苦瓜Momordica charantia別名涼瓜、錦荔枝、賴(lài)葡萄,為葫蘆科苦瓜屬一年生草本植物,作為一種蔬菜作物廣泛種植于非洲和亞洲,具有抗糖尿病,抑菌等功效 (Grover and Yadav,2004;Beloin et al.,2005;Maurya,2009)。與其他葫蘆科作物相比,在其生長(zhǎng)發(fā)育過(guò)程中很少被植食性昆蟲(chóng)為害 (Yasui et al.,1998;Abe and Matsuda,2000),表現(xiàn)出較強(qiáng)的抗蟲(chóng)性 (Kumar et al.,1979;Mishraetal,2006)。大量的研究結(jié)果表明,苦瓜葉片提取物對(duì)多種害蟲(chóng)有明顯的拒食、忌避產(chǎn)卵和抑制生長(zhǎng)發(fā)育的作用,并從中分離鑒定出多種對(duì)昆蟲(chóng)有生物活性的苦瓜素類(lèi)化合物,如Momordicin I、II、Ⅳ和 3β,7β,25-三羥葫蘆烷-5,23二烯-19-醛-3-O-β-D-吡喃葡糖苷等,這些化合物是一類(lèi)葫蘆烷型四環(huán)三萜類(lèi)植物次生物質(zhì),對(duì)昆蟲(chóng)的作用方式主要是拒食、忌避產(chǎn)卵和抑制生長(zhǎng)發(fā)育 (Chandravadana et al.,1983;陳宏等,1996;李紹勤等,2001;Boeke,2004;Mekuria et al.,2005,2006;凌冰等,2008,2009;向亞林等,2009)。
我們前期的研究結(jié)果表明,苦瓜葉乙酸乙酯萃取物對(duì)小菜蛾幼蟲(chóng)有明顯的拒食和抑制生長(zhǎng)發(fā)育活性 (凌冰等,2008;向亞林等,2009),對(duì)美洲斑潛蠅成蟲(chóng)的拒食和產(chǎn)卵忌避作用顯著 (凌冰等,2009),對(duì)黃曲條跳甲也有明顯的拒食作用(楊舒媚等,2010)。駱?lè)f等 (另文發(fā)表)測(cè)定了苦瓜葉乙酸乙酯萃取物對(duì)斜紋夜蛾實(shí)驗(yàn)種群增長(zhǎng)的抑制作用,發(fā)現(xiàn)苦瓜葉乙酸乙酯萃取物對(duì)斜紋夜蛾幼蟲(chóng)的生長(zhǎng)發(fā)育也有明顯的抑制作用。為了研究苦瓜葉乙酸乙酯萃取物及其活性化合物Momordicin II對(duì)斜紋夜蛾的作用機(jī)理,我們測(cè)定了苦瓜葉片乙酸乙酯萃取物及其活性化合物Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)中腸淀粉酶和蛋白酶活性的影響,旨在探索苦瓜植株的抗蟲(chóng)機(jī)理,為尋找新的昆蟲(chóng)拒食活性物質(zhì),為合理利用苦瓜資源提供科學(xué)依據(jù)。
1.1.1 供試?yán)ハx(chóng)
斜紋夜蛾Spodoptera litura(Fabricius)由華南農(nóng)業(yè)大學(xué)資源環(huán)境學(xué)院昆蟲(chóng)生態(tài)研究室在人工氣候室內(nèi) [T:(27±1)℃;RH:70% ~80%;L∶D=14∶10]用人工飼料飼養(yǎng)的種群。斜紋夜蛾人工飼料參照陳其津等 (2000)的方法配制。
1.1.2 供試藥品
苦瓜葉乙酸乙酯萃取物及其活性化合物Momordicin II均由本課題組提供。其他試劑均為市售的化學(xué)純?cè)噭?,水由SZ-93自動(dòng)雙重純水蒸餾器制備 (上海亞榮生化儀器廠)。
