摘要:采用RT-PCR、Western Blot、L-[35S]放射性標(biāo)記物摻入等方法,觀察甘蔗蠟提取物多廿烷醇對(duì)Hep G2細(xì)胞羥甲基戊二酸輔酶A(HMG-CoA)還原酶mRNA轉(zhuǎn)錄及蛋白質(zhì)表達(dá)量的影響,及其對(duì)HMG-CoA還原酶合成和降解的作用。結(jié)果表明,多廿烷醇降低HMG-CoA還原酶mRNA的轉(zhuǎn)錄和蛋白質(zhì)的表達(dá)量,且作用效果隨多廿烷醇濃度的升高而增強(qiáng)。此外,多廿烷醇抑制HMG-CoA還原酶的合成并加速其降解,多廿烷醇處理后HMG-CoA還原酶的半衰期明顯縮短。

Study on the Hypolipidemic Mechanism of Policosanol Extracted from Sugarcane Wax

Abstract: The effects of policosanol extracted from sugarcane wax on mRNA transcription and protein expression level, synthesis and degeneration of 3-hydroxy-3-methylglutarl-coenzyme (HMG-CoA) reductase in Hep G2 cell were determined by RT-PCR, Western Blot and L-[35S] radioactive isotope tracing technology. The results showed that the mRNA transcription and protein expression level of HMG-CoA reductase decreased with the increase of policosanol concentration(10,20,30 μg/L). Policosanol also suppressed the synthesis of HMG-CoA reductases while sped up its degeneration; the half life of HMG-CoA reductases treated by policosanol was shortened obviously.
