藥有效成分提取法有浸漬法[7]、滲漉法[8]、煎煮法[9]、回流法[10]、索氏提取法[11]、超聲波法[12]、超臨界CO2萃取法[13]、超高壓法[14]等。這些方法提取率一般較低,提取時間較長且能耗較大。微波輔助提取法是由微波提取和溶劑提取結合而成的一種新的提取技術,提取時間短、提取效率高并且重現(xiàn)性較好。
隨著現(xiàn)代醫(yī)藥科學的發(fā)展研究,對人參主要活性成分人參皂苷及多糖的研究日益深入。在傳統(tǒng)的人參有效成分提取中,人參皂苷和多糖的提取往往是單獨進行的,本實驗將提取人參皂苷后的人參渣經(jīng)干燥后用于人參多糖的提取,實現(xiàn)了人參原材料的充分利用。本研究通過單因素實驗考察了乙醇濃度、提取時間、提取溫度、液固比對人參皂苷提取率的影響;研究了提取時間、提取溫度、液固比對人參多糖提取率的影響;通過響應曲面中的Box-Behnken法優(yōu)化了提取工藝,得到微波輔助提取人參皂苷及多糖的優(yōu)化工藝參數(shù)。比較了水浴回流和微波輔助對人參皂苷和多糖的提取。