1.2.1 苦瓜葉乙酸乙酯萃取物和Momordicin II混合飼料的配制
用丙酮分別配置不同濃度的苦瓜葉乙酸乙酯萃取物和Momordicin II藥液。將配制好的尚未凝固的人工飼料分別定量地放入保鮮盒內(nèi),按飼料:藥液等于5∶1的比例,將藥液加入人工飼料中,以丙酮作對(duì)照。待各濃度的含藥混合飼料中的溶劑自然揮發(fā)并凝固后,放入冰箱保存?zhèn)溆谩?/p>
1.2.2 試蟲(chóng)處理及酶液的制備
將大小一致的頭一天蛻皮成4齡的斜紋夜蛾幼蟲(chóng)饑餓4 h后,以不同濃度苦瓜葉乙酸乙酯萃取物和Momordicin II配制的人工飼料喂養(yǎng)斜紋夜蛾4齡幼蟲(chóng),于處理后24 h、48 h和72 h,每處理挑取大小一致的試蟲(chóng)在冰浴下迅速解剖出中腸,去圍食膜及食物殘?jiān)?,將整個(gè)中腸轉(zhuǎn)移到5 mL的組織勻漿器中,加4 mL 0.02 mol/L pH 8.0磷酸緩沖液,充分研磨后在4℃、4000 r/min條件下離心10 min,取上清液作酶液。
1.2.3 淀粉酶活性的測(cè)定
參考畢明娟等 (2010)的方法并略加改進(jìn)。取A、B兩支試管,在A管中加入0.2 mL酶液和0.8 mL 1%淀粉溶液作為底物,37℃水浴保溫15 min,再加入3,5-二硝基水楊酸試劑1.0 mL,沸水浴5 min,用蒸餾水定容至5 mL,然后用UV300紫外分光光度計(jì) (美國(guó)熱電公司)測(cè)定其在波長(zhǎng)520 nm處的吸光值。B管中加入0.8 mL磷酸緩沖液代替酶液,其余操作同A管。試驗(yàn)重復(fù)3次。根據(jù)A、B管吸光值差計(jì)算比活力,以每分鐘每毫克蛋白催化水解生成1 mg麥芽糖為1個(gè)單位 (U)。
1.2.4 蛋白酶活性的測(cè)定
參考潘科等 (2010)的酪蛋白水解比色方法。取A、B兩支試管,在A管中先加入酶液1 mL,再加0.5%酪蛋白溶液2.0 mL,37℃水浴保溫15 min后加入3 mL 10%三氯乙酸終止反應(yīng),以4000 r/min離心10 min,取上清液2.0 mL,按標(biāo)準(zhǔn)曲線制作方法測(cè)定其在波長(zhǎng)680 nm處的吸光值。B管中加入1.0 mL磷酸緩沖液代替酶液,其余操作同A管,試驗(yàn)重復(fù)3次。根據(jù)A、B管吸光值差計(jì)算比活力,以每分鐘每毫克蛋白催化水解生成1 μg酪氨酸為1個(gè)單位 (U)。
1.2.5 蛋白質(zhì)含量的測(cè)定
采用Bradford(1976)的考馬斯亮藍(lán)比色法。
本文中,所有數(shù)據(jù)均以“平均數(shù)±標(biāo)準(zhǔn)誤”表示,方差分析采用鄧肯氏新復(fù)極差檢驗(yàn)法(Duncan's multiple range test,DMRT)。數(shù)據(jù)分析采用SAS system 9.0版軟件進(jìn)行處理。
2.1.1 對(duì)淀粉酶活性的影響
以含有0.04%和0.08%濃度的苦瓜葉乙酸乙酯萃取物的人工飼料喂養(yǎng)斜紋夜蛾4齡幼蟲(chóng),24 h、48 h和72 h后對(duì)中腸淀粉酶活力的影響見(jiàn)圖1。由圖1可以看出,0.04%和0.08%2個(gè)濃度處理后24 h和48 h,斜紋夜蛾幼蟲(chóng)中腸的淀粉酶活性與對(duì)照相比差異不顯著,但72 h后,各處理的淀粉酶活性明顯降低,0.04%和0.08%濃度的苦瓜葉乙酸乙酯萃取物處理對(duì)斜紋夜蛾幼蟲(chóng)中腸淀粉酶活性的抑制率分別為22.7%和23.0%。

2.1.2 苦瓜葉乙酸乙酯萃取物對(duì)斜紋夜蛾幼蟲(chóng)蛋白酶活性的影響
以含有0.04%和0.08%濃度的苦瓜葉乙酸乙酯萃取物的人工飼料喂養(yǎng)斜紋夜蛾4齡幼蟲(chóng),24 h、48 h和72 h后對(duì)中腸蛋白酶酶活力的影響見(jiàn)圖2。由圖2可以看出,0.08%濃度處理24 h后,斜紋夜蛾幼蟲(chóng)中腸的蛋白酶活性就明顯低于對(duì)照組,抑制率達(dá)50.9%;48 h和72 h后,抑制率分別為38.4%和40.6%。0.04%濃度處理在24 h后對(duì)蛋白酶活性的影響與對(duì)照相比差異不顯著,但48 h和72 h后,對(duì)蛋白酶活性的抑制作用明顯增強(qiáng),抑制率達(dá)38.9%和39.3%。說(shuō)明苦瓜葉乙酸乙酯提取物對(duì)斜紋夜蛾幼蟲(chóng)中腸的蛋白酶活性有明顯的影響。從上述結(jié)果可以看出,苦瓜葉乙酸乙酯提取物對(duì)斜紋夜蛾幼蟲(chóng)中腸蛋白酶活性的抑制作用大于對(duì)淀粉酶活性的抑制作用。

2.2.1 Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)中腸淀粉酶活性的影響
以含有 0.05%、0.1%、0.2%、0.4%和0.6%濃度的Momordicin II的人工飼料喂養(yǎng)斜紋夜蛾4齡幼蟲(chóng),24 h、48 h和72 h后對(duì)中腸淀粉酶活力的影響見(jiàn)圖3。由圖3可以看出,各濃度Momordicin II處理24 h、48 h和72 h后,斜紋夜蛾幼蟲(chóng)中腸淀粉酶的活性與對(duì)照相比雖然有所降低但差異不顯著,表明Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)中腸的淀粉酶活性無(wú)明顯的抑制作用。
2.2.2 Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)中腸蛋白酶活性的影響
以含有 0.05%、0.1%、0.2%、0.4%和0.6%濃度的Momordicin II的人工飼料喂養(yǎng)斜紋夜蛾4齡幼蟲(chóng),24 h、48 h和72 h后對(duì)中腸蛋白酶活力的影響見(jiàn)圖4。由圖4可以看出,MomordicinⅡ?qū)π奔y夜幼蟲(chóng)中腸的蛋白酶活力有明顯的影響,且隨著使用濃度的增加抑制作用也隨著增強(qiáng)。MomordicinⅡ處理24 h和48 h后,0.05%和0.10%濃度處理的蛋白酶活性與對(duì)照差異不顯著,而濃度為0.2%、0.4%和0.6%的Momordicin II處理24 h和48 h后,其蛋白酶活性均顯著低于對(duì)照,24h的抑制率分別為44.9%、53.1%和63.5%;48 h的抑制率分別為27.0%、31.9%和42.9%。處理72 h后,5個(gè)濃度處理蛋白酶活性均顯著低于對(duì)照,對(duì)蛋白酶活性抑制作用最強(qiáng),各濃度處理的抑制率分別為 43.1%、40.2%、67.4%、71.8%和75.6%。說(shuō)明Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)中腸的蛋白酶活性有顯著的抑制作用。

本試驗(yàn)研究結(jié)果表明苦瓜葉乙酸乙酯萃取物對(duì)斜紋夜蛾幼蟲(chóng)中腸淀粉酶活性和蛋白酶活性均有一定的抑制作用,且對(duì)蛋白酶活性的抑制作用更明顯。斜紋夜蛾幼蟲(chóng)取食了含有Momordicin II的人工飼料后,對(duì)蛋白酶活性的影響與苦瓜葉乙酸乙酯萃取物大體相同,但Momordicin II對(duì)淀粉酶活性的影響不明顯,說(shuō)明苦瓜葉乙酸乙酯萃取物中還存在抑制斜紋夜蛾幼蟲(chóng)中腸淀粉酶活性的其他活性物質(zhì)。
近年來(lái),大量的研究結(jié)果表明,苦瓜葉片萃取物及其次生化合物對(duì)多種害蟲(chóng)有明顯的拒食、忌避產(chǎn)卵和抑制生長(zhǎng)發(fā)育的作用 (Chandravadana et al.,1983,1987;陳宏等,1996;Yasui,1998;Abe etal.,2000;李 紹勤 等,2001;Boeke,2004;Mekuria et al.,2005,2006;Kashiwagi et al.,2007;任立云等,2008;凌冰等,2008,2009;向亞林等,2009)。目前已從苦瓜葉片提取物中分離鑒定出多種對(duì)昆蟲(chóng)有生物活性的苦瓜素類(lèi)化合物,它們是苦瓜抗蟲(chóng)的重要活性物質(zhì),但有關(guān)這些化合物對(duì)昆蟲(chóng)的作用機(jī)理研究尚未見(jiàn)報(bào)道。
MomordicinⅡ是苦瓜葉提取物中的最主要苦瓜素類(lèi)化合物,生物活性測(cè)定結(jié)果表明MomordicinⅡ具有拒食作用,拒產(chǎn)卵作用和抑制生長(zhǎng)發(fā)育的作用 (Yasui,1998;Mekuria et al.,2005;凌冰等,2008)。與楝科植物中的印楝素相同,均為四環(huán)三萜類(lèi)化合物。印楝素對(duì)多種害蟲(chóng)有拒食、觸殺、胃毒、抑制生長(zhǎng)發(fā)育和影響卵巢發(fā)育的作用(Schmutterer,1990),通過(guò)影響口器上厭食及其他化學(xué)感器產(chǎn)生初生拒食反應(yīng);通過(guò)抑制昆蟲(chóng)中腸酯酶和脂肪體中蛋白酶、淀粉酶、脂肪酶、磷酸酶和葡萄糖酶的活性,降低昆蟲(chóng)的取食率和對(duì)食物的轉(zhuǎn)化利用率形成次生拒食反應(yīng) (程?hào)|美等2007)。消化酶是由昆蟲(chóng)中腸上皮細(xì)胞形成和分泌的,其活性直接影響昆蟲(chóng)對(duì)食物中營(yíng)養(yǎng)物質(zhì)的消化、吸收。其中,淀粉酶和蛋白酶是昆蟲(chóng)最主要的消化酶??喙先~乙酸乙酯萃取物和Momordicin II對(duì)斜紋夜蛾幼蟲(chóng)生長(zhǎng)發(fā)育及存活的影響可能與其抑制了幼蟲(chóng)中腸內(nèi)的消化酶有關(guān),淀粉酶和蛋白酶活性的顯著降低直接影響了斜紋夜蛾幼蟲(chóng)對(duì)食物中淀粉、纖維素及蛋白質(zhì)等的消化和吸收,進(jìn)一步抑制了斜紋夜蛾幼蟲(chóng)對(duì)食物的攝取及維持其生理功能和生長(zhǎng)發(fā)育需要的營(yíng)養(yǎng)物質(zhì)的吸收,因而影響了斜紋夜蛾的正常生長(zhǎng)發(fā)育。
